Abstract:
From tumour material of bovine skin leukosis a permanent cell line designated SBL‐H12578 was established. The cells were shown to have T‐cell character and induced transplantable tumours in nude mice. They produced type C particles as demonstrated by electron microscopy, simultaneous detection of 70 S RNA and reverse transcriptase activity associated with particles from the density range 1.15 to 1.17 g/ml. The skin tumour virus isolate did not show genetic sequence homology or serological cross reactivity with bovine leukemia virus. However, a reactivity of SBL‐H12578 antigen with a number of sera from cases of calf and skin leukosis was demonstrated. Zusammenfassung Etablierung einer Zellinie aus einem Hautleukosetumor eines Rindes und Nachweis von Retrovirus‐Aktivität Aus Biopsiematerial von einem Hautleukosetumor vom Rind wurde eine permanente Zellinie mit T‐Zellcharakter mit der Bezeichnung SBL‐H12578 etabliert. Die Zellen erzeugten in Nacktmäusen transplantable Tumoren. Durch Elektronenmikroskopie, den Nachweis einer 70 S RNS im „Simultaneous Detection Test” und dem Nachweis von Reverser Transkriptase, die mit Partikeln mit einer Dichte von 1,15 bis 1,17 g/ml assoziiert war, konnte die Produktion von C‐Typ‐Viren durch die SBL‐H12578‐Zellen nachgewiesen werden. Das Virusisolat zeigte keine genetische oder serologische Verwandtschaft mit dem Rinderleukose‐Virus. Es wurden jedoch serologische Reaktionen des SBL‐H12578‐Antigens mit einigen Seren von Kalbs‐ und Hautleukosetieren beobachtet. Résumé Etablissement d'une ligne cellulaire à partird'une tumeur de leucémie de la peau d'un bovin et mise en évidence de l'activité du Retrovirus A partir de matérial de biopsie d'une tumeur de leucémie de la peau d'un bovin on a pu établir une ligne cellulaire permanente, denommé SBL‐H12578 ayant le charactère des cellules T. Les cellules provoquent chez les souris nues des tumeurs transplantables. La production des virus du type C par les cellules SBL‐H12578 pouvait être prouvée avec le microscope électronique, la mise en évidence d'un ARN 70 S à l'aide du «simultaneous detection test» et par ailleurs avec la détection «reverse transcriptase» associés à des particules ayant une densité de 1,15 à 1,17 g/ml. Le virus isolé n'avait aucune analogie génétique et sérologique avec le virus de la leucémie enzootique des bovins. Cependant des reactions serologiques de l'antigène des cellules SBL‐H12578 avec certains serums de veaux a l'leucémie et d'autres animaux atteints de l'leucémie de la peau pouvait être observé. Resumen Puesta en marcha de una línea celular de un tumor de leucosis vacuna cutánea y puesta en evidencia de la actividad de los retrovirus A partir de un material de biopsia de un tumor de leucosis cutánea de un vacuno, se estableció una línea celular permanente con caraćter celular T, la cual se denominó SBL‐H12578. Las células engendraban en ratones coritos tumores transplantables. Mediante el microscopio electrónico, la puesta en evidencia de un 70 S RNS en la «prueba simultánea de detección» y la identificación de transcriptasa reversa, la cual se hallaba asociada con partículas de una densidad de 1,15 hasta 1,17 g/ml, se pudo demostrar la producción de virus tipo C por las células SBL‐H12578. El virus aislado no presentaba parentesco genético o serológico con el virus de la leucosis bovina. Sin embargo, se observaron reacciones serológicas del antígeno SBL‐H12578 con algunos sueros de terneros y otros animales con leucosis cutáneas.
