    Abstract: Diabetic retinopathy (DR) is a common microvascular complication caused by diabetes mellitus (DM) and is a leading cause of vision impairment and loss among adults. Here, we performed a comprehensive proteomic analysis to discover biomarkers for DR. First, to identify biomarker candidates that are specifically expressed in human vitreous, we performed data-mining on both previously published DR-related studies and our experimental data; 96 proteins were then selected. To confirm and validate the selected biomarker candidates, candidates were selected, confirmed, and validated using plasma from diabetic patients without DR (No DR) and diabetics with mild or moderate nonproliferative diabetic retinopathy (Mi or Mo NPDR) using semiquantitative multiple reaction monitoring (SQ-MRM) and stable-isotope dilution multiple reaction monitoring (SID-MRM). Additionally, we performed a multiplex assay using 15 biomarker candidates identified in the SID-MRM analysis, which resulted in merged AUC values of 0.99 (No DR versus Mo NPDR) and 0.93 (No DR versus Mi and Mo NPDR). Although further validation with a larger sample size is needed, the 4-protein marker panel (APO4, C7, CLU, and ITIH2) could represent a useful multibiomarker model for detecting the early stages of DR.

    Abstract: Purpose. 18 F-FC119S is a positron emission tomography (PET) tracer for imaging β -amyloid (A β ) plaques in Alzheimer’s disease (AD). The aim of this study is to evaluate the efficacy of 18 F-FC119S in quantitating A β deposition in a mouse model of early amyloid deposition (5xFAD) by PET. Method . Dynamic 18 F-FC119S PET images were obtained in 5xFAD ( n = 5 ) and wild-type (WT) mice ( n = 7 ). The brain PET images were spatially normalized to the M. Mirrione T2-weighted mouse brain MR template, and the volumes of interest were then automatically drawn on the cortex, hippocampus, thalamus, and cerebellum. The specific binding of 18 F-FC119S to A β was quantified as the distribution volume ratio using Logan graphical analysis with the cerebellum as a reference tissue. The A β levels in the brain were also confirmed by immunohistochemical analysis. Result . For the 5xFAD group, radioactivity levels in the cortex, the hippocampus, and the thalamus were higher than those for the WT group. In these regions, specific binding was approximately 1.2-fold higher in 5xFAD mice than in WT. Immunohistochemistry supported these findings; the 5xFAD showed severe A β deposition in the cortex and hippocampus in contrast to the WT group. Conclusion . These results demonstrated that 18 F-FC119S PET can successfully distinguish A β depositions in 5xFAD mice from WT.

    Abstract: Metagenomic library was constructed from a deep-sea sediment sample and screened for lipolytic activity. Open-reading frames of six positive clones showed only 33–58% amino acid identities to the known proteins. One of them was assigned to a new group while others were grouped into Families I and V or EstD Family. By employing a combination of approaches such as removing the signal sequence, coexpression of chaperone genes, and low temperature induction, we obtained five soluble recombinant proteins in Escherichia coli . The purified enzymes had optimum temperatures of 30–35°C and the cold-activity property. Among them, one enzyme showed lipase activity by preferentially hydrolyzing p -nitrophenyl palmitate and p -nitrophenyl stearate and high salt resistance with up to 4 M NaCl. Our research demonstrates the feasibility of developing novel lipolytic enzymes from marine environments by the combination of functional metagenomic approach and protein expression technology.

    Abstract: Hepatitis C virus (HCV) infection is a major cause of liver disease, including cirrhosis and hepatocellular carcinoma. Due to significant adverse effects and emergence of resistant strains of currently developed anti-HCV agents, plant extracts have been considered to be potential sources of new bioactive compounds against HCV. The aim of this study was to evaluate the functional effects of triterpenoid saponins contained in the root extract of Platycodon grandiflorum (PG) on viral enzyme activities and replication in both HCV replicon cells and cell culture grown HCV- (HCVcc-) infected cells. Inhibitory activities of triterpenoid saponins from PG were verified by NS5B RNA-dependent RNA polymerase assay and were further confirmed in the context of HCV replication. Six triterpenoid saponins (platycodin D, platycodin D 2 , platycodin D 3 , deapioplatycodin D, deapioplatycodin D 2 , and platyconic acid A), PG saponin mixture (PGSM), were identified as active components exerting anti-HCV activity. Importantly, PGSM exerted synergistic anti-HCV activity in combination with either interferon- α or NS5A inhibitors. We demonstrated that combinatorial treatment of PGSM and IFN- α efficiently suppressed colony formation with significant reduction in drug resistant variant of HCV. These data suggest that triterpenoid saponin may represent a novel anti-HCV therapeutic agent.

    Abstract: Purpose . To estimate the prevalence of glaucoma and costs associated with glaucoma care in South Korea between 2008 and 2013 using the Korean national claims database. Design . Retrospective cross-sectional study from a national claims database. Methods . Patients who were diagnosed with glaucoma between 2008 and 2013 were retrospectively identified in the national claims database using glaucoma diagnostic codes. For each year, the prevalence of glaucoma and direct medical costs associated with glaucoma care were estimated. Result . The prevalence of glaucoma in patients ≥40 years of age increased from 0.79% in 2008 to 1.05% in 2013. The number of patients with glaucoma increased by 54% between 2008 and 2013 (9% average annual increase). The prevalence of glaucoma increased with age and was higher in males than in females. The cost to care for glaucoma patients increased from $16.5 million in 2008 to $29.2 million in 2013, which translated into an 81% increase over the 6 years examined (12.7% average annual increase). Conclusion . The estimated prevalence and socioeconomic burden of glaucoma have steadily increased each year in South Korea. Nevertheless, many glaucoma patients remain undiagnosed in the present study using national claims database.

    Abstract: T helper 17-related cytokines have been implicated in rheumatoid arthritis (RA) pathogenesis. The study aimed to identify cytokines associated with the treatment response of RA patients to tocilizumab (TCZ), a humanized monoclonal antibody against the interleukin- (IL-) 6 receptor. As an independent substudy of the 24-week, randomized, double-blinded CWP-TCZ301 trial of TCZ in RA patients with an inadequate response to disease-modifying antirheumatic drugs, serum levels of cytokines including tumor necrosis factor-alpha, IL-17A, IL-21, IL-23, IL-6, and soluble IL-6 receptor were measured. Baseline IL-17A levels were significantly lower in RA patients who achieved disease activity score 28 (DAS28) remission at 12 weeks of TCZ treatment, compared to patients not in remission. Patients were stratified into IL-17A low group and IL-17A high group. Significantly more patients in the IL-17A low group achieved remission as compared to the IL-17A high group (47.6 versus 17.4%, P = 0.032 ). DAS28 improvement was significantly better in the IL-17A low group than in the IL-17A high group at 12 weeks ( P = 0.045 ) and 24 weeks ( P = 0.046 ) after adjustment. Other baseline cytokines were not associated with treatment response to TCZ. The data demonstrate that low baseline IL-17A levels are associated with better clinical response to TCZ treatment in RA patients.

    Abstract: This study investigates the effect of a new pore filling material, named sodium silicate-based nanosilica (SS), on resisting the diffusion of the chloride ions. The proposed SS is chosen, mainly due to its smaller particle size, compared to the conventional ethyl silicate-based nanosilica. Each particle of SS is chemically treated to have the negative (−) charge on its surface. Four types of mixes with different amounts of partial replacement with fly ash and slag are prepared. Effect of water to binder ratios (0.35, 0.40, and 0.45) is also examined. Test results showed that the inclusion of SS was significantly beneficial for protecting the concrete from chloride attack. At a given strength, the SS inclusion in concrete was up to three times more effective than the control concrete without SS. It is believed that these excellent results are attributed to the small particle size and the chemical surface treatment of SS. In this study, experiments of compressive strength, hydration heat, accelerated neutralization, and sulfate erosion tests were also conducted to find the general effect of SS inclusion on the fundamental properties and durability of concrete.

    Abstract: Wood plastic composites (WPCs) are increasingly being utilized these days due to their excellent mechanical properties and low maintenance cost. Despite these advantages, poor UV resistance and low impact strength are drawbacks. To overcome these shortcomings, coextrusion technology has recently been applied in the production of WPCs and it has been showing good results. However, further research on the combination of different functional enhancements is still needed. This study, therefore, manufactured polypropylene (PP) composites filled with UV stabilizer and inorganic fillers (i.e., nano-CaCO 3 and talc) and then investigated the influence of weathering on the mechanical and morphological properties of the filled composites as a function of filler type and content. UV stabilizer effectively protected the filled composites from UV-induced photodegradation. At 2.5 wt% nano-CaCO 3 (NCC), the NCC particles were well dispersed in the PP matrix, thereby improving the mechanical properties of the filled composites. The best results were observed in 2.5 wt% NCC and 10 wt% talc hybrid filled composites. The composites prepared by adding UV stabilizer to the PP matrix together with NCC and talc exhibited high mechanical properties and improved weathering resistance, and, thus, the combination of NCC, talc, and UV stabilizer in the PP matrix is applicable for shell layer to be used in coextruded WPCs.

    Abstract: Background . To assess if the variants of (R)-alpha-methyl-CoA racemase (AMACR) gene would be associated with the risk of sporadic prostate cancer in ethnically homogenous Koreans. Materials and Methods . We enrolled 194 patients with prostate cancer and 169 healthy controls. A total of 17 single nucleotide polymorphisms of the AMACR gene were selected. The distribution of each genotype and haplotype was analyzed and their association with the incidence of prostate cancer was evaluated. Further, we detected AMACR expression in tumor with immunohistochemistry and analyzed its association with genotype regarding prostate cancer risk. Results . AG or GG genotype of rs2278008 (E277K) tended to lower prostate cancer risk. The minor G allele was found to be a significant allele that decreased the risk of prostate cancer (adjusted OR, 0.57; 95% CI, 0.35–0.93, P value = 0.025). In patients expression AMACR, AG or GG genotype was also significant genotype in terms of prostate cancer risk (adjusted OR, 0.47; 95% CI, 0.26–0.87, P value = 0.017). Further, [GGCGG] haplotype consisted of five coding SNPs of rs2278008, rs34677, rs2287939, rs10941112, and rs3195676 which decreased the risk of prostate cancer ( P value = 0.047). Conclusions . Genetic variations of AMACR are associated with the risk of sporadic prostate cancer that underwent radical prostatectomy in Koreans.

    Abstract: Background and Aims . While several case reports on ischemic colitis (IC) suggest the presence of predisposing causative factors, a few studies have investigated the predisposing factors in IC. This study aimed to identify the characteristics of patients with IC, particularly focusing on the predisposing factors. Methods . We conducted a single-center, retrospective analysis of 159 patients with IC. Clinical characteristics, laboratory data, endoscopic findings, and medical records were reviewed. Data were compared between groups of patients defined according to the predisposing factors. The predisposing factors are defined as temporary states or episodic events occurring within a week before the development of IC such as colonoscopy, enema, use of laxatives, heavy drinking, pancreatitis, shock, and burn. Results . Compared to the group of patients without predisposing factors of IC, the group of patients with predisposing factors was characterized by a relatively higher prevalence of male sex (56.9% versus 33.3%, p = 0.005 ), younger age (60.9 ± 15.4 versus 67.2 ± 13.4 years, p = 0.010 ), lower incidence of hypertension (43.1% versus 60.2%, p = 0.044 ), and fewer risk factors (1.24 ± 1.18 versus 1.82 ± 1.22, p = 0.005 ). Conclusions . Among men with predisposing factors, IC may develop even at a relatively younger age and in the absence of multiple risk factors, suggesting that predisposing factors may be involved in the pathogenesis of IC.
