    Abstract: Psoriasis is a chronic skin disease affecting 2–3% of the global population. The proinflammatory IL-17A is a key cytokine in psoriasis. Accumulating evidence has revealed that IL-36γ plays also a pathogenic role. To understand more precisely the role of the IL-17A–IL-36γ cytokine network in skin pathology, we used an ear injection model. We injected IL-17A or IL-36γ alone and in combination into the ear pinnae of mice. This resulted in a significant increase in ear thickness measured over time. Histological evaluation of IL-17A + IL-36γ-treated skin showed a strong acanthosis, hyperparakeratosis and infiltration of neutrophils. The same histological features were found in mice after injection of IL-36γ alone, but to a lesser extent. IL-17A alone was not able to induce psoriasis-like changes. Genes encoding proteins of the S100 family, antimicrobial peptides and chemo-attractants for neutrophils were upregulated in the IL-17A + IL-36γ group. A much weaker expression was seen after the injection of each cytokine alone. These results strengthen the hypothesis that IL-17A and IL-36γ drive psoriatic inflammation via a synergistic interaction. Our established intradermal ear injection model can be utilized in the future to monitor effects of various inhibitors of this cytokine network.

    Abstract: Oxidative stress (OS) represents a state of an imbalanced amount of reactive oxygen species (ROS) and/or a hampered efficacy of the antioxidative defense system. Cells of the central nervous system are particularly sensitive to OS, as they have a massive need of oxygen to maintain proper function. Consequently, OS represents a common pathophysiological hallmark of neurodegenerative diseases and is discussed to contribute to the neurodegeneration observed amongst others in Alzheimer’s disease and Parkinson’s disease. In this context, accumulating evidence suggests that OS is involved in the pathophysiology of Niemann-Pick type C1 disease (NPC1). NPC1, a rare hereditary neurodegenerative disease, belongs to the family of lysosomal storage disorders. A major hallmark of the disease is the accumulation of cholesterol and other glycosphingolipids in lysosomes. Several studies describe OS both in murine in vivo and in vitro NPC1 models. However, studies based on human cells are limited to NPC1 patient-derived fibroblasts. Thus, we analyzed OS in a human neuronal model based on NPC1 patient-specific induced pluripotent stem cells (iPSCs). Higher ROS levels, as determined by DCF (dichlorodihydrofluorescein) fluorescence, indicated oxidative stress in all NPC1-deficient cell lines. This finding was further supported by reduced superoxide dismutase (SOD) activity. The analysis of mRNA and protein levels of SOD1 and SOD2 did not reveal any difference between control cells and NPC1-deficient cells. Interestingly, we observed a striking decrease in catalase mRNA and protein levels in all NPC1-deficient cell lines. As catalase is a key enzyme of the cellular antioxidative defense system, we concluded that the lack of catalase contributes to the elevated ROS levels observed in NPC1-deficient cells. Thus, a restitution of a physiological catalase level may pose an intervention strategy to rescue NPC1-deficient cells from the repercussions of oxidative stress contributing to the neurodegeneration observed in NPC1.

    Abstract: The reaction of amiloride hydrochloride (1.HCI) with bromine in alkaline solution generated a yellow-brown dehydrogenation product, which tumed out as 3-(3-amino-I,2,4-oxadiazol-5-yl)-5-chloro-2,6-pyrazinediamine (2). The structure was deduced from the MS and the NMR spectra of 2 with the help of comparisons with corresponding spectra of amiloride (1) and reference substances 3 - 5. The agreement of all relevant data of the product and of authentical oxadiazolylpyrazine 2 as weil as the accomplished X-ray analysis confirmed the postulated structure. The mechanism of the formation of 2 is also discussed.

    Abstract: Globally, the number of dams increased dramatically during the 20th century. As a result, monitoring water levels and storage volume of dam-reservoirs has become essential in order to understand water resource availability amid changing climate and drought patterns. Recent advancements in remote sensing data show great potential for studies pertaining to long-term monitoring of reservoir water volume variations. In this study, we used freely available remote sensing products to assess volume variations for Lake Mead, Lake Powell and reservoirs in California between 1984 and 2015. Additionally, we provided insights on reservoir water volume fluctuations and hydrological drought patterns in the region. We based our volumetric estimations on the area–elevation hypsometry relationship, by combining water areas from the Global Surface Water (GSW) monthly water history (MWH) product with corresponding water surface median elevation values from three different digital elevation models (DEM) into a regression analysis. Using Lake Mead and Lake Powell as our validation reservoirs, we calculated a volumetric time series for the GSWMWH–DEMmedian elevation combinations that showed a strong linear ‘area (WA) – elevation (WH)’ (R2 〉 0.75) hypsometry. Based on ‘WA-WH’ linearity and correlation analysis between the estimated and in situ volumetric time series, the methodology was expanded to reservoirs in California. Our volumetric results detected four distinct periods of water volume declines: 1987–1992, 2000–2004, 2007–2009 and 2012–2015 for Lake Mead, Lake Powell and in 40 reservoirs in California. We also used multiscalar Standardized Precipitation Evapotranspiration Index (SPEI) for San Joaquin drainage in California to assess regional links between the drought indicators and reservoir volume fluctuations. We found highest correlations between reservoir volume variations and the SPEI at medium time scales (12–18–24–36 months). Our work demonstrates the potential of processed, open source remote sensing products for reservoir water volume variations and provides insights on usability of these variations in hydrological drought monitoring. Furthermore, the spatial coverage and long-term temporal availability of our data presents an opportunity to transfer these methods for volumetric analyses on a global scale.

    Abstract: Forests in Germany cover around 11.4 million hectares and, thus, a share of 32% of Germany’s surface area. Therefore, forests shape the character of the country’s cultural landscape. Germany’s forests fulfil a variety of functions for nature and society, and also play an important role in the context of climate levelling. Climate change, manifested via rising temperatures and current weather extremes, has a negative impact on the health and development of forests. Within the last five years, severe storms, extreme drought, and heat waves, and the subsequent mass reproduction of bark beetles have all seriously affected Germany’s forests. Facing the current dramatic extent of forest damage and the emerging long-term consequences, the effort to preserve forests in Germany, along with their diversity and productivity, is an indispensable task for the government. Several German ministries have and plan to initiate measures supporting forest health. Quantitative data is one means for sound decision-making to ensure the monitoring of the forest and to improve the monitoring of forest damage. In addition to existing forest monitoring systems, such as the federal forest inventory, the national crown condition survey, and the national forest soil inventory, systematic surveys of forest condition and vulnerability at the national scale can be expanded with the help of a satellite-based earth observation. In this review, we analysed and categorized all research studies published in the last 20 years that focus on the remote sensing of forests in Germany. For this study, 166 citation indexed research publications have been thoroughly analysed with respect to publication frequency, location of studies undertaken, spatial and temporal scale, coverage of the studies, satellite sensors employed, thematic foci of the studies, and overall outcomes, allowing us to identify major research and geoinformation product gaps.

    Abstract: The multiple-use approach to forestry applied in Germany aims to combine timber production and habitat management by preserving specific stand structures. We selected four forest stand types comprising (i) pure oak, (ii) equal oak–pine mixtures, (iii) single tree admixtures of oak in pine forest and (iv) pure pine. We analysed the effects of stand composition parameters on species representative of the larger carabid beetles (Carabus arvensis, C. coriaceus, C. hortensis, C. violaceus, Calosoma inquisitor). The main statistical methods used were correlation analyses and generalised linear mixed models. Cal. inquisitor was observed in pure oak forests exclusively. C. coriaceus and C. hortensis were absent from pure pine stands. High activity densities of C. arvensis and C. violaceus were observed in all four forest types. When assessed at the smaller scales of species crown cover proportions and spatial tree species effect zones, C. hortensis was found to be positively related to oak trees with a regular spatial distribution, whereas C. coriaceus preferred lower and more aggregated oak tree proportions. C. violaceus showed strong sex-specific tree species affinities. Information about preferences of carabid beetles is necessary for management activities targeting the adaptation of forest structures to habitat requirements.

    Abstract: This study evaluated the effect of differences in residual feed intake (RFI) of Charolaise candidate young bulls on feeding behavior and self-performance test results. Bulls were classified into high and low RFI (H-RFI, L-RFI) groups. Bulls were fed in a HOKOFARM system to measure individual animal intake and behavior. L-RFI bulls had significantly lower feed intakes (p = 0.002) and higher gain to feed ratio (p = 0.001), lower intake per day/kg DM (dry matter) (p = 0.002) and lower intake g/body weight/day (p 〈 0.001). L-RFI animals had lower visits number per day (p = 0.02), but spent longer time per visit (p = 0.02), and tended to have higher intake g/visit (p = 0.06) on feeders. The correlation between RFI and DMI (dry matter intake)/bodyweight/day as well as intake per day/kg were large and positive. Back-loin length and rump length, and moreover muzzle width and frame, showed negative correlations with RFI value. However, bulls with better RFI values associated with lower legs score. Results reveal that RFI was shown beneficial correlations with economically relevant self-performance traits. Further investigations are needed to seek additional indicator traits that are predictive for RFI.

    Abstract: In the past decades, various Earth observation-based time series products have emerged, which have enabled studies and analysis of global change processes. Besides their contribution to understanding past processes, time series datasets hold enormous potential for predictive modeling and thereby meet the demands of decision makers on future scenarios. In order to further exploit these data, a novel pixel-based approach has been introduced, which is the spatio-temporal matrix (STM). The approach integrates the historical characteristics of a specific land cover at a high temporal frequency in order to interpret the spatial and temporal information for the neighborhood of a given target pixel. The provided information can be exploited with common predictive models and algorithms. In this study, this approach was utilized and evaluated for the prediction of future urban/built-settlement growth. Random forest and multi-layer perceptron were employed for the prediction. The tests have been carried out with training strategies based on a one-year and a ten-year time span for the urban agglomerations of Surat (India), Ho-Chi-Minh City (Vietnam), and Abidjan (Ivory Coast). The slope, land use, exclusion, urban, transportation, hillshade (SLEUTH) model was selected as a baseline indicator for the performance evaluation. The statistical results from the receiver operating characteristic curve (ROC) demonstrate a good ability of the STM to facilitate the prediction of future settlement growth and its transferability to different cities, with area under the curve (AUC) values greater than 0.85. Compared with SLEUTH, the STM-based model achieved higher AUC in all of the test cases, while being independent of the additional datasets for the restricted and the preferential development areas.

    Abstract: Poor agricultural productivity has led to food shortages for smallholder farmers in Ethiopia. Agroforestry may improve food security by increasing soil fertility, crop production, and livelihoods. Agroforestry simulation models can be useful for predicting the effects of tree management on crop growth when designing modifications to these systems. The Agricultural Production Systems sIMulator (APSIM) agroforestry tree-proxy model was used to simulate the response of maize yield to N fertilizer applications and tree pruning practices in the parkland agroforestry system in the Central Rift Valley, Ethiopia. The model was parameterized and tested using data collected from an experiment conducted under trees and in crop-only plots during the 2015 and 2016 growing seasons. The treatments contained three levels of tree pruning (100% pruned, 50% pruned, and unpruned) as the main plots, and N fertilizers were applied to maize at two rates (9 or 78 kg N ha−1) as sub-plots. Maize yield predictions across two years in response to tree pruning and N applications under tree canopies were satisfactorily simulated (NSE = 0.72, RSR = 0.51, R2 = 0.8). Virtual experiments for different rates of N, pruning levels, sowing dates, and cultivars suggest that maize yield could be improved by applying fertilizers (particularly on crop-only plots) and by at least 50% pruning of trees. Optimal maize yield can be obtained at a higher rate of fertilization under trees than away from them due to better water relations, and there is scope for improving the sowing date and cultivar. Across a 34-year range of recent climate, small increases in yields due to optimum N-fertilizing and pruning were probably limited by nutrient limitations other than N, but the highest yields were consistently in the 2–4 m zone under trees. These virtual experiments helped to form hypotheses regarding fertilizers, pruning, and the effects of trees on soil that warrant further field evaluation.
