Anmerkung: With contributions from experts in the field, this book provides a comprehensive overview of the oxidative folding of cysteine-rich peptides, The formation of disulfide bonds is probably the most influential modification of peptides and proteins. An elaborate set of cellular machinery exists to catalyze and guide this process. In recent years, significant developments have been made in both our understanding of the in vivo situation and the in vitro manipulation of disulfide bonds. This is the first monograph to provide a comprehensive overview of this exciting and rapidly developing area. It offers in-depth insights into the mechanisms of in vivo and in vitro oxidative folding of proteins as well as mono- and multiple-stranded peptides. Procedures applied for laboratory and industrial purposes are also discussed by top experts in the field. The book describes the enzymes involved in the correct oxidative folding of cysteine-containing proteins in prokaryotes and eukaryotes. It then goes on to discuss the mimicking of these enzymes for successful in vitro folding of proteins (including synthetic replicates) and to deal with important issues concerning cysteine-rich peptides. The ability of natural bioactive peptides to fold correctly, and in high yields, to form defined structural motifs using cysteine sequence patterns is still puzzling. With this in mind, synthetic procedures for establishing native cysteine frameworks are discussed using selected examples, such as the potential of selenocysteines. The biotechnological and pharmaceutical relevance of proteins, peptides, their variants and synthetic replicates is continuously increasing. Consequently, this book is invaluable for peptide and protein chemists involved in related research and production

Inhalt: There is growing evidence that adult stem cells age. This process can result in alterations in the number and function of stem cells, leading to distinct phenotypic outcomes in different organ systems. This publication provides an outstanding overview of this emerging field. The molecular causes of stem cell aging remain to be defined. Stem cell aging can involve cell-intrinsic as well as cell-extrinsic alterations affecting the stem cell niche or the macroenvironment. Stem cells have a longer life span than other cell populations and retain a capacity to proliferate and differentiate in adult organs. The aging of adult stem cells plays a key role in the decline of organ maintenance and regenerative potential during aging and during the end stage of chronic diseases. In addition, it can contribute to stem cell transformation and carcinogenesis.Molecular Mechanisms of Adult Stem Cell Aging will appeal to scientists working in the fields such as stem cells, aging, regeneration and cancer. This subject matter should be of interest to physicians and scientists specializing in geriatric medicine, internal medicine, and surgery. It is also likely to be an invaluable resource for medical students and biologists who wish to enhance their understanding of molecular and stem cell biology

Serie: Frontiers of hormone research Vol. 25
Inhalt: Not much research has been done on the regulation of post-transcriptional events, much more is known about transcription. This book illustrates how hormones may influence post-transcriptional nuclear events and that some post-transcriptional modification is a requirement for the expression and function of most higher eukaryotic genes. Topics such as the regulation of pre-mRNA splicing by hormonal signals and tissue-specific mechanisms, pre-mRNA processing during spermatogenesis, to the link between the cell cycle and splicing, the role of phosphorylation and dephosphorylation in the function of the basal splicing machinery, RNA editing, the neuroendocrine regulation of polyadenylation all contribute to make up this volume. If you are an endocrinologist searching for interesting and important answers unearthed by RNA experiments, this book provides them

    ISBN: 978-3-318-00266-9
Serie: Frontiers of oral biology Vol. 10
Inhalt: Saliva is essential for oral health and influences all events in the mouth. A deficiency of saliva can have devastating consequences. Therefore, it is important to have a book about the basic tissue mechanisms involved in the secretion of saliva, based on an holistic approach. With such an aim in mind, this book contains chapters covering the histological basis for secretion, electrophysiological events, electrolyte and water secretion, protein synthesis and secretion, bloodflow, capillary dynamics, myoepithelial activity, glandular permeability, hormonal influences, including the effects of diabetes, and the synthesis and secretion of IgA in man. The chapters have been written by international experts in the field, who present balanced accounts of their subjects. The secretory mechanisms described in this book have a far-reaching impact beyond the salivary field and have applicability to all forms of exocrine secretion. Salivary glands offer many advantages for experimental study of these phenomena since they can be tested more precisely than most other glands, having ducts that can readily be cannulated for assessing the secretion. As a single-source of up-to-date data, this book will benefit doctors and dentists, and anyone interested in secretory phenomena, including physiologists, biochemists and cell biologists. It not only brings together all available data on this subject, as it stands at the turn of the century, but also lays down a sound platform of knowledge on which further investigations can be based

Serie: Frontiers of oral biology Vol. 11
Inhalt: Saliva is essential for oral health and influences all events in the mouth. In 1850 Ludwig discovered that autonomic nerve impulses evoke salivary secretion and all work since has shown that the nerves normally control the flow and protein output of saliva. This publication, written by international experts, is the first one devoted to the neuroglandular mechanisms of this control. The chapters contained deal with: the intimate details of the nerves and their different transmitters in the glands; central connections of the glandular nerves; receptors in the glands; nerve-induced glandular secretion of proteins; autonomic transmitters on salivary cells; the roles of non-conventional transmitters in the glands; the effects of denervations on the glands, on their capacity to synthesize and secrete proteins, and the development of supersensitivities to transmitter substances. Further chapters prescribe the inter-relationship between taste and saliva formation, and the reflexes involved in salivary secretion. The book lays a sound platform of knowledge to all with an interest in and around the mouth including dentists, pharmacologists, biochemists and cell biologists as well as neurobiologists working on autonomic nerve activities
