    Abstract: The influence of freezing on the protein profile and quality traits in bovine Longissimus thoracic (LT) muscle was investigated by the data-independent acquisition (DIA) technique. Compared to fresh meat, a total of 262 proteins were identified as differential abundance proteins (DAPs) in four frozen groups (−12 °C, −18 °C, −38 °C, and −80 °C). According to the bioinformatics analysis, most of the DAPs in the significant Go terms and the KEGG pathway were structure proteins and enzymes. Proteome changes in the frozen bovine muscle at −12 °C and −18 °C were more significant than those at −38 °C and −80 °C. The result was consistent with the deterioration trend of the meat quality. The correlation analysis revealed that 17 proteins were correlated closely with the color, shear force, thawing loss, and cooking loss of the frozen meat, which could be used as putative biomarkers for frozen meat quality. MYO18A and ME3 are newly discovered proteins that are associated with frozen beef quality. In addition, CTTN and SERPINB6 were identified in frozen groups, which exhibited a significant inverse correlation with thawing loss (p 〈 0.01). These findings reveal the quality changes induced by freezing at the protein molecular level and provide new insights into the control of quality deterioration.

    Abstract: We analyzed whole-genome bisulfite sequencing (WGBS) and RNA sequencing data of two young (1 year old) and two adult (9 years old) rhesus macaques (Macaca mulatta) to characterize the genomic DNA methylation profile of the thymus and explore the molecular mechanism of age-related changes in the thymus. Combining the two-omics data, we identified correlations between DNA methylation and gene expression and found that DNA methylation played an essential role in the functional changes of the aging thymus, especially in immunity and coagulation. The hypomethylation levels of C3 and C5AR2 and the hypermethylation level of C7 may lead to the high expressions of these genes in adult rhesus macaque thymuses, thus activating the classical complement pathway and the alternative pathway and enhancing their innate immune function. Adult thymuses had an enhanced coagulation pathway, which may have resulted from the hypomethylation and upregulated expressions of seven coagulation-promoting factor genes (F13A1, CLEC4D, CLEC4E, FCN3, PDGFRA, FGF2 and FGF7) and the hypomethylation and low expression of CPB2 to inhibit the degradation of blood clots. Furthermore, the functional decline in differentiation, activation and maturation of T cells in adult thymuses was also closely related to the changes in methylation levels and gene expression levels of T cell development genes (CD3G, GAD2, ADAMDEC1 and LCK) and the thymogenic hormone gene TMPO. A comparison of the age-related methylated genes among four mammal species revealed that most of the epigenetic clocks were species-specific. Furthermore, based on the genomic landscape of allele-specific DNA methylation, we identified several age-related clustered sequence-dependent allele-specific DNA methylated (cS-ASM) genes. Overall, these DNA methylation patterns may also help to assist with understanding the mechanisms of the aging thymus with the epigenome.

    Abstract: Background: This study evaluated the influence of intramuscular connective tissue (IMCT) on structural shrinkage and water loss during cooking. Longissimus thoracis (LT), semimembranosus (SM) and semitendinosus (ST) muscles were cut and boiled for 30 min in boiling water, followed by detection of water holding capacity (WHC), tenderness, fiber volume shrinkage and protein denaturation. Results: Compared with LT and SM, ST had the best WHC and lowest WBSF and area shrinkage ratio. The mobility of immobilized water (T22) was key to holding the water of meat. ST contained the highest content of total and heat-soluble collagen. On the contrary, ST showed the lowest content of cross-links and decorin, which indicate the IMCT strength of ST is weaker than the other two. The heat-soluble collagen is positively correlated to T22. Conclusions: The shrinkage of heat-insoluble IMCT on WHC and WBSF may partly depend on the structural strength changes of IMCT components rather than solely caused by quantitative changes of IMCT.

    Abstract: To understand the pollution characteristics of volatile organic compounds (VOCs) in the glass deep-processing industry, samples were collected using polyvinyl fluoride bags and quickly transferred to summa tanks for GC/MS/FID analysis. The emission characteristics of VOCs, the ozone formation potential and the secondary aerosol formation potential were studied. The results showed that the VOCs emitted by the six enterprises were mainly aromatics and OVOCs, accounting for 35% to 97% of the emissions, with high emission loads of alkanes and halocarbons from individual enterprises. The stack emissions from Enterprise 2 were as high as 38 mg/m3, while the emissions from the remaining five enterprises were all in the range of 1.7~4.1 mg/m3, probably because the terminal treatment facilities were not updated in a timely manner, resulting in excessive stack emissions from Enterprise 2. The characteristic pollutants, including OVOCs, aromatics and alkanes, which are mainly derived from spray painting and gluing, were screened in the six enterprises. Aromatics and OVOCs contributed the most to the ozone formation potential in the six enterprises, with some enterprises having a high contribution from alkanes and alkenes. On the basis of the secondary aerosol formation potential, toluene, benzene, ethylbenzene, o-xylene and m/p-xylene account for 98% of the six enterprises’ emissions. Glass enterprises should prioritise the control of benzene and OVOCs emissions. The glass processing industry mainly emits aromatics, OVOCs and alkanes. Through a preliminary study on the emission characteristics of VOCs in the glass deep-processing industry, we provided basic data for the reduction and control of VOCs in the glass deep-processing industry in China.

    Abstract: Twin-field quantum key distribution (TF-QKD) can break the repeaterless linear bound and possess the measurement-device-independent security, and thus seems very promising in practical applications of quantum secure communication. In most reported TF-QKD protocols, light sources are assumed to possess trusted and fixed photon number distributions (PND), which are unrealistic assumptions in practical applications. Fortunately, the light source monitoring (LSM) method is proposed to circumvent this problem by actively adjusting the attenuation coefficient and monitoring the photon number distribution of light sources. However, the active light source monitoring (ALSM) method may induce additional modulation errors due to imperfect attenuation devices, deteriorating practical performances of TF-QKD systems. In this manuscript, we propose a passive light source monitoring (PLSM) scheme for TF-QKD, and employ the sending-or-not-sending (SNS) TF-QKD as an example for illustration. Simulation results show that our present work can greatly exceed both the original SNS protocol and the ALSM scheme when light source fluctuations and modulation errors are taken into account.

    Abstract: Noctuidae is known to have high species diversity, although the genomic diversity of Noctuidae species has yet to be studied extensively. Investigation of transposable elements (TEs) in this family can improve our understanding of the genomic diversity of Noctuidae. In this study, we annotated and characterized genome-wide TEs in ten noctuid species belonging to seven genera. With multiple annotation pipelines, we constructed a consensus sequence library containing 1038–2826 TE consensus. The genome content of TEs showed high variation in the ten Noctuidae genomes, ranging from 11.3% to 45.0%. The relatedness analysis indicated that the TE content, especially the content of LINEs and DNA transposons, is positively correlated with the genome size (r = 0.86, p-value = 0.001). We identified SINE/B2 as a lineage-specific subfamily in Trichoplusia ni, a species-specific expansion of the LTR/Gypsy subfamily in Spodoptera exigua, and a recent expansion of SINE/5S subfamily in Busseola fusca. We further revealed that of the four TE classes, only LINEs showed phylogenetic signals with high confidence. We also examined how the expansion of TEs contributed to the evolution of noctuid genomes. Moreover, we identified 56 horizontal transfer TE (HTT) events among the ten noctuid species and at least three HTT events between the nine Noctuidae species and 11 non-noctuid arthropods. One of the HTT events of a Gypsy transposon might have caused the recent expansion of the Gypsy subfamily in the S. exigua genome. By determining the TE content, dynamics, and HTT events in the Noctuidae genomes, our study emphasized that TE activities and HTT events substantially impacted the Noctuidae genome evolution.

    Abstract: Greenhouses, as important parts of facility agriculture, can reduce the restrictions on agricultural production imposed by the natural environment and make rational and efficient use of production resources. We conducted long-term, continuous testing of temperature, humidity, light intensity, and CO2 concentration parameters in a heliostat greenhouse in the central and western parts of the Inner Mongolia Autonomous Region, a cold and arid region of northern China. A large amount of data was processed by statistical observation, simulation analysis, and 3D reconstruction to obtain the overall distribution, variation pattern, and mathematical model of the regional greenhouse environment in time and space. The results show that the temperature, humidity, light intensity, and CO2 concentration in the greenhouse have significant daily variation patterns, that there are strong coupling relationships between light intensity–CO2 concentration–time and indoor temperature–light intensity–CO2 concentration, that the coefficients of determination (R2) of the mathematical models are 0.88 and 0.89, and that the standard errors (RMSE) are 49.67 ppm and 45.30 ppm, respectively. The environmental parameters were fitted with high accuracy in order to provide scientific data for the cultivation of heliostats in the region.

    Abstract: In this paper, we propose an end-to-end automatic walking system for construction machinery, which uses binocular cameras to capture images of construction machinery for environmental perception, detects target information in binocular images, estimates the relative distance between the current target and cameras, and predicts the real-time control signal of construction machinery. This system consists of two parts: the binocular recognition ranging model and the control model. Objects within 5 m can be quickly detected by the recognition ranging model, and at the same time, the distance of the object can be accurately ranged to ensure the full perception of the surrounding environment of the construction machinery. The distance information of the object, the feature information of the binocular image, and the control signal of the previous stage are sent to the control model; then, the prediction of the control signal of the construction machinery can be output in the next stage. In this way, the automatic walking experiment of the construction machinery in a specific scenario is completed, which proves that the model can control the machinery to complete the walking task smoothly and safely.
