    Abstract: Background. The effect of GuizhiFuling Wan (GFW) on adenomyosis (AM) is definite. This study aimed to explore the mechanism and key therapeutic targets of GFW in treating AM through network pharmacology combined with molecular docking and experimental verification. Materials and Methods. In network pharmacology, firstly, the active components of GFW, its drug, and disease targets were screened through several related public databases, and GFW-AM common targets were obtained after the intersection. Then, the biological function (Gene Ontology, GO) and pathway (Kyoto Encyclopedia of Genes and Genomes, KEGG) of GFW in treating AM were enriched and analyzed. Finally, the interaction and binding force between key components and key targets of GFW were verified by molecular docking. In the animal part, the effect of GFW on the expression of matrix metallopeptidase 2 (MMP-2), matrix metallopeptidase 9 (MMP-9), and vascular endothelial growth factor (VEGF) in mice with AM was observed by HE staining, ELISA, and immunohistochemistry. Results. In this study, 89 active components of GFW, 102 related targets, and 291 targets of AM were collected. After the intersection, 26 common targets were finally obtained. The key active compounds were baicalein, sitosterol, and β-sitosterol, and the key targets were MMP-2, MMP-9, and VEGF. GO and KEGG enrichment analyses showed that biological processes such as the positive regulation of vascular endothelial migration and signaling pathways such as TNF and HIF-1 were involved in regulating angiogenesis, invasion, and metastasis in AM. The molecular docking results showed that baicalein, β-sitosterol, and stigmasterol had better binding potential with MMP-2, MMP-9, and VEGF. The results of in vivo analysis showed that GFW could decrease the serum content and protein expression of MMP-2, MMP-9, and VEGF in mice with AM. Conclusions. GFW could reduce the expression of MMP-2, MMP-9, and VEGF, which might be an essential mechanism for GFW to inhibit the invasion and metastasis of ectopic tissues of AM.

    Abstract: With the rapid development of e-commerce, recommendation system has become one of the main tools that assists users in decision-making, enhances user’s experience, and creates economic value. Since it is difficult to explain the implicit features generated by matrix factorization, explainable recommendation system has attracted more and more attention recently. In this paper, we propose an explainable fast recommendation model by combining fine-grained sentiment in review data (FSER, (Fast) Fine-grained Sentiment for Explainable Recommendation). We innovatively construct user-rating matrix, user-aspect sentiment matrix, and item aspect-descriptive word frequency matrix from the review-based data. And the three matrices are reconstructed by matrix factorization method. The reconstructed results of user-aspect sentiment matrix and item aspect-descriptive word frequency matrix can provide explanation for the final recommendation results. Experiments in the Yelp and Public Comment datasets demonstrate that, compared with several classical models, the proposed FSER model is in the optimal recommendation accuracy range and has lower sparseness and higher training efficiency than tensor models or neural network models; furthermore, it can generate explanatory texts and diagrams that have high interpretation quality.

    Abstract: Colorectal cancer (CRC) is one of the most common malignant tumors worldwide, causing a large number of cancer-related deaths each year. Patients are usually diagnosed at advanced and incurable stages due to the lack of suitable screening methods for early detection. Noncoding RNAs (ncRNAs), including small and long noncoding RNAs (lncRNA), are known to have significant regulatory functions, and accumulating evidence suggests that circulating ncRNAs have potential applications as noninvasive biomarkers for diagnosing CRC, evaluating its prognosis, or predicting chemosensitivity in the general population. In this review, we summarize the origins of circulating ncRNAs and provide details of single and multiple circulating ncRNAs that might have roles as diagnostic and prognostic biomarkers in CRC. We end by discussing circulating ncRNAs that may distinguish patients with resistance to chemotherapy.

    Abstract: Objective. To evaluate whether traditional Chinese medicine compound preparations (TCMCPs) are associated with rheumatoid arthritis- (RA-) related complications (including readmission, Sjogren’s syndrome, surgical treatment, and all-cause death) in patients with RA. Methods. Clinical outcome data were retrospectively collected from patients with RA discharged from the Department of Rheumatology and Immunology of the First Affiliated Hospital of Anhui University of Chinese Medicine from January 2009 to June 2021. The propensity score matching method was used to match baseline data. Multivariate analysis was conducted to analyze sex, age, the incidence of hypertension, diabetes, and hyperlipidemia and identify the risk of readmission, Sjogren’s syndrome, surgical treatment, and all-cause death. Users of TCMCP and nonusers of TCMCP were defined as the TCMCP and non-TCMCP groups, respectively. Results. A total of 11,074 patients with RA were included in the study. The median follow-up time was 54.85 months. After propensity score matching, the baseline data of TCMCP users corresponded with those of non-TCMCP users, with 3517 cases in each group. Retrospective analysis revealed that TCMCP significantly reduced clinical, immune, and inflammatory indices in patients with RA, and these indices were highly correlated. Notably, the composite endpoint prognosis for treatment failure in TCMCP users was better than that in non-TCMCP users ( HR = 0.75 (0.71-0.80)). The risk of RA-related complications in TCMCP users with high-exposure intensity ( HR = 0.669 (0.650-0.751)) and medium-exposure intensity ( HR = 0.796 (0.691-0.918)) was significantly lower than those in non-TCMCP users. An increase in exposure intensity was associated with a concomitant decrease in the risk of RA-related complications. Conclusion. The use of TCMCPs, as well as long-term exposure to TCMCPs, may lower RA-related complications, including readmission, Sjogren’s syndrome, surgical treatment, and all-cause death, in patients with RA.

    Abstract: Residues from herbal medicine processing in pharmaceutical plants create a large amount of waste (herb residues), which consists mainly of environmental pollution and medicinal waste. In order to resolve this problem, probiotics of Bacillus (B.) subtilis , Aspergillus (A.) oryzae , and Lactobacillus (L.) plantarum M3 are selected to reuse herb residue of Jianweixiaoshi tablets (JT), and an antibiotic-associated diarrhea (AAD) mouse model was established to evaluate the therapeutic effects of the herb residue fermentation supernatant. Our results indicated that the fermentation supernatant had scavenged 77.8% of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 78% of O 2 •− , 36.7% of • OH, 39% of Fe 2+ chelation, and 716 mg/L reducing power. The inhibition zones for Salmonella (S.) typhimurium , S. enteritidis , Shigella (Sh.) flexneri , Escherichia (E.) coli , Listeria (L.) monocytogenes , Sh. dysenteriae 301, and Staphylococcus (S.) aureus were 17, 14, 19, 18, 20, 19, and 20 mm, respectively. The in vivo results indicated that the fermentation supernatant resulted in a high diarrhea inhibition rate (56%, p 〈 0.05 ), greatly enhanced the disruption of bacterial diversity caused by antibiotics, and restored the dominant position of L. johnsonii in the treatment and recovery stages. Therefore, the combination of the herb residue and probiotics suggests a potential to explore conversion of these materials for the possible development of therapies for AAD.

    Abstract: The purpose of this study was to prepare a novel paclitaxel (PTX) microemulsion containing a reduced amount of Cremophor EL (CrEL) which had similar pharmacokinetics and antitumor efficacy as the commercially available PTX injection, but a significantly reduced allergic effect due to the CrEL. The pharmacokinetics, biodistribution, in vivo antitumor activity and safety of PTX microemulsion was evaluated. The results of pharmacokinetic and distribution properties of PTX in the microemulsion were similar to those of the PTX injection. The antitumor efficacy of the PTX microemulsion in OVCRA-3 and A 549 tumor-bearing animals was similar to that of PTX injection. The PTX microemulsion did not cause haemolysis, erythrocyte agglutination or simulative reaction. The incidence and degree of allergic reactions exhibited by the PTX microemulsion group, with or without premedication, were significantly lower than those in the PTX injection group ( P 〈 . 01 ). In conclusion, the PTX microemulsion had similar pharmacokinetics and anti-tumor efficacy to the PTX injection, but a significantly reduced allergic effect due to CrEL, indicating that the PTX microemulsion overcomes the disadvantages of the conventional PTX injection and is one way of avoiding the limitations of current injection product while providing suitable therapeutic efficacy.

    Abstract: Objective. To explore the protective effect of special electromagnetic field treated water (SEW) and far infrared rays (IFR) on endotoxin (lipopolysaccharide, LPS) induced ARDS rats and the effect on inflammatory factors. Methods. 40 healthy male SD rats were randomly divided into 4 groups with 10 in each group. Preventive experiment: Adaptive feeding was carried out for 1 week according to animal feeding standards. Rats in SEW group drank SEW daily. Samely, rats in SEW and FIR group also drank SEW daily. Besides, rats in SEW and FIR group were also given far infrared rays for 20min/d. Rats in model group drank distilled water daily. After 7 days, rats in each group were injected with LPS (2 mg/kg) via the tail vein for making models. Rats in blank control group were given distilled water for 7 days, without modeling. All rats in the 4 groups were put to death under anesthesia 16 hours after modeling. Lung tissue and abdominal aortic blood were taken from these rats. Results. Pathological observation of lung and lung tissue indicated that rats in model group showed great pathological difference from rats in blank group. Rats in intervention group showed more symptomatic improvement in relation to alveolar and pulmonary interstitial congestion, edema, and inflammatory cell infiltration than rats in model group. The level of inflammatory factors like IL-1 β and IL-6 in serum of rats in model group increased compared to blank control group (p 〈 0.05). Comparing SEW group and SEW and FIR group with model group, levels of IL-1 β and IL-6 in serum of rats both decreased remarkably (IL-I β : P 〈 0.05; IL-6: P 〈 0.01) while there was no obvious difference between SEW group and SEW and FIR group (p 〉 0.05). The lung coefficient (LI) in SEW and FIR group was significantly lower than that in model group ( P 〈 0.01), which was higher than that of blank control group ( P 〈 0.05), while there was no obvious difference between model group and SEW group ( P 〉 0.05). Compared with blank control group, lung permeability index (LPI) in model group showed no obvious difference (P 〉 0.05). Conclusion. Special electromagnetic field treated water and far infrared rays can alleviate lung tissue damage of endotoxin-induced ARDS rats, relieving symptoms of alveolar and pulmonary interstitial congestion, edema, and inflammatory cell infiltration. The protective effect of special electromagnetic field treated water and far infrared rays on endotoxin-induced acute respiratory distress syndrome may result from their role in reducing the levels of IL-1 β and IL-6 in serum and the expression level of p65 protein in lung tissue, in addition to reliving inflammatory response, lung coefficient, and lungs edema.

    Abstract: Little work is done to develop Aloe vera (AV) using probiotics. To explore the potential benefits, the antioxidant effects and the antibacterial effects on foodborne pathogens of Aloe fermentation supernatant were evaluated in vitro. Our results indicated that the Aloe fermentation supernatant fermented by Lactobacillus plantarum HM218749.1 had very strong scavenging capacities of the DPPH (86%), O 2 • - (85%), O H • (76%), and F e 2 + chelation (82%) and reducing powers (242.5 mg/L), and the inhibition zones for Salmonella typhimurium , Salmonella enteritidis , Shigella flexneri , Escherichia coli , Listeria monocytogenes , S. dysenteriae 301, Staphylococcus aureus Cowan1, and Propionibacterium acnes were 16, 15, 19, 20, 21, 20, and 27 mm. Moreover, the low concentration of Aloe fermentation supernatant had significantly reduced the production of IL-1 β , TNF- α , and IL-6 in both mRNA and protein levels ( P 〈 0.01 ). Therefore, the Aloe fermentation supernatant can be used as functional beverage or cosmetic ingredients to guard human intestinal health, delaying senescence, and prevent chronic diseases.

    Abstract: Type 2 diabetes constitutes a serious threat to the health of patients, but there is currently no ideal treatment in the clinic. Glucagon-like peptide-1 and human umbilical cord mesenchymal stem cells have been confirmed to have antidiabetic effects, but both of them have certain defects in the process of antidiabetes, which cannot meet the need of clinical treatment. We hypothesized that human umbilical cord mesenchymal stem cells can be used as a vector to construct a novel cell line that expresses GLP-1 in vivo for a long time. And this cell strain results in lowering blood glucose in type 2 diabetic mice. The results showed that after 3 weeks of intramuscular injection of the new cell line, the fasting blood glucose of type 2 diabetic mice returned to the normal range, and the hypoglycemic effect was maintained within 3 weeks after putting an end to the drug. At the same time, during the administration, the mice lost weight, the food intake decreased, the half-life of GLP-1 in the body prolonged, the IR reduced, and the pancreatic function recovered. The results of this study indicate that the novel cell line can prolong the half-life of GLP-1 in vivo and effectively lower blood sugar, which is a feasible method to improve type 2 diabetes.

    Abstract: Background . The gut microbiota is involved in the occurrence and development of chronic liver diseases. Zonulin is considered a marker of intestinal permeability. The purpose of this study was to assess zonulin levels in patients with chronic hepatitis B (CHB), HBV-associated liver cirrhosis (LC), and HBV-associated hepatocellular carcinoma (HCC). Materials and Methods . The study population consisted of 90 HBV-associated HCC patients, 90 HBV-associated LC patients, 90 CHB patients, and 90 healthy subjects. Serum levels of zonulin and AFP were determined. The diagnostic accuracy of each marker was evaluated using receiver operating characteristic (ROC) curve analysis (AUC). Results . Serum zonulin levels were significantly higher in patients with HCC than in patients with LC or CHB or healthy subjects ( p 〈 0.001 ). Moreover, the zonulin levels were increased in the advanced stage of LC and HCC. ROC curve analysis revealed that serum zonulin could be used to differentiate CHB from cirrhosis. In addition, the combination of zonulin and AFP exhibited a significantly larger AUC compared with zonulin or AFP alone. Conclusions . Serum zonulin levels were significantly increased both in LC and in HCC and correlated with the advanced stage of LC and HCC. Moreover, the combination of zonulin and AFP confers significant benefit to diagnostic accuracy in differentiating LC from HCC.
