    Abstract: Objective. This study systematically explored the mechanism of Rhizoma Coptidis-Eupatorium fortunei in treating type 2 diabetes mellitus (T2DM) by using network pharmacology and molecular docking methods. Methods. The TCMSP database was used to screen out the active ingredients and related targets of Rhizoma Coptidis-Eupatorium fortunei (R-E) drug pair. GeneCards, OMIM, DrugBank, and other databases were used to screen the related targets of T2DM, and then, the UniProt database was used to standardize the relevant targets of T2DM. Then, the Venn analysis was performed on the active ingredient-related targets and disease-related targets of R-E drugs to find the intersection targets. Using the STRING database and Cytoscape software, the PPI network and “drug-active ingredient-target-disease” network are constructed by intersecting targets and corresponding active ingredients. Through the cluster profiler package in the R software, GO function enrichment analysis and KEGG pathway enrichment analysis were carried out on the intersection targets and the screened core targets, and the prediction results were verified by molecular docking. Results. Taking OB ≥ 30 % and DL ≥ 0.18 as the standard, a total of 25 effective active ingredients of R-E drug pairs were screened, including berberine, palmatine, coptisine, and so on. After corresponding, 19 effective chemical components and 284 targets of the R-E drug pair were obtained. After searching multiple disease databases, 1289 T2DM-related targets were screened. After the summary, 159 common targets were obtained in this study. Finally, in the bioinformatics analysis, this study concluded that quercetin, luteolin, berberine, palmatine, and coptisine are the main chemical components of the R-E drug pair. ESR1, MAPK1, AKT1, TP53, IL6, and JUN are the important core targets. GO and KEGG enrichment analyses showed that Rhizoma Coptidis-Eupatorium fortunei could improve T2DM by regulating multiple biological processes and pathways. Molecular docking results showed that berberine, palmatine, and coptisine had higher binding to the core target, and MAPK1, AKT1, and IL6 could stably bind to the active ingredients of Rhizoma Coptidis-Eupatorium fortunei. Conclusion. Rhizoma Coptidis-Eupatorium fortunei may have therapeutic effects on T2DM such as anti-inflammatory and regulating glucose and lipid metabolism through multiple components, multiple targets, and multiple signaling pathways, which provides a scientific basis for further research on the hypoglycemic effect of Rhizoma Coptidis-Eupatorium fortunei drug pair.

    Abstract: Lumbar intervertebral disc degeneration (IDD) has been the major contributor to low back pain (LBP). IDD is an chronic inflammation process, with the activation of plentiful inflammation-related cytokines and ECM degradation-related enzymes. In the past few years, hypertension has been reported to correlate with LBP. In addition, the local tissue renin-angiotensin system (tRAS) has been identified in multiple tissues, including the spinal cord, skin, kidney, heart, and bone. Recently, tRAS has also been established in both bovine and human intervertebral disc tissues, especially in the degenerated disc tissue. However, the exact of tRAS and IDD remains unknown. In this present study, proteomic analysis, molecular biology analysis, and animal model were all used. Firstly, we revealed that tRAS was excessively activated in the human degenerated intervertebral disc tissue via proteomic analysis and molecular biology analysis. Then, in vitro experiment suggested that Ang II could decrease the cell viability of human NP cells and promote NP cell apoptosis, senescence, oxidative stress, and NLRP3 activation in human NP cells. In addition, Ang II could also trigger degeneration and fibrosis phenotype in human NP cells. Finally, the animal model demonstrated that the local activated ACE/Ang II axis in the NP tissue could accelerate IDD in aging spontaneously hypertensive rats (SHR). Collectively, the degenerated intervertebral disc tissue showed excessively activated tRAS, and local activated tRAS could induce NP cell senescence, apoptosis, oxidative stress, and inflammatory reaction to promote IDD. These biological effects of Ang II on human NP cells may provide novel insight into further treatment of IDD.

    Abstract: Low back pain (LBP) has been a wide public health concern worldwide. Among the pathogenic factors, intervertebral disc degeneration (IDD) has been one of the primary contributors to LBP. IDD correlates closely with inflammatory response and oxidative stress, involving a variety of inflammation-related cytokines, such as interleukin 1 beta (IL-1β), which could result in local inflammatory environment. Ulinastatin (UTI) is a kind of acidic protein extracted from human urine, which inhibits the release of tumor necrosis factor alpha (TNF-α) and other inflammatory factors to protect organs from inflammatory damage. However, whether this protective effect of UTI on human nucleus pulposus (NP) exists, and how UTI affects the biological behaviors of human NP cells during IDD remain elusive. In this current study, we revealed that UTI could improve the viability of NP cells and promote the proliferation of NP cells. Additionally, UTI could protect human NP cells via ameliorating IL-1β-induced apoptosis, inflammatory response, oxidative stress, and extracellular matrix (ECM) degradation. Molecular mechanism analysis suggested that the protective effect from UTI on IL-1β-treated NP cells were through activating nuclear factor- (erythroid-derived 2-) like 2 (Nrf2)/heme oxygenase-1 (HO-1) signaling pathway and the suppression of NF-κB signaling pathway. Therefore, UTI may be a promising therapeutic medicine to ameliorate IDD.

    Abstract: Objectives . This study aims to assess the effectiveness and safety of moxibustion for the correction of nonvertex presentation. Methods . Records without language restrictions were searched up to February 2013 for randomized controlled trials (RCTs) comparing moxibustion with other therapies in women with a singleton nonvertex presentation. Cochrane risk of bias criteria were used to assess the methodological quality of the trials. Results . Seven of 392 potentially relevant studies met the inclusion criteria. When moxibustion was compared with other interventions, a meta-analysis revealed a significant difference in favor of moxibustion on the correction of nonvertex presentation at delivery (risk ratio (RR) 1.29, 95% confidence interval (CI) 1.12 to 1.49, and I 2 = 0 ). The same findings applied to the cephalic presentation after cessation of treatment (RR 1.36, 95% CI 1.08 to 1.71, and I 2 = 80 %). A subgroup analysis that excluded two trials with a high risk of bias also indicated favorable effects (RR 1.63, 95% CI 1.42 to 1.86, and I 2 = 0 %). With respect to safety, moxibustion resulted in decreased use of oxytocin. Conclusion . Our systematic review and meta-analysis suggested that moxibustion may be an effective treatment for the correction of nonvertex presentation. Moreover, moxibustion might reduce the need for oxytocin.

    Abstract: With the continuous enhancement of computer technology, digital technology brings more convenience to people with its powerful functions. After the digital technology is applied to the design field, it greatly expands the creative space of art design. In the digital age, fabric printing and dyeing design needs to be combined with digital technology under the guidance of art theory. Traditional flat patterns can no longer satisfy people’s pursuit of visual illusion patterns with a sense of space and three-dimensionality. The use of computer representation can realize the innovative artistic design and can directly generate various highly creative visual images in the computer according to the theme and creative needs. We input the digital data related to the fabric, such as graphics, photographic photos, logos, fonts, and textures, into the computer and then use various application software to perform various special effects on them to produce creative, unique, and surprising picture effects. In the aspect of exploring the meaning of the theme, the application of spatial expression means is studied, and practical research is carried out to guide the design of works with spatial beauty and creativity. In view of the neglect of artistic design, on the basis of studying the space art expression of traditional printing and dyeing process patterns, the traditional space art expression is combined with computer image processing technology. And special techniques are used to create differences in various textile fabrics and garments. We use the software of digital art design to carry out the artistic creation practice of spatial expression. Through the application of computer software functions, combined with the guidance of spatial expression techniques, the design comparison of patterns is carried out, which provides design practice ideas for the integration and innovation of traditional printing and dyeing technology and modern design. The implementation of the visual material colouring programming system reduces the coupling between artists and programmers and reduces the coupling between art collections and producers. The colouring program in the process of art pattern development improves the efficiency of art design.

    Abstract: Purpose . Urothelial carcinoma-associated 1 (UCA1) has been reported to be overexpressed and correlated with progression in various cancers. However, the association between UCA1 expression and some clinicopathological features of digestive system malignancies, such as metastasis and survival, remains inconclusive. Therefore, a meta-analysis was performed to investigate the clinical significance of UCA1 in digestive system malignancies. Methods . Relevant literatures were searched in PubMed, Web of Science, Cochrane Library, and Embase databases updated to May 2016. Results . A total of 1089 patients from 10 studies were included in this meta-analysis. Meta-analysis results showed that digestive system malignancy patients with UCA1 overexpression were significantly more susceptible to developing lymph node metastasis (LNM) (OR = 1.85, 95% CI: 1.28–2.67) and distant metastasis (DM) (OR = 3.14, 95% CI: 1.77–5.58) and suffer from poor overall survival (OS) (HR = 2.31, 95% CI: 1.89–2.82, univariate analysis; HR = 2.24, 95% CI: 1.69–2.98, multivariate analysis) and poor disease-free survival (DFS) (HR = 2.65, 95% CI: 1.59–4.43, univariate analysis; HR = 2.50, 95% CI: 1.62–3.86, multivariate analysis). Conclusion . UCA1 overexpression was correlated with LNM, DM, poor OS, and poor DFS. UCA1 may serve as an indicator for metastasis and poor prognosis in digestive system malignancies.

    Abstract: In both mouse models and clinical patients with lupus, autophagy levels were significantly elevated and correlated with disease activity. Furthermore, autophagy can promote the survival of B and T cells, plasma cell differentiation, and antibody production. These results suggest that autophagy may promote the progression of lupus by regulating the survival of autoreactive immune cells. Therefore, we aimed at studying whether suppressing autophagy can modulate lupus progression in vivo . First, we found that the autophagy levels in splenocytes and lymphocytes of peripheral blood (PB) were elevated and positively correlated with disease severity in lupus-prone mice. The shAtg5-lentivirus, which effectively inhibits autophagy in vitro , was then injected into the lupus-prone mice. Autophagy levels in lymph node cells and PB lymphocytes were reduced following Atg5 suppression. We also found that lymphadenopathy and the numbers of plasma cells, CD4 - CD8 - , and CD4 + T cells decreased in mice treated with the shAtg5-lentivirus. The mice treated with shAtg5-lentivirus exhibited lower levels of proteinuria, serum anti-dsDNA antibody, B-cell activating factor (BAFF), and glomerular immune complex deposition. Therefore, targeting autophagy to moderate overactivated autophagy in immune cells seems to be a novel strategy for combination therapy of lupus.

    Abstract: Circular RNAs are a class of RNAs with a covalently closed configuration, and several members of them have been reported to be capable of regulating various biological processes and predicting the outcome of disease. Among them, circular RNA circ-ITCH has been identified to be aberrantly expressed and associated with disease progression in diverse cancers. However, the correlation of circ-ITCH expression with clinicopathological features, as well as the prognosis of cancers, remains inconclusive. Therefore, a meta-analysis was performed to investigate the clinical significance of circ-ITCH in cancers by systematically summarizing all eligible literatures. Up to August 31, 2020, relevant articles were searched in PubMed, Web of Science, Cochrane library, Embase, CNKI, and Wanfang databases. Pooled hazard ratios (HRs) and odds ratios (ORs) with corresponding 95% confidence intervals (CIs) were calculated. A total of 1604 patients from 14 studies were included in this meta-analysis. The results showed that cancer patients with low circ-ITCH expression were more susceptible to develop lymph node metastasis ( OR = 2.25 , 95% CI: 1.67-3.02, p ≤ 0.01 ), larger tumor size ( OR = 3.01 , 95% CI: 2.01-4.52, p ≤ 0.01 ), advanced TNM stage ( OR = 2.82 , 95% CI: 1.92-4.14, p ≤ 0.01 ), and poor overall survival (OS) ( HR = 2.45 , 95% CI: 2.07–2.90, p ≤ 0.01 , univariate analysis; HR = 2.69 , 95% CI: 1.82-3.96, p ≤ 0.01 , multivariate analysis). Thus, low circ-ITCH expression was significantly associated with aggressive clinicopathological features and unfavorable outcome in various cancers. Therefore, circ-ITCH may serve as a molecular therapy target and a prognostic marker in human cancers.

    Abstract: Objective. Deep vein thrombosis (DVT) is the third-largest cardiovascular disease, and accurate segmentation of venous thrombus from the black-blood magnetic resonance (MR) images can provide additional information for personalized DVT treatment planning. Therefore, a deep learning network is proposed to automatically segment venous thrombus with high accuracy and reliability. Methods. In order to train, test, and external test the developed network, total images of 110 subjects are obtained from three different centers with two different black-blood MR techniques (i.e., DANTE-SPACE and DANTE-FLASH). Two experienced radiologists manually contoured each venous thrombus, followed by reediting, to create the ground truth. 5-fold cross-validation strategy is applied for training and testing. The segmentation performance is measured on pixel and vessel segment levels. For the pixel level, the dice similarity coefficient (DSC), average Hausdorff distance (AHD), and absolute volume difference (AVD) of segmented thrombus are calculated. For the vessel segment level, the sensitivity (SE), specificity (SP), accuracy (ACC), and positive and negative predictive values (PPV and NPV) are used. Results. The proposed network generates segmentation results in good agreement with the ground truth. Based on the pixel level, the proposed network achieves excellent results on testing and the other two external testing sets, DSC are 0.76, 0.76, and 0.73, AHD (mm) are 4.11, 6.45, and 6.49, and AVD are 0.16, 0.18, and 0.22. On the vessel segment level, SE are 0.95, 0.93, and 0.81, SP are 0.97, 0.92, and 0.97, ACC are 0.96, 0.94, and 0.95, PPV are 0.97, 0.82, and 0.96, and NPV are 0.97, 0.96, and 0.94. Conclusions. The proposed deep learning network is effective and stable for fully automatic segmentation of venous thrombus on black blood MR images.
