    Abstract: Objective . To investigate the risk factors for hepatic steatosis in chronic hepatitis B (CHB), to determine its correlation with liver necroinflammation and fibrosis and response to peginterferon alpha-2a (PEG-IFN α -2a) antiviral therapy, and to explore the mechanisms underlying the poor antiviral effect of PEG-IFN α -2a in CHB patients with hepatic steatosis. Methods . We analysed the impact of hepatic steatosis on the antiviral effect of PEG-IFN α -2a on CHB patients in a cohort of 226 patients who underwent pretherapeutic liver biopsy. To assess the complete response (CR), virological response (VR), and biochemical response (BR), the 226 patients were treated with PEG-IFN α -2a for 48 weeks and were followed-up for 24 weeks. The expressions of hepatitis B surface antigen (HBsAg) and hepatitis B core antigen (HBcAg) in the liver tissue were detected in all patients to explore the possible mechanism of hepatic steatosis with regard to antiviral effects. Results . The patients were divided into four groups based on the severity of hepatic steatosis: 119 with no steatosis, 76 with mild steatosis, 22 with moderate steatosis, and 9 with severe steatosis. In the hepatic steatosis groups, the proportions of male patients, patients aged 〉 40 years, patients with hyperuricaemia, patients with a BMI 〉 23   kg / m 2 , and total cholesterol (TC), triglyceride (TG), glucose (GLU), and uric acid (UA) levels were significantly higher than those in the group without steatosis, whereas the alanine aminotransferase (ALT) and aspartate transaminase (AST) levels were significantly lower than those in the group without steatosis. The multivariate analysis results indicated that a BMI 〉 23   kg / m 2 was independently associated with CHB patients with hepatic steatosis; the levels of baseline AST and UA were independently associated with CHB patients with significant hepatic steatosis, and the baseline AST level was independently associated with significant liver fibrosis. After 48 weeks of treatment and 24 weeks of follow-up, the rates of CR, VR, and BR had gradually decreased, whereas the severity of hepatic steatosis had increased. Conclusion . Hepatic steatosis can reduce the efficacy of PEG-IFN α -2a in the treatment of CHB patients, and its mechanism may be related to the different HBcAg expression patterns in liver tissue.

    Abstract: Ethyl carbamate (EC), a probable human carcinogen, occurs widely in many fermented foods. Previous studies indicated that EC-induced cytotoxicity was associated with oxidative stress. Wild raspberries are rich in polyphenolic compounds, which possess potent antioxidant activity. This study was conducted to investigate the protective effect of wild raspberry extracts produced before (RE) and after in vitro simulated gastrointestinal digestion (RD) on EC-induced oxidative damage in Caco-2 cells. Our primary data showed that ethyl carbamate could result in cytotoxicity and genotoxicity in Caco-2 cells and raspberry extract after digestion (RD) may be more effective than that before digestion (RE) in attenuating toxicity caused by ethyl carbamate. Further investigation by fluorescence microscope revealed that RD may significantly ameliorate EC-induced oxidative damage by scavenging the overproduction of intracellular reactive oxygen species (ROS), maintaining mitochondrial function and preventing glutathione (GSH) depletion. In addition, HPLC-ESI-MS results showed that the contents of identified polyphenolic compounds (esculin, kaempferol O-hexoside, and pelargonidin O-hexoside) were remarkably increased after digestion, which might be related to the better protective effect of RD. Overall, our results demonstrated that raspberry extract undergoing simulated gastrointestinal digestion may improve the protective effect against EC-induced oxidative damage in Caco-2 cells.

    Abstract: Background. Both Lowe syndrome and Dent-2 disease are caused by variants in the OCRL gene. However, the reason why patients with similar OCRL gene mutations presented with different phenotypes remains uncertain. Methods. Children with hemizygous pathogenic or likely pathogenic variants in OCRL were compiled from published and unpublished consecutive cases from China. Furthermore, a Chi-square test was employed to analyze the correlation of the location and types of mutations on the phenotype of children with Lowe syndrome or Dent-2 disease. Results. Among the total 83 patients, 70.8% (34/48) cases of Lowe syndrome presented with truncating mutations, while only 31.4% (11/35) cases of Dent-2 disease presented with truncating mutation (Χ2 = 12.662; P 〈 0.001 ). Meanwhile, the majority of mutations in Dent-2 disease are located in Exon 2–12 (21/35, 60.0%), while the majority of mutations in Lowe syndrome are located in Exon 13–23 (39/48, 81.3%; Χ2 = 14.922; P 〈 0.001 ). Conclusions. Truncating mutations of the OCRL gene were more common in patients with Lowe syndrome than in Dent-2 disease, while mutation is more likely located at exon 2–12 in Dent-2 disease than that in Lowe syndrome. The type and location of mutation are important indicators for the phenotypes in patients with OCRL mutation. This is a large cohort study analyzing the genotype-phenotype correlation in patients with Lowe syndrome and Dent-2 disease in China. Our data may improve the interpretation of new OCRL variants and genetic counseling. Furthermore, a large international study would be necessary to illustrate the genotype-phenotype correlation in patients with OCRL mutations.

    Abstract: This work is aimed to study the possibility of recycling plastic waste (polypropylene (PP)) as aggregate instead of sand in the manufacturing of mortar or concrete. For this, an experimental study was carried out to evaluate the influence of nano-SiO 2 and recycled PP plastic particles' content on physical, mechanical, and shrinkage properties and microstructure of the mortars with recycled PP plastic particles. The sand is substituted with the recycled PP plastic particles at dosages (0%, 20%, 40%, and 60% by volume of the sand). The nano-SiO 2 content is 5% by weight of cement. The physical (porosity, water absorption, and density), mechanical (compressive and flexural strength) and shrinkage properties of the mortars were evaluated, and a complementary study on microstructure of the interface between cementitious matrix and PP plastic particles was made. The measurements of physical and mechanical properties showed that PP-filled mortar had lower density and better toughness (higher ratio of flexural strength to compressive strength). However, the compressive strength and flexural strength of PP-filled mortar is reduced, and the porosity, water absorption, autogenous shrinkage, and dry shrinkage increased as compared to normal cement mortar. The addition of nano-SiO 2 reduced the porosity, water absorption, and drying shrinkage of PP-filled mortar and effectively improved the mechanical properties, but increased its autogenous shrinkage. A microscopic study of the interfacial zone (plastic-binder) has shown that there is poor adhesion between PP plastic particles and cement paste. From this work, it is found that recycled PP plastic waste has a great potential to be a construction material. It can be used as partial replacement of natural aggregates instead.

    Abstract: Myeloid-derived suppressor cells (MDSCs) have attracted attention due to their important role in inflammation. Several studies have investigated the involvement of MDSCs in chronic liver disease. However, due to the difference of MDSC phenotypes, patient types, and sample sources among the studies, the results are inconsistent and controversial. We took advantage of a large well-defined cohort of 98 (24 patients with CHB, 18 with NAFLD, 13 with HCC, 16 with PBC, and 27 with AIH) patients with liver inflammation and 12 healthy controls to investigate the expression of MDSCs, and the relationships between the expression of hepatic MDSCs and the clinical characteristics were analyzed. We found that the expression of CD11b+CD33+ MDSCs is closely related to chronic liver disease and positively correlated with clinical parameters such as ALT, AST, and globulin. Ultimately, the present study suggests that hepatic CD11b+CD33+ MDSCs are increased in HCC and AIH and positively correlate with the liver stages of hepatitis activity and liver fibrosis stage.

    Abstract: Cardiovascular disease remains a substantial cause of morbidity and mortality in the developed world and is becoming an increasingly important cause of death in developing countries too. While current cardiovascular treatments can assist to reduce the risk of this disease, a large number of patients still retain a high risk of experiencing a life-threatening cardiovascular event. Thus, the advent of new treatments methods capable of reducing this residual risk remains an important healthcare objective. This paper proposes a deep learning-based method for section recognition of cardiac ultrasound images of critically ill cardiac patients. A convolution neural network (CNN) is used to classify the standard ultrasound video data. The ultrasound video data is parsed into a static image, and InceptionV3 and ResNet50 networks are used to classify eight ultrasound static sections, and the ResNet50 with better classification accuracy is selected as the standard network for classification. The correlation between the ultrasound video data frames is used to construct the ResNet50 + LSTM model. Next, the time-series features of the two-dimensional image sequence are extracted and the classification of the ultrasound section video data is realized. Experimental results show that the proposed cardiac ultrasound image recognition model has good performance and can meet the requirements of clinical section classification accuracy.
