    Abstract: Creep-induced cavitation is an important failure mechanism in steel components operating at high temperature. Robust techniques are required to observe and quantify creep cavitation. In this paper, the use of two complementary analysis techniques: small-angle neutron scattering (SANS), and quantitative metallography, using scanning electron microscopy (SEM), is reported. The development of creep cavities that is accumulated under uniaxial load has been studied as a function of creep strain and life fraction, by carrying out interrupted tests on two sets of creep test specimens that are prepared from a Type-316H austenitic stainless steel reactor component. In order to examine the effects of pre-strain on creep damage formation, one set of specimens was subjected to a plastic pre-strain of 8%, and the other set had no pre-strain. Each set of specimens was subjected to different loading and temperature conditions, representative of those of current and future power plant operation. Cavities of up to 300 nm in size are quantified by using SANS, and their size distribution, as a function of determined creep strain. Cavitation increases significantly as creep strain increases throughout creep life. These results are confirmed by quantitative metallography analysis.

    Abstract: A partially (50%) perforated flooring system showed positive effects on health- and behavior-based welfare indicators without affecting production performance. Ammonia (NH3) is the most common air pollutant in poultry production, with effects on animal welfare and the environment. The objectives of animal welfare and environmental protection are often incompatible. Therefore, this study addresses the question of how a partially perforated flooring system affects NH3 emissions. According to German regulations, three fattening periods were carried out with 500 Ross 308 broilers per barn (final stocking density: 39 kg m−2). The experimental barn was equipped with an elevated perforated area in the supply section, accessible by perforated ramps. The remaining area in the experimental barn and the control barn were equipped with wood shavings (600 g m−2). Besides the different floor types, management was identical. Air temperature (Temp), relative air humidity (RH), NH3 concentration, and ventilation rate (VR) were measured continuously. Furthermore, dry matter (DM) content, pH, and litter quality were assessed. Towards the end of the fattening periods, the NH3 emission rate (ER) of the partially perforated flooring system was higher compared with that of the littered control barn (all p 〈 0.001). This effect is mainly caused by the higher NH3 concentrations, which are promoted by the lack of compaction underneath the elevated perforated area and the increase in pH value under aerobic conditions. Nevertheless, the partially perforated flooring system offers different approaches for NH3 reduction that were previously not feasible, potentially contributing equally to animal welfare and environmental protection.

    Abstract: Guanine-rich DNA sequences self-assemble into highly stable fourfold structures known as DNA-quadruplexes (or G-quadruplexes). G-quadruplexes have furthermore the tendency to associate into one-dimensional supramolecular aggregates termed G-wires. We studied the formation of G-wires in solutions of the sequences d(G4C2)n with n = 1, 2, and 4. The d(G4C2)n repeats, which are associated with some fatal neurological disorders, especially amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD), represent a challenging research topic due to their extensive structural polymorphism. We used dynamic light scattering (DLS) to measure translational diffusion coefficients and consequently resolve the length of the larger aggregates formed in solution. We found that all three sequences assemble into longer structures than previously reported. The d(G4C2) formed extremely long G-wires with lengths beyond 80 nm. The d(G4C2)2 formed a relatively short stacked dimeric quadruplex, while d(G4C2)4 formed multimers corresponding to seven stacked intramolecular quadruplexes. Profound differences between the multimerization properties of the investigated sequences were also confirmed by the AFM imaging of surface films. We propose that π-π stacking of the basic G-quadruplex units plays a vital role in the multimerization mechanism, which might be relevant for transformation from the regular medium-length to disease-related long d(G4C2)n repeats.

    Abstract: The equipment used in broiler breeder houses is an important factor in allowing the expression of the various behaviours of the animals, and thus realizing high welfare standards. Presently, detailed requirements for the equipment in broiler breeder houses are not specified in Germany, especially feed space and perch design allowance. One reason is that basic biometric data on broiler breeders are lacking. To close this gap, a pilot study was conducted, and birds’ width, weight, and feet were measured. Broiler breeders at 22 weeks of age (50 female and 17 male) were weighed and photographed digitally, and their body widths were calculated from the photographs. Female broiler breeders weighed 2791.80 ± 334.99 g on average and showed a body width of 20.63 ± 1.88 cm. For males, a mean of 3615.88 ± 432.46 g was measured with a body width of 21.94 ± 2.32 cm. Our examinations revealed that a trough side length of 21.00 cm per hen and 22.00 cm per cock must be provided to ensure that all broiler breeders have equal access to feed. The same dimensions should be planned as the perch length for each animal. Measurements of broiler breeders’ feet (506 female and 150 male Cobb 500) were performed at 60 weeks of age. The length of each toe and the width and length of the foot pad of both sexes were photographed and measured digitally. Female broiler breeders’ feet showed an average length of 10.14 cm, and male birds showed a length of 12.05 cm. Based on recommendations for the perch design for laying hens, round and oval perches for broiler breeders should have a circumference of at least 11.30 cm (female) or 13.40 cm (male). For angular perches, the upper contact area should have a width of 2.80 cm (female) or 3.40 cm (male). The obtained biometric data could be a useful basis for the development of legal requirements for broiler breeders.

    Abstract: Conventionally, hyperimmune globulin drugs manufactured from pooled immunoglobulins from vaccinated or convalescent donors have been used in treating infections where no treatment is available. This is especially important where multi-epitope neutralization is required to prevent the development of immune-evading viral mutants that can emerge upon treatment with monoclonal antibodies. Using microfluidics, flow sorting, and a targeted integration cell line, a first-in-class recombinant hyperimmune globulin therapeutic against SARS-CoV-2 (GIGA-2050) was generated. Using processes similar to conventional monoclonal antibody manufacturing, GIGA-2050, comprising 12,500 antibodies, was scaled-up for clinical manufacturing and multiple development/tox lots were assessed for consistency. Antibody sequence diversity, cell growth, productivity, and product quality were assessed across different manufacturing sites and production scales. GIGA-2050 was purified and tested for good laboratory procedures (GLP) toxicology, pharmacokinetics, and in vivo efficacy against natural SARS-CoV-2 infection in mice. The GIGA-2050 master cell bank was highly stable, producing material at consistent yield and product quality up to 〉 70 generations. Good manufacturing practices (GMP) and development batches of GIGA-2050 showed consistent product quality, impurity clearance, potency, and protection in an in vivo efficacy model. Nonhuman primate toxicology and pharmacokinetics studies suggest that GIGA-2050 is safe and has a half-life similar to other recombinant human IgG1 antibodies. These results supported a successful investigational new drug application for GIGA-2050. This study demonstrates that a new class of drugs, recombinant hyperimmune globulins, can be manufactured consistently at the clinical scale and presents a new approach to treating infectious diseases that targets multiple epitopes of a virus.

    Abstract: The objective of this study was to analyze sexual dimorphism with regard to the bone quality of Ross 308 broilers. The relationship between carcass traits, performance and bone quality was analyzed. The effect of sex and age at slaughter was examined in four replicates, always on day 31 and day 38. The weight, length, minimum diameter and breaking strength of the tibiotarsi were measured to determine the bone quality. Female tibiotarsi were shorter, lighter and had a lower minimum diameter and breaking strength compared to male ones. The tibiotarsi of older broilers were found to show higher values in the three-point bending test. The effects on broiler performance were determined by measuring the live body weight, carcass and cut weights. The results demonstrate significant effects of the age at slaughter and the sex (p 〈 0.001) on all measured parameters, with males always reaching higher values compared to females. A strong correlation of performance and bone parameters could be observed between live weight and weight of the tibiotarsi on both days of data collection. Therefore, it could be beneficial to raise female and male broilers separately. However, in order to make specific recommendations for practice, further knowledge is necessary.

    Abstract: The available floor space is an important welfare factor for cull sows during transportation. Sows of modern genetics reach a size and weight far exceeding those of fattening pigs. In most countries, there are no binding, consistent regulations for the maximum loading densities, especially for sows during road transportation. As a first step towards such recommendations, the physical floor space requirement (static space) of 100 sows of a current breed, while standing and lying down, were determined using contrast-based planimetry. An average sow covered about 0.42–0.47 m2 (standing postures) up to 0.53–0.63 m2 (lying postures). The largest measured area was 0.72 m2 for a sow lying in the belly-chest position. We detected a significant dependency of the covered floor area and the live weight, which supports the common practice to derive space requirements and recommendations based on live weight. Also, our results suggest that especially heavy sows, under currently usual loading densities, are at risk of having insufficient floor space requirements during transport. The results cannot be used to define the space required by a sow to carry out movements or sustain the individual need for distance (social/dynamic space) but provide data on the static space covered by sows of current breeds.

    Abstract: Relatively common behavioral patterns in adolescence are problematic use of video games (PG), social media (PSMU) or alcohol (PAU). According to theoretical models, personality traits are relevant for Internet-related problematic behaviors. In the present study, associations of the Big Five personality domains and their 15 facets with PG, PSMU and PAU were compared for the first time. Therefore, 492 adolescents (mean age: 16.83 years) were examined with the established Big Five Inventory-2 as well as other standardized questionnaires on PG, PSMU and PAU. For statistical evaluation, correlation analyses were used as bivariate procedures and multiple regression analyses as multivariable procedures. At the personality domain level, consistently in bivariate and multivariate analyses, statistically significant associations between higher Negative Emotionality (Neuroticism) and PG, PSMU and PAU as well as between lower Open-Mindedness and PG and PAU were observed. At the level of facets, higher Anxiety (facet of Negative Emotionality) was related to PG and PSMU as well as lower Aesthetic Sensitivity (facet of Open-Mindedness) and lower Productiveness (facet of Conscientiousness) to PG. Considering the overlap of 95% confidence intervals, very comparable patterns of associations between PG, PSMU and PAU and the Big Five and their facets were observed in adolescence (indicating similarities in etiology).

    Abstract: Telerehabilitation (TR) has gained attention as a promising rehabilitation format. Our study examined how patients responded to TR and whether it provided adequate support for their lifestyle changes and self-care efforts when compared to conventional rehabilitation (CR). Cardiac patients (n = 136) were randomly assigned to a TR or CR group. The TR group was provided with relevant health care technology for a period of three months, and both groups filled in questionnaires on their motivation for lifestyle changes and self-care psychological distress, and quality of life at 0, 3, 6, and 12 months. Patients in both groups were found to be equally motivated for lifestyle changes and self-care (p 〈 0.05) and they experienced similar levels of psychological distress and quality of life. TR is comparable to conventional rehabilitation in motivating patients, preventing psychological distress and improving quality of life. Although we observed an initial increase in autonomous motivation in the telerehabilitation group, this positive difference in motivation does not last over time. As such, neither rehabilitation format seems able to ensure long-term motivation. Therefore, TR may serve as a viable replacement for conventional rehabilitation when considered relevant. Further research is needed to enhance long-term motivation, and maybe telerehabilitation can help to achieve this.
