In dieser Arbeit wird die prognostische und /oder prädiktive Relevanz prätherapeutischer Werte (PTW) für das progressionsfreie Überleben (PFS) und das Gesamtüberleben (ÜL) unter Therapie mit Bevazizumab (BEV) und/oder Cetuximab (CET) bei metastasiertem kolorektalen Karzinom (mCRC) untersucht. Betrachtet wurden die PTW von Alkalischer Phosphatase (AP), Carbohydrat Antigen 19 9 (CA 19 9), Carcinoembryonales Antigen und Laktatdehydrogenase, sowie die Wahl des monoklonalen Antikörpers (mAb) insgesamt oder als Starttherapie. Keine der untersuchten Variablen war prädiktiv. Multivariat ließen sich AP und CA 19-9 als Prognosefaktoren für das ÜL bestätigen wobei ein niedriger PTW für eine gute Prognose spricht. Für das PFS zeigten sich CA 19 9 und die Wahl der mAb-Starttherapie mit BEV oder CET als prognostisch. Auch hier war ein niedrigerer PTW prognostisch von Vorteil, bezüglich der Starttherapie zeigte sich für die Wahl von BEV ein progressionsfreier Überlebensvorteil.

In this dissertation the prognostic and/or predictive valuesofpretherapeutic levels (PTL)for the overall survival (OS) and progressive free survival (PFS)in patients treated for metastatic colorectal cancer with bevacizumab (BEV) and/ or cetuximab were investigated. PTLof alkaline phophatase (AP), carbohydrate antigen 19 9 (CA 19 9), carcinoembryonic antigen and lactatedehydrogenase, as well as the administrationof certain monoclonal antibodies (mAb) altogether or as start therapy were examined. In a multivariate analysis PTL of AP and CA 19-9 were found to be prognostic for OS, whereupon a low PTL militates for a better OS. Forthe PFS, CA19-9 and the first givenmAbproved to be prognostic. A low PTL leads to a better PFS as does the administration of BEV as first mAb. None of the investigated variables were predictive.
