    Kurzfassung: Young Zucker lean (Fa/-) and obese (fa/fa) female rats were fed the fatty acid synthesis inhibitor (-)-hydroxy-citrate as a dietary admixture for 39 days. In the lean rats, (-)-hydroxycitrate treatment decreased body weight, food intake, percent of body fat, and fat cell size. In the obese rat, food intake and body weight were reduced but the percent of body fat remained unchanged. Throughout the treatment period, obese rats maintained a fat cell size equivalent to their obese controls. Although a reduction in fat cell number in the obese rats occurred during the treatment period, marked hyperplasia was observed during the posttreatment period. The results of this study indicate that the obese rat, despite a substantial reduction in body weight produced by (-)-hydroxycitrate, still defends its obese body composition.

    Kurzfassung: Fat cell hypertrophy occurs in most forms of obesity. In the obese Zucker rat, fat cell hypertrophy and increased lipoprotein lipase activity (LPL) are the earliest known signs of obesity. We studied the regulation of fat cell size in the obese Zucker rat by measuring changes in fat cell LPL activity and lipolysis in response to an overnight fast in 6- and 14-wk-old lean and obese rats. At both ages, fed obese rats had significantly increased fat cell size, LPL activity, and basal glycerol release in three adipose tissue depots compared to fed lean rats. Obese rats decreased LPL activity in response to fasting, but levels always remained equal to or greater than those in fed lean rats. Obese rats also showed a reduced lipolytic response to fasting. Thus, the obese rat after an overnight fast could not produce a coordinated response to fasting similar to the lean rat, and its homeostatic adjustments to this mild stimulus favored preservation of its enlarged fat cell size.

    Kurzfassung: The properties of neuronal spike potentials ranging from 20 to 60 mV in size were studied in stable recordings made from the motor cortex of unanesthetized, unparalyzed cats. Histologic evidence that injection of horseradish peroxidase (HRP) was confined to single neurons and appropriately high levels of potassium measured with K+ ion-sensitive electrodes indicated that these recordings were obtained from single, intracellularly penetrated neurons. Pressure injection of small volumes of 4% HRP in 1 M KC1, intracellularly, was characteristically associated with transient increases in spike size and decreases in input resistance. Recoveries of HRP-filled dendritic processes without filling of somata were obtained from penetrations giving spikes smaller than the recorded resting potential (spike-undershoot recordings). Recordings with dendritic recoveries had higher input resistances and showed greater increases in spike size during pressure injection than did recordings with both somata and dendrites recovered. The activity in response to a weak click elicited in cells with spike potentials between 20 and 40 mV was as great or greater than that elicited in cells with larger spike potentials or in extracellularly recorded units. We conclude that many stable spike-undershoot recordings of neurons of the motor cortex of awake cats reflect penetrations of dendritic processes as opposed to injury because they show a) a normal functional response to weak auditory stimuli delivered over periods of many minutes, b) a reversible increase in spike size during pressure injection of small volumes of HRP in KC1 consistent with penetration of a cable remote from the site of spike generation, and c) the frequent recovery of HRP-marked dendrites without somata.
