    Abstract: This study aimed to investigate whether visceral adipose tissue index (VATI) is a significant risk factor for the early recurrence (ER) of HBV-related hepatocellular carcinoma (HCC) (≤5 cm) after hepatectomy. Methods The recruited cohort patients who were positive for hepatitis B virus, presented with surgically confirmed HCC (≤5 cm) from Army Medical University (internal training cohort: n  = 192) and Chongqing Medical University (external validation group: n  = 46). We measured VATI, subcutaneous adipose tissue index (SATI) via computed tomography (CT). ER was defined as recurrence within 2 years after hepatectomy. The impact of parameters on outcome after hepatectomy for HCC was analyzed. Results Univariate analysis showed that alpha-fetoprotein levels ( p  = 0.044), body mass index (BMI) ( p   & lt; 0.001), SATI ( p   & lt; 0.001), and VATI ( p   & lt; 0.001) were significantly different between ER and non-ER groups in internal training cohort. Multivariate analysis identified VATI as an independent risk factor for ER (odds ratio = 1.07, 95% confidence interval: 1.047–1.094, p   & lt; 0.001), with a AUC of 0.802, based on the cut-off value of VATI, which was divided into high risk (≥37.45 cm 2 /m 2 ) and low risk ( & lt;37.45 cm 2 /m 2 ) groups. The prognosis of low risk group was significantly higher than that of high risk group ( p  & lt; 0.001). The AUC value of VATI in external validation group was 0.854. Conclusion VATI was an independent risk factor for the ER, and higher VATI was closely related to poor outcomes after hepatectomy for HBV-related HCC (≤5 cm).

    Abstract: When manufacturers construct a dual-channel distribution system, which includes online and offline sales channels, they need to solve the inventory management problem to ensure supply and reduce inventory costs of the supply chain system. The dual-channel supply chain is the research object, and the inventory decision model is designed to achieve optimal profit when market demand is divided into online and offline demands. The results of the numerical analysis and simulations, conducted using MATLAB, indicate that both the manufacturer and the retailer increase their inventories and that their profits decrease when demand uncertainty increases. Besides, the increase in the online demand ratio causes the increase in the manufacturer’s inventory and reduces the profits of the retailer and the entire supply chain.

    Abstract: Lightning rods have been developed to prevent damage caused by lightning to organisms. However, the biological effect of the current transmitted into the soil through lightning rods is unknown. In this study, we analyzed the effects of lightning on soil properties, the microbial community, and the active components of Pu-erh tea ( Camellia sinensis var. assamica ) near lightning rods. The results showed that the contents of organic matter and available potassium, copper, and calcium in rhizosphere soil near the lightning rod were significantly higher than those in control soil ( P & lt; 0.05), while the contents of total potassium, phosphorus, iron, magnesium, and aluminum decreased. Lightning significantly increased the bacterial diversity of Pu-erh rhizosphere soil compared to control soil samples ( P & lt; 0.05). Sphingomonas, Nitrospira , and Reyranella were significantly enriched in soil samples near the lightning rod compared to soil samples far from the lightning rod. Clusters of Orthologous Groups (COG) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses indicated that adenosine/AMP kinase, chitodextrinase, flavorubredoxin, nucleotide metabolism, and carbohydrate digestion and absorption were significantly enriched in the rhizosphere soil samples near the lightning rod compared to the control samples ( P & lt; 0.05). β diversity analysis indicated the grounding of the lightning rod contributed to the community differentiation of rhizosphere bacteria. Amino acids, polyphenols, and soluble sugar increased in Pu-erh tea near the lightning rod, while the contents of catechin and anthocyanin decreased in Pu-erh tea near the lightning rod compared with the control sample ( P & lt; 0.05). Significant correlations were found among microbial indicators, soil properties, and Pu 'er tea components. This study serves as the first report on the effects of lightning rods on soil properties, microecology, and plant metabolism, which promotes the understanding of the biological effects of lightning, and provides a reference for the rational use of lightning resources.

    Abstract: Increasing evidence has demonstrated that in addition to dysfunction of neuronal circuitry, oligodendroglial dysfunction and/or disruption of white matter integrity are found in the brains of patients with schizophrenia. DNA methylation, a well-established risk factor for schizophrenia, has been demonstrated to cause neuronal dysfunction; however, whether dysregulation of DNA methylation contributes to oligodendroglial/myelin deficits in the pathogenesis of schizophrenia remains unclear. In the present study, by using L-methionine-treated mice, we confirmed that mice with DNA hypermethylation exhibited an anxious phenotype, impaired sociability, and sensorimotor gating deficits. Notably, DNA hypermethylation in oligodendroglial cells led to dysregulation of multiple oligodendroglia-specific transcription factors, which indicated disruption of the transcriptional architecture. Furthermore, DNA hypermethylation caused a reduction of oligodendroglial lineage cells and myelin integrity in the frontal white matter of mice. Taken together, these results indicate that DNA hypermethylation leads to oligodendroglial and/or myelin deficits, which may, at least in part, contribute to schizophrenia-like behaviors in mice. This study provides new insights into the possibility that precise modulation of DNA methylation status in oligodendroglia could be beneficial for the white matter pathology in schizophrenia.

    Abstract: The objective of this research is to investigate the clinical application value of cerebrospinal fluid (CSF) cytology and circulating tumor DNA (ctDNA) in lung adenocarcinoma (LUAD) meningeal metastasis-meningeal carcinomatosis (MC), and to further explore the possible molecular mechanisms and drug treatment targets of LUAD meningeal metastasis by next-generation sequencing (NGS). Methods We retrospectively analyzed LUAD with MC in 52 patients. CSF cytology was carried out using the slide centrifugation precipitation method and May-Grüwald-Giemsa (MGG) staining. Tumor tissue, plasma and CSF ctDNA of some MC patients were detected by NGS. Results Of the 52 MC patients, 46 (88.46%) were positive for CSF cytology and 34 (65.38%) were positive for imaging, with statistically significant differences in diagnostic positivity ( P & lt; 0.05). In 32 of these patients, CSF cytology, cerebrospinal fluid ctDNA, plasma ctDNA and MRI examination were performed simultaneously, and the positive rates were 84.38, 100, 56.25, and 62.50% respectively, the difference was statistically significant ( P & lt; 0.001). Analysis of the NGS profiles of tumor tissues, plasma and CSF of 12 MC patients: the mutated gene with the highest detection rate was epidermal growth factor receptor (EGFR) and the detection rate were 100, 58.33, and 100% respectively in tumor tissues, plasma and CSF, and there were 6 cases of concordance between plasma and tissue EGFR mutation sites, with a concordance rate of 50.00%, and 12 cases of concordance between CSF and tissue EGFR mutation sites, with a concordance rate of 100%. In addition, mutations not found in tissue or plasma were detected in CSF: FH mutation, SETD2 mutation, WT1 mutation, CDKN2A mutation, CDKN2B mutation, and multiple copy number variants (CNV), with the most detected being CDKN2A mutation and MET amplification. Conclusion CSF cytology is more sensitive than traditional imaging in the diagnosis of meningeal carcinomatosis and has significant advantages in the early screening and diagnosis of MC patients. CSF ctDNA can be used as a complementary diagnostic method to negative results of CSF cytology and MRI, and CSF ctDNA can be used as an important method for liquid biopsy of patients with MC, which has important clinical significance in revealing the possible molecular mechanisms and drug treatment targets of meningeal metastasis of LUAD.

    Abstract: Forty-nine susceptible loci have been reported to be significantly associated with vitiligo by genome-wide association studies (GWASs) in European-derived whites. To date, some of these reported susceptibility loci have not yet been validated in the Chinese Han population. The purpose of this study was to examine whether the 16 reported susceptible loci in European-derived whites were associated with vitiligo in the Chinese Han population. Imputation was performed using our previous GWAS dataset by IMPUTE v2.2.2. The 16 imputed top single-nucleotide polymorphisms (SNPs) with suggestive signals, together with the reported SNPs, were genotyped in a total of 2581 patients and 2579 controls by the Sequenom MassARRAY system. PLINK 2.0 software was used to perform association analysis. The dbSNP database, HaploReg, and eQTL data were adopted to annotate the biological function of the SNPs. Finally, four SNPs from three loci were significantly associated with vitiligo, including rs3747517 ( P = 1.29 × 10 –3 , OR = 0.87) in 2q24.2, rs4807000 ( P = 7.78 × 10 –24 , OR = 0.66) and rs6510827 ( P = 3.65 × 10 –5 , OR = 1.19) in 19p13.3, and rs4822024 ( P = 6.37 × 10 –10 , OR = 0.67) in 22q13.2. According to the dbSNP database, rs3747517 is a missense variant of IFIH1 , rs4807000 and rs6510827 are located in TICAM1 , and rs4822024 is located 6 kb upstream of TEF . Further bioinformatics analysis by HaploReg and eQTL found that rs4807000, rs6510827, and rs4822024 are involved in regulating gene expression. Our study revealed the strong association of 2q24.2 (rs3747517), 19p13.3 (rs4807000, rs6510827), and 22q13.2 (rs4822024) with the risk of vitiligo in the Chinese Han population, which implicates common factors for vitiligo across different ethnicities, and helps expand the understanding of the genetic basis of this disease.

    Abstract: The reconstruction of Asian summer monsoon (ASM) changes during the last glacial period is of great significance for better understanding monsoon dynamics. The phase relationship between the Indian summer monsoon (ISM) and East Asian summer monsoon (EASM) subsystems on different timescales is still unclear. The comparative analysis of speleothem records in the ISM region, EASM region, and central China helps to clarify the relationship between the ISM and EASM. Based on the well-dated isotope records of stalagmite DDH-B15 from the Didonghe (DDH) Cave in Hanzhong, Shaanxi, we reconstructed ASM changes during the past 34–13 thousand years before the present (kyr BP). The small average error (61 years) of 18 uranium-series ages enables a precise comparison of the stalagmite δ 18 O record with other well-dated records from the orbital to the millennial timescales. The δ 18 O signal of the DDH-B15 stalagmite is controlled by changes of the low latitude northern hemisphere summer insolation (NHSI) on the orbital timescale. It records cold Heinrich Stadial (HS) and Dansgaard–Oeschger (DO) cycles which are originated from the northern high latitude on the millennial time scale. The δ 18 O changes of stalagmites from the three regions are similar on the millennial and centennial timescales. But on the orbital-suborbital timescale, stalagmite δ 18 O changes during the last glacial cycle have different characteristics. The stalagmite δ 18 O values in eastern China became gradually negative, and the stalagmite δ 18 O values in the Indian monsoon domain showed a increasing trend, but the stalagmite δ 18 O values in Central China adopted an intermediate state between the EASM and ISM. Then we argued that the δ 18 O value of stalagmites in Central China is a mixed signal of the ISM and EASM, which indicates a change of the water vapor source as an important influence on the Chinese stalagmite δ 18 O record.
