    Abstract: Under stress conditions, luteinizing hormone (LH)-mediated ovulation is inhibited, resulting in insufficient oocyte production and excretion during follicular development. When the body is stressed, a large amount of corticosterone (CORT) is generated, which will lead to a disorder of the body’s endocrine system and damage to the body. Our previous work showed that CORT can block follicular development in mice. Since LH acts through binding with the luteinizing hormone receptor (Lhcgr), the present study aimed to investigate whether and how corticosterone (CORT) influences Lhcgr expression in mouse ovarian granulosa cells (GCs). For this purpose, three-week-old ICR female mice were injected intraperitoneally with pregnant mare serum gonadotropin (PMSG). In addition, the treatment group was injected with CORT (1 mg/mouse) at intervals of 8 h and the control group was injected with the same volume of methyl sulfoxide (DMSO). GCs were collected at 24 h, 48 h, and 55 h after PMSG injection. For in vitro experiments, the mouse GCs obtained from healthy follicles were treated with CORT alone, or together with inhibitors against the glucocorticoid receptor (Nr3c1). The results showed that the CORT caused a downregulation of Lhcgr expression in GCs, which was accompanied by impaired cell viability. Moreover, the effect of the CORT was mediated by binding to its receptor (Nr3c1) in GCs. Further investigation revealed that Nr3c1 might regulate the transcription of Lhcgr through inhibiting the expression of Lhcgr transcription factors, including AP1 and Creb. Taken together, our findings suggested a possible mechanism of CORT-induced anovulation involving the inhibition of Lhcgr expression in GCs by the CORT–Nr3c1–AP1/Creb axis.

    Abstract: Oocytes are efficient at reprogramming terminally differentiated cells to a totipotent state. Nuclear transfer techniques can exploit this property to produce cloned animals. However, the overall efficiency is low. The use of umbilical cord mesenchymal stem cells (UC-MSCs) as donor nuclei may increase blastocyst rates, but the exact reasons for this remain unexplored. A single-cell transcriptomic approach was used to map the transcriptome profiles of eight-cell embryos that were in vitro-fertilized and handmade-cloned using umbilical cord mesenchymal stem cells and fibroblasts as nuclear donors. Differences were examined at the chromatin level, the level of differentially expressed genes, the level of histone modifications and the level of DNA methylation. This research provides critical information regarding the use of UC-MSCs as a preferred donor nucleus for nuclear transfer techniques. It also offers unique insights into the mechanism of cellular reprogramming.

    Abstract: The Meishan pig is a traditional Chinese native breed, known for its excellent reproduction performance that is widely used in commercial pig production through two-way or three-way crossbreeding systems. However, the lean meat yield of Meishan crossbred pigs is still very low and cannot meet the market demand. To evaluate the lean meat yield of Meishan crossbred pigs, six wild-type Meishan sows were artificially inseminated by using the MSTN+/− Duroc boar semen in this experiment. Some reproductive performance-related traits of Meishan sows were recorded to ensure that semen from MSTN knockout Duroc boar did not affect offspring production, including total births, live births, sex, and litter weight. In total, 73 piglets were obtained and 63 were alive. Male to female ratio was close to 1: 1. because of factors such as disease, only 43 pigs were utilized, including 28 MSTN mutant pigs (MSTN+/−) and 15 MSTN homozygous pigs (MSTN+/+). We compared the growth performance and carcass performance of these full or half-sib populations and found that there were no differences between MSTN+/− and MSTN+/+ genotypes for live animal measures including average daily gain (ADG), body dimensions, or ultrasonic measurement of fat thickness when pigs were harvested after 120 days of feeding. Conversely, the MSTN+/− pigs had higher dressing percentage and lean meat percentage, lower level of carcass fat, larger longissimus muscle area, less percentage of skin and skeleton, thinner average backfat thickness, and lower intramuscular fat (IMF) content than MSTN+/+ pigs. In conclusion, the production of MSTN+/− mutant progeny from Meishan females resulted in improved carcass composition, providing a feasible solution to improve the lean meat yield of Chinese local fat-type pig breeds.

    Abstract: Metacognitive experience, measured by processing fluency, contributes to divergent thinking performance; however, whether it exhibits varying effects on insight problem-solving remains unknown. Additionally, as individuals’ interpretation of metacognitive experience is influenced by their creative mindset, whether creative mindset plays a role in the relationship between metacognitive experience and insight problem-solving is another issue. In Experiment 1, a Chinese logogriph task was used to investigate insight problem-solving performance. The font style of logogriphs (easy versus difficult) was used to alter the ease of processing. The results showed that individuals had lower performance accuracy for logogriphs presented in difficult font styles, suggesting the negative effect of metacognitive disfluency experience on logogriph solving. In Experiment 2, different creative mindsets (entity versus incremental) were activated in individuals via prime manipulation. Individuals with an incremental creative mindset had a significantly higher performance accuracy and longer reaction time for logogriphs presented in difficult font styles than individuals with an entity creative mindset, suggesting that an incremental creative mindset might counteract the negative effect of metacognitive disfluency experience on logogriphs solving. These findings suggest that metacognitive disfluency experience has a negative effect on insight problem-solving and that a creative mindset moderated this effect.

    Abstract: Urchin-like mullite whiskers synthesized by the vapor–liquid–solid growth method were used to improve the high-temperature performance of porous gelcast SiO2-based ceramic molds. Aluminum was used to facilitate the synthesis of polycrystal urchin-like mullite whiskers which acted as bridges between particles. Scanning electron microscopy (SEM) and X-ray diffraction (XRD) were used to investigate the microstructures and phase compositions of the sintered ceramic samples, respectively. Urchin-like mullite whiskers with diameters of 0.2~1.0 µm and lengths of 1.0~8.0 µm were successfully synthesized in SiO2-based ceramic. When 15 vol% Al was added, the high-temperature strength at 1200 °C was improved from 8.5 to 27.5 MPa, and the creep deformation was decreased to 0.56 mm. Meanwhile, a sintering shrinkage below 0.3% was obtained, and the de-coring rate was accelerated by 67% compared to that of the pure SiO2-based ceramic. This method showed excellent high-temperature strength and high precision, having remarkable potential in the fabrication of hollow turbine blades.

    Abstract: Vegetable-oils-based pressure-sensitive adhesives (PSAs) are being developed as a substitute for petrochemical-based PSAs for application in daily life. However, vegetable-oils-based PSAs face the problems of unsatisfactory binding strengths and easy aging. In this work, the grafting of antioxidants (tea polyphenol palmitates, caffeic acid, ferulic acid, gallic acid, butylated hydroxytoluene, tertiary butylhydroquinone, butylated hydroxyanisole, propyl gallate (PG), tea polyphenols) was introduced into an epoxidized soybean oils (ESO)/di-hydroxylated soybean oils (DSO)-based PSA system to improve the binding strengths and aging-resistant properties. PG was screened out as the most suitable antioxidant in the ESO/DSO-based PSA system. Under optimal conditions (ESO/DSO mass ratio of 9/3, 0.8% PG, 55% rosin ester (RE), 8% phosphoric acid (PA), 50 °C, and 5 min), the peel adhesion, tack, and shear adhesion of the PG-grafted ESO/DSO-based PSA increased to 1.718 N/cm, 4.62 N, and 〉 99 h, respectively, in comparison with the control (0.879 N/cm, 3.59 N, and 13.88 h), while peel adhesion residue reduced to 12.16% in comparison with the control (484.07%). The thermal stability of the ESO/DSO-based PSA was enhanced after PG grafting. PG, RE, PA, and DSO were partially crosslinked in the PSA system, with the rest being free in the network structures. Thus, antioxidant grafting is a feasible method for improving the binding strengths and aging-resistant properties of vegetable-oils-based PSAs.

    Abstract: Herein, a new geopolymer is recognized as a potential alternative cementing material of ordinary Portland cement (OPC), which is used for reducing carbon emissions and efficiently recycling the waste. Therefore this paper mainly studied the alkali-activated coal gangue-slag concrete (ACSC) was prepared by using the coal gangue-slag and Na2SiO3 and NaOH complex activator. The ratio of coal gangue (calcined and uncalcined) coarse aggregate replacing the gravel was 0%, 30%, 50%, 70%, and 100%. The water and salt freeze-thaw resistance, compressive strength, chloride permeation, microstructure, performance mechanism, inner freeze-thaw damage distribution, and mechanics models of ACSC were investigated. Results show that ACSC displayed excellent early age compressive strength, and the compact degree and uniformity of structure were better compared with the ordinary Portland cement (OPC) when the coal gangue replacement rate was less than 50%. The ACSC demonstrated the best chloride penetration resistance under 30% uncalcined coal gangue content, which was less than 27.75% lower than that of using OPC. At the same number cycles, especially in the salt freezing, the calcined coal gangue had lowered advantages of improving resistance freeze-thaw damage resistance. Water and salt accumulative freeze-thaw damage mechanics models of ACSC were established by using the relative dynamic elasticity modulus. The exponential function model was superior to the power function model with better precision and relativity, and the models accurately reflected the freeze-thaw damage effect.

    Abstract: Purpose: To investigate the prognostic value of serum transferrin (TRF) level before intensity-modulated radiation therapy (IMRT) on radio-sensitivity and overall survival (OS) in patients with nasopharyngeal carcinoma (NPC). Methods: From October 2012 to October 2016, a total of 348 patients with NPC in the First Affiliated Hospital of Fujian Medical University were retrospectively analyzed in our study. The concentration of serum TRF was detected by the method of enzyme-linked immunosorbent assay (ELISA). In the whole group, 46 patients received IMRT, and 302 patients received IMRT plus chemotherapy. The radio-sensitive tumor was defined when the local tumor lesions disappeared completely in the nasopharyngeal MRI scan and no tumor residues were found under the electronic nasopharyngoscope one month after the end of radiotherapy. Results: The serum TRF level before IMRT was (1.34–3.89) g/L, with a median of 2.16 g/L and a mean of (2.20 ± 0.42) g/L. In the whole group, 242 cases (69.5%) were radiosensitive, and 106 cases (30.5%) were insensitive. The number of radiosensitive patients in the group of HTRF (transferrin 〉 2.16 g/L) and LTRF (transferrin ≤ 2.16 g/L) before radiotherapy was 129 (74.6%) and 113 (64.6%), respectively. The difference in radio-sensitivity between the two groups was statistically significant (χ2 = 4.103, p = 0.043). Logistic regression analysis showed that the level of TRF before radiotherapy (OR = 1.702; 95% CI 1.044~2.775; p = 0.033) was an independent factor for radio-sensitivity. The log-rank test showed that patients in the LTRF group achieved a significantly worse OS (χ2 = 5.388, p = 0.02) than those in the HTRF group. Cox regression analysis showed that baseline TRF level (HR = 1.706; 95% CI 1.065~2.731; p = 0.026) was an independent prognostic factor for overall survival. Conclusions: The low level of TRF before IMRT is a risk factor for radio-sensitivity and a prognostic factor for poor OS in NPC patients. It may be a promising marker to predict radio-sensitivity and OS in NPC patients who accept IMRT.

    Abstract: Mast cells can recognize foot-and-mouth disease virus-like particles (FMDV-VLPs) via mannose receptors (MRs) to produce differentially expressed cytokines. The regulatory role of chromatin accessibility in this process is unclear. Bone marrow-derived mast cells (BMMCs) were cultured, and an assay of transposase-accessible chromatin sequencing (ATAC-seq) was applied to demonstrate the regulation of chromatin accessibility in response to the BMMCs’ recognition of FMDV-VLPs. A pathway enrichment analysis showed that peaks associated with the nuclear factor-κB (NF-κB), mitogen-activated protein kinase (MAPK), phosphatidylinositol 3 kinase-protein kinase B (PI3K-Akt), and other signaling pathways, especially the NF-κB pathway, were involved in the BMMCs’ recognition of VLPs. Moreover, transcription factors including SP1, NRF1, AP1, GATA3, microphthalmia-associated transcription factor (MITF), and NF-κB-p65 may bind to the motifs with altered chromatin accessibility to regulate gene transcription. Furthermore, the expression of NF-κB, interleukin (IL)-9, tumor necrosis factor (TNF)-α, and interferon (IFN)-γ in the BMMCs of the VLP group increased compared with that of the BMMCs in the control group, whereas the expression of IL-10 did not differ significantly between groups. After inhibiting the MRs, the expression of NF-κB, IL-9, TNF-α, and IFN-γ decreased significantly, whereas the expression of IL-10 increased. The expression of MAPK and IL-6 showed no significant change after MR inhibition. This study demonstrated that MRs expressed on BMMCs can affect the NF-κB pathway by changing chromatin accessibility to regulate the transcription of specific cytokines, ultimately leading to the differential expression of cytokines. These data provide a theoretical basis and new ideas for the development of a novel vaccine for FMD.
