    Abstract: Evidence had shown the detrimental effect of prostaglandin (PG) E2 in diabetic nephropathy (DN) of STZ-induced type-1 diabetes but its role in the development of DN of type-2 diabetes remains uncertain. The present study was undertaken to investigate the regulation of PGE2 synthetic pathway and the interaction between peroxisome proliferator-activated receptor (PPAR) γ and PGE2 synthesis in the kidneys of db/db mice. Strikingly, urinary PGE2 was remarkably elevated in db/db mice paralleled with the increased protein expressions of COX-2 and mPGES-1. In contrast, the protein expressions of COX-1, mPGES-2, cPGES, and 15-hydroxyprostaglandin dehydrogenase (15-PGDH) were not altered. Following 1-week rosiglitazone (Rosi) therapy, urinary PGE2, but not other prostanoids, was reduced by 57% in parallel with significant reduction of mPGES-1 protein and EP4 mRNA expressions. By immunohistochemistry, mPGES-1 was significantly induced in the glomeruli of db/db mice, which was almost entirely abolished by Rosi. In line with the reduction of glomerular mPGES-1, the glomerular injury score showed a tendency of improvement after 1 week of Rosi therapy. Collectively, the present study demonstrated an inhibitory effect of PPAR γ activation on renal mPGES-1/PGE2/EP4 pathway in type-2 diabetes and suggested that mPGES-1 may potentially serve as a therapeutic target for treating type-2 diabetes-associated DN.

    Abstract: Nitroalkene derivatives of nitro-oleic acid (OA- NO 2 ) are endogenous lipid products with novel signaling properties, particularly the activation of PPARs. The goal of this proposal was to examine the therapeutic potential of this OA- NO 2 in treatment of obesity and obesity-related conditions in obese Zucker rats. The animals were randomly divided to receive OA- NO 2 , oleic acid (OA), both at 7.5  μ g /kg/d, or vehicle ethanol via osmotic mini-pumps for 2 weeks. Following OA- NO 2 treatment, food intake was decreased as early as the first day and this effect appeared to persist throughout the experimental period. At day 14, body weight gain was significantly reduced by OA- NO 2 treatment. This treatment significantly reduced plasma triglyceride and almost normalized plasma free fatty acid and significantly increased plasma high-density lipid (HDL). The plasma TBARS and proteinuria were paralelly decreased. In contrast, none of these parameters were affected by OA treatment. After 14 days of OA- NO 2 treatment, hematocrit, a surrogate of fluid retention associated with PPAR γ agonists, remained unchanged. Together, these data demonstrated that OA- NO 2 may offer an effective and safe therapeutic intervention for obesity and obesity-related conditions.

    Abstract: In order to achieve the goal of ecological restoration, plant irrigation, and water retention in the external dump of open-pit coal mine in the arid desert area, it is proposed to use the mudstone in the stripped material to reconstruct the surface aquiclude and improve the water holding capacity of the topsoil. By taking the Hongshaquan Open-Pit Coal Mine as the study object, the red mudstone of mining level +650 was selected as the topsoil aquiclude material through the geological survey. XRD diffraction experiments are used to determine the composition of red mudstone including kaolinite mineral, quartz, potash feldspar, albite, and illite. The moisture content of the red mudstone is 4.16% as measured by the indoor drying experiment. And the particle size of 0.5 mm, 1 mm, and 2 mm and the thickness of 5 cm, 10 cm, and 20 cm rock samples were selected to conduct the indoor soil column experiment. Three indicators of initial infiltration rate, stable infiltration rate, and average infiltration rate were obtained according to the analysis and calculation of the water column drop rate, wet peak drop rate, and cumulative infiltration rate so that the permeability law and coefficient of each group of tests can be known. The finite element transient analysis theory and numerical simulation method were adopted to verify the results of physical experiments. The research results show that +650 level red mudstone has a strong antiseepage ability, and the smaller the particle size, the better the antiseepage performance. For 0.5 mm and 2 mm particle sizes, as the paving height increases, the water-proof effect is better. The laying height of 1 mm particle size has no obvious influence on the water barrier effect. The physical simulation was confirmed to have the same result trend as the numerical simulation, both of which are quadratic functions with the error within a reasonable range.

    Abstract: Accumulating evidence demonstrated that hydrogen sulfide (H 2 S) is highly involved in inflammation, oxidative stress, and apoptosis and contributes to the pathogenesis of kidney diseases. However, the role of H 2 S in cisplatin nephrotoxicity is still debatable. Here we investigated the effect of GYY4137, a novel slow-releasing H 2 S donor, on cisplatin nephrotoxicity in mice. Male C57BL/6 mice were pretreated with GYY4137 for 72 h prior to cisplatin injection. After cisplatin treatment for 72 h, mice developed obvious renal dysfunction and kidney injury as evidenced by elevated blood urea nitrogen (BUN) and histological damage. Consistently, these mice also showed increased proinflammatory cytokines such as TNF- α , IL-6, and IL-1 β in circulation and/or kidney tissues. Meanwhile, circulating thiobarbituric aid-reactive substances (TBARS) and renal apoptotic indices including caspase-3, Bak, and Bax were all elevated. However, application of GYY4137 further aggravated renal dysfunction and kidney structural injury in line with promoted inflammation, oxidative stress, and apoptotic response following cisplatin treatment. Taken together, our results suggested that GYY4137 exacerbated cisplatin-induced nephrotoxicity in mice possibly through promoting inflammation, oxidative stress, and apoptotic response.
