image The transcription factor SOX 9 is a key regulator of stem cell programs with potential oncogenic activities. Its degradation via the tumor suppressor FBW 7 shows that its regulation directly impinges on medulloblastoma malignancy. FBW7 is commonly mutated or transcriptionally downregulated in human medulloblastoma. FBW7 recognizes a conserved SOX9 degron phosphorylated by GSK3 and targets SOX9 for ubiquitylation and proteasomal degradation. Inactivation of FBW7 leads to elevated SOX9 levels, promoting cell migration, metastasis, and cisplatin treatment resistance. PI3K/AKT/mTOR pathway inhibition sensitizes medulloblastoma cells to cisplatin treatment through GSK3/FBW7‐mediated SOX9 degradation.
