    Abstract: A highly efficient Agrobacterium-mediated transformation method is needed for the molecular study of model tree species such as hybrid poplar 84K (Populus alba × P. glandulosa cv. ‘84K’). In this study, we report a callus-based transformation method that exhibits high efficiency and reproducibility. The optimized callus induction medium (CIM1) induced the development of calli from leaves with high efficiency, and multiple shoots were induced from calli growing on the optimized shoot induction medium (SIM1). Factors affecting the transformation frequency of calli were optimized as follows: Agrobacterium concentration sets at an OD600 of 0.6, Agrobacterium infective suspension with an acetosyringone (AS) concentration of 100 µM, infection time of 15 min, cocultivation duration of 2 days and precultivation duration of 6 days. Using this method, transgenic plants are obtained within approximately 2 months with a transformation frequency greater than 50%. Polymerase chain reaction (PCR), reverse transcription-PCR (RT-PCR) and β-galactosidase (GUS) histochemical staining analyses confirmed the successful generation of stable transformants. Additionally, the calli from leaves were subcultured and used to obtain new explants; the high transformation efficiency was still maintained in subcultured calli after 6 cycles. This method provides a reference for developing effective transformation protocols for other poplar species.

    Abstract: To investigate the effect of active immunisation with gonadotropin-releasing hormone (GnRH) on the reproductive function in male Sprague Dawley (SD) rats, 24 42-day-old rats were randomly assigned to treatment with GnRH6-MAP, GnRH-OVA, a surgical castration group, and a blank control group. Each rat in the treatment groups was intramuscularly injected at 6, 8, and 10 weeks of age. The serum concentrations of testosterone (T), follicle-stimulating hormone (FSH), luteinising hormone (LH), and anti-GnRH antibodies were determined using enzyme-linked immunosorbent assays. The results showed that active immunisation with recombinant GnRH6-MBP and GnRH-OVA significantly increased the serum levels of anti-GnRH antibodies and reduced the serum concentrations of testosterone compared to the black control. Eight weeks after immunisation, the rats’ testes were surgically removed for morphological evaluation, showing atrophy of the convoluted vasculature, relative emptying of the lumen, and insignificant differentiation of spermatogonial cells, which were increased in weight and volume compared with the blank control group. These findings indicated that active immunisation with GnRH can lead to testicular atrophy and reduce gonadal hormone concentrations, suggesting that GnRH is a highly effective immunogen.

    Abstract: With the rapid growth of energy consumption, enhanced oil recovery (EOR) methods are continually emerging, the most effective and widely used was polymer flooding. However, the shortcomings were gradually exposed. A novel decorated polyacrylamide might be a better alternative than polymer. In this work, the molecular structure and the properties reflecting the viscosity of decorated polyacrylamide, interfacial tension, and emulsification were examined. In order to better understand the interactions between decorated polyacrylamide and oil as well as the displacement mechanism, the displacement experiment were conducted in the etched-glass microscale model. Moreover, the coreflooding comparison experiments between decorated polyacrylamide and polymer were performed to investigate the displacement effect. The statistical analysis showed that the decorated polyacrylamide has excellent characteristics of salt tolerance, viscosity stability, and viscosification like polymer. Besides, the ability to reduce the interfacial tension in order 10−1 and emulsification, which were more similar to surfactant. Therefore, the decorated polyacrylamide was a multifunctional polymer. The displacement process captured by camera illustrated that the decorated polyacrylamide flooded oil mainly by means of ‘pull and drag’, ‘entrainment’, and ‘bridging’, based on the mechanism of viscosifying, emulsifying, and viscoelasticity. The results of the coreflooding experiment indicated that the recovery of decorated polyacrylamide can be improved by approximately 11–16% after water flooding when the concentration was more than 800 mg/L, which was higher than that of conventional polymer flooding. It should be mentioned that a new injection mode of ‘concentration reduction multi-slug’ was first proposed, and it obtained an exciting result of increasing oil production and decreasing water-cut, the effect of conformance control was more significant.

    Abstract: This study aimed to compare the microbial diversity in meatballs with or without blown pack spoilage (BPS) to determine the cause of BPS and to assess the synergistic effect of static magnetic field (SMF) and modified atmosphere packaging (MAP) to reduce the phenomenon of BPS. Results showed that the BPS group with a 2.26-fold larger volume and packaging containing 71.85% CO2 had Klebsiella spp. (46.05%) and Escherichia spp. (39.96%) as the dominant bacteria, which was different from the spoilage group. The results of isolation and identification of strains from the BPS group and their inoculation test confirmed that Klebsiella pneumoniae was the major strain-inducing BPS in meatballs due to its pack-swelling ability. SMF (5 mT) treatment combined with MAP (40%CO2 + 60%N2), which did not influence the sensory quality of meatballs, had a significant synergistic effect on preventing the increase in pack volume. Compared with the control group, this synergistic treatment effectively delayed bacterial growth, drop in pH, and the increase of TBARS. The findings of this study will provide further guidance for meatball manufacturers to adopt effective strategies to reduce the BPS of meatballs.

    Abstract: Temperature and its distribution are crucial for combustion monitoring and control. For this application, digital camera-based pyrometers become increasingly popular, due to its relatively low cost. However, these pyrometers are not universally applicable due to the dependence of calibration. Compared with pyrometers, monitoring cameras exist in all most every combustion chamber. Although these cameras, theologically, have the ability to measure temperature, due to lack of calibration they are only used for visualization to support the decisions of operators. Almost all existing calibration methods are laboratory-based, and hence cannot calibrate a camera in operation. This paper proposes an online calibration method. It uses a pre-calibrated camera as a standard pyrometer to calibrate another camera in operation. The calibration is based on a photo taken by the pyrometry-camera at a position close to the camera in operation. Since the calibration does not affect the use of the camera in operation, it sharply reduces the cost and difficulty of pyrometer calibration. In this paper, a procedure of online calibration is proposed, and the advice about how to set camera parameters is given. Besides, the radio pyrometry is revised for a wider temperature range. The online calibration algorithm is developed based on two assumptions for images of the same flame taken in proximity: (1) there are common regions between the two images taken at close position; (2) there are some constant characteristic temperatures between the two-dimensional temperature distributions of the same flame taken from different angles. And those two assumptions are verified in a real industrial plants. Based on these two verified features, a temperature distribution matching algorithm is developed to calibrate pyrometers online. This method was tested and validated in an industrial-scale municipal solid waste incinerator. The accuracy of the calibrated pyrometer is sufficient for flame monitoring and control.

    Abstract: Harsh parenting can be regarded as a harsh behaviors, feelings, and attitudes toward children in the process of parenting. According to the theory of intergenerational transmission of violence, harsh parenting is an important factor affecting children’s aggressive behavior, but the theory does not clarify the specific action path between harsh parenting and aggressive behavior. In order to reveal the relationship between harsh parenting and the aggressive behavior of juvenile delinquents, 604 male juvenile delinquents (N = 604; Mage = 16.57 years; SD = 0.612 years) were investigated using the Harsh Parenting Questionnaire, the Buss–Perry Aggression Questionnaire, the Multidimensional Peer Victimization Scale, and the Word Sentence Association Paradigm for Hostility in Chinese. Analysis using structural equation modeling procedures showed that (a) all variables were positively associated with each other; (b) the partial indirect effect of harsh parenting on aggressive behaviors was determined through the mediators of peer victimization and hostile attribution bias independently; and (c) the partial indirect effect was determined through the mediators of peer victimization and hostile attribution bias sequentially. The results suggest that harsh parenting can explain the highly aggressive behavior of male juvenile delinquents. Moreover, harsh parenting may also predict the risk of peer victimization and hostile attribution bias, thereby predicting the development of highly aggressive behaviors.

    Abstract: One of the most severe soil-borne pathogens in the world is the root-knot nematode (Meloidogyne incognita). Biological control is gaining more importance as environmental awareness increases. Thus, keeping this in mind, a total of 712 bacterial strains were isolated from 117 rhizosphere soil samples and investigated for potential biological control activity against M. incognita. Strain Sneb518 (Clostridium beijerinckii) was identified as having solid biocontrol activity against M. incognita. Sneb518 demonstrated significant inhibition against M. incognita, with J2 mortality reaching 90.73% at 12 h and with eggs hatching at a rate of 6.00% at 24 h, compared to a hatchability level of 29.07% for the control. Additionally, Sneb518 was excellent for enhancing seed germination. The seeds coated with a fermentation broth containing Sneb518 efficiently boosted the germination rate to 88.49%. The effectiveness and stability of C. beijerinckii Sneb518 against M. incognita were then further evaluated in a greenhouse. According to the pot experiment data, Sneb518 considerably (p 〈 0.05) reduced the number of root galls and egg masses on roots and also significantly (p 〈 0.05) increased tomato plant growth. C. beijerinckii Sneb518-treated tomato seedlings exhibited 50.26% biocontrol effectiveness compared to the control group. Our results demonstrate that C. beijerinckii Sneb518 can be a potential biological control agent against root-knot nematode disease and a biomass enhancer. This research will give new options for the sustainable control of root-knot nematode disease in tomatoes and other host plants.
