    Abstract: In both mouse models and clinical patients with lupus, autophagy levels were significantly elevated and correlated with disease activity. Furthermore, autophagy can promote the survival of B and T cells, plasma cell differentiation, and antibody production. These results suggest that autophagy may promote the progression of lupus by regulating the survival of autoreactive immune cells. Therefore, we aimed at studying whether suppressing autophagy can modulate lupus progression in vivo . First, we found that the autophagy levels in splenocytes and lymphocytes of peripheral blood (PB) were elevated and positively correlated with disease severity in lupus-prone mice. The shAtg5-lentivirus, which effectively inhibits autophagy in vitro , was then injected into the lupus-prone mice. Autophagy levels in lymph node cells and PB lymphocytes were reduced following Atg5 suppression. We also found that lymphadenopathy and the numbers of plasma cells, CD4 - CD8 - , and CD4 + T cells decreased in mice treated with the shAtg5-lentivirus. The mice treated with shAtg5-lentivirus exhibited lower levels of proteinuria, serum anti-dsDNA antibody, B-cell activating factor (BAFF), and glomerular immune complex deposition. Therefore, targeting autophagy to moderate overactivated autophagy in immune cells seems to be a novel strategy for combination therapy of lupus.

    Abstract: Hypoxia-inducible factor (HIF)-1 α and vascular endothelial growth factor (VEGF) are important angiogenic factors in human cancers. Relative to VEGF-C, prognostic significance of VEGF-D expression and its association with HIF-1 α expression remain elusive in esophageal squamous cell cancer (ESCC). We studied expression of HIF-1 α and VEGF-D using immunohistochemistry in 85 resected ESCC specimens and correlated results with patients' clinicopathologic parameters and survival. Association between expression of HIF-1 α and VEGF-D was investigated using a concordance analysis. High expression of HIF-1 α and VEGF-D was observed in 52 (61.2%) and 56 (65.9%) patients, respectively. HIF-1 α expression correlated well with tumor stage ( P = 0.041), whereas VEGF-D expression correlated with tumor stage ( P = 0.027) and N status ( P = 0.019). Groups of high HIF-1 α and VEGF-D showed worse survivals than those of low expression ( P = 0.002 and 0.001, respectively). Multivariate analysis supported expression of HIF-1 α and VEGF-D as significant survival predictors ( P = 0.044 and 0.035, respectively). A concordance rate of 69.5% was observed between expression of HIF-1 α and VEGF-D. In conclusion, protein expression of HIF-1 α and VEGF-D are independent prognostic predictors. An association between expression of HIF-1 α and VEGF-D suggests that these two angiogenic factors are essential in progression of ESCC.

    Abstract: Purpose . Urothelial carcinoma-associated 1 (UCA1) has been reported to be overexpressed and correlated with progression in various cancers. However, the association between UCA1 expression and some clinicopathological features of digestive system malignancies, such as metastasis and survival, remains inconclusive. Therefore, a meta-analysis was performed to investigate the clinical significance of UCA1 in digestive system malignancies. Methods . Relevant literatures were searched in PubMed, Web of Science, Cochrane Library, and Embase databases updated to May 2016. Results . A total of 1089 patients from 10 studies were included in this meta-analysis. Meta-analysis results showed that digestive system malignancy patients with UCA1 overexpression were significantly more susceptible to developing lymph node metastasis (LNM) (OR = 1.85, 95% CI: 1.28–2.67) and distant metastasis (DM) (OR = 3.14, 95% CI: 1.77–5.58) and suffer from poor overall survival (OS) (HR = 2.31, 95% CI: 1.89–2.82, univariate analysis; HR = 2.24, 95% CI: 1.69–2.98, multivariate analysis) and poor disease-free survival (DFS) (HR = 2.65, 95% CI: 1.59–4.43, univariate analysis; HR = 2.50, 95% CI: 1.62–3.86, multivariate analysis). Conclusion . UCA1 overexpression was correlated with LNM, DM, poor OS, and poor DFS. UCA1 may serve as an indicator for metastasis and poor prognosis in digestive system malignancies.

    Abstract: To improve the detection rate and reduce the correction error of abnormal data for water quality, an outlier detection and correction method is proposed based on the improved Variational Mode Decomposition (improved VMD) and Least Square Support Vector Machine (LSSVM) algorithms. The correlation coefficient is introduced, for solving the optimal parameter k of VMD algorithm, and an improved VMD algorithm is obtained. Combined with LSSVM algorithm, the outliers of water quality can be detected and repaired. This method is applied for the detection and correction of water quality monitoring outliers using dissolved oxygen which is retrieved from the water quality monitoring station in Hangzhou, Zhejiang Province, China. The result shows that the improved VMD algorithm is of higher detection rate and lower error rate than those of Empirical Mode Decomposition (EMD) and Ensemble Empirical Mode Decomposition (EEMD). The LSSVM algorithm increases the fitting accuracy and decreases correction error in comparison with SVM and BP neural network, which provides important references for the implementation of environmental protection measures.

    Abstract: Background . Pathogenic infection in broilers has become an important issue in the development of poultry industry. Xylooligosaccharides released from xylan via xylanase and fermented polysaccharide of Hericium caputmedusae (FPHC) have antimicrobial potential against many pathogens. Objective . We aimed to explore the effects of xylanase and FPHC on pathogenic infection in the broilers ( Gallus gallus domesticus ). Methods . Three hundred and thirty 21-day male broilers were assigned into four groups: control group (CG, basic diet), xylanase group (XG, basic diet + xylanase), FPHC group (HG, basic diet + FPHC), and XHG group (basic diet + xylanase + FPHC). Average daily feed intake (ADFI) and daily gain (ADG) were measured. Microflora from broiler feces was analyzed using 16S rRNA sequencing. Serum tumor necrosis factor- (TNF-) α , interleukin-1 β (IL-1 β ), IL-1 receptor antagonist (IL-1ra), IL-10, total antioxidant capacity (T-AOC), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and malondialdehyde (MDA) contents were detected using kits. The variables were compared using the Student t -test between two groups. Results . Microbiological investigations showed that 75% of broilers were affected by bacterial pathogens in the CG group, most notably by coagulase-negative staphylococci. Comparatively, 15%, 26%, and 5% of broilers were affected by bacterial pathogens in the XG, HG, and XHG groups, respectively. Xylanase and FPHC treatment increased the ratio of ADG to ADFI and antioxidant capacity by increasing the levels of T-AOC, SOD, and GSH-Px and reducing the levels of MDA ( P 〈 0.05 ). Xylanase and FPHC treatment improved anti-inflammatory capacity by increasing serum levels of IL-1ra and IL-10 and reducing the levels of IL-1 β and TNF- α . On the other hand, the treatment increased probiotic concentration of Bacillus licheniformis , Bacillus subtilis , and Lactobacillus plantarum ( P 〈 0.05 ), which were also proved in cell culture. Conclusions . Xylanase and FPHC ameliorate pathogen infection by increasing antioxidant and anti-inflammatory activities of broilers via the increase of probiotics.

    Abstract: In this paper, stope structure optimization during sublevel caving mining is considered under the condition that the isolated draw zones (IDZs) are nonstandard ellipsoid, which is realized by dynamically adjusting the arrangement of IDZs and quantifying the degree of intersection of IDZs according to an ore profit and loss calculation model. A dynamic intersecting arrangement model based on IDZs was proposed, which can dynamically adjust the sublevel height and drift spacing according to the ore-rock bulk flow parameters, economic indicators, occurrence condition of the ore body, drilling machine, and so forth. Based on the model, the range of drift spacing, the lower volume of crestal residual ore, and the higher volume of mixing waste rock are calculated. By deducing the function of ore profit and loss, a calculation model for ore profit and loss is established to quantify the degree of intersection of IDZs and determine the best stope structure. Using the constructed dynamic intersecting arrangement model, a stope structure of −213 m to −303 m in the Yanqianshan Iron Mine was designed, with a sublevel height of 22.5 m and a drift spacing of 20.5 m. A physical drawing model was designed, and three physical simulation experiment schemes were conducted to compare and analyse the ore loss and dilution of the intersecting arrangement model and the traditional tangent arrangement model. The results showed that the loss rate decreased by 3.66% and the dilution rate increased by only 0.22%, thus verifying the effectiveness and applicability of the model to optimize the stope structure.
