Am 26. August 1768 stach Cook mit dem für seine Zwecke umgebauten ehemaligen Kohlenfrachter 'Endeavour' in See, um für die Königlich Geographische Gesellschaft astronomische Beobachtungen auf Tahiti vorzunehmen und von dort zu dem noch unbekannten 'Südkontinent' (Terra australis) vorzustoßen. Drei Monate lang beobachtete er auf Tahiti das Leben der Eingeborenen, machte sich außerdem mit der Pflanzen- und Tierwelt vertraut, kartographierte die umliegenden Inseln, die er zu Ehren seiner Auftraggeber 'Gesellschaftsinseln' nannte, und stellte fest, dass Neuseeland aus zwei Hauptinseln bestand, es somit das 'Südland' gar nicht gab. Seine Weiterreise führte ihn von Neuseeland an die fast unbekannte Ostküste Australiens und durch die Tores-Straße nach Batavia (Djakarta). Über das Kap der Guten Hoffnung kehrte er 1771 nach England zurück. 1772-1775 überquerte er zweimal den südlichen Polarkreis, erreichte die Osterinseln, die Neuen Hebriden, Neukaledonien, Kap Hoorn und Südgeorgien. 1776 brach er zu seiner dritten Expedition auf, um die nördliche Durchfahrt zwischen dem Pazifik und dem Atlantik zu erkunden. Dabei entdeckte er eine Inselgruppe, die er zu Ehren des Earl of Sandwich 'Sandwich-Inseln' nannte, das heutige Hawaii. Nach Erreichen der Westküste Nordamerikas segelte er durch die Beringstraße ins Nordpolarmeer, wurde wegen großer Massen an Treibeis zur Umkehr gezwungen und erreichte am 26. November 1778 Hawaii. Die Anfangs freundliche Gesinnung der Eingeborenen schlug infolge eines Verstoßes seiner Mannschaft gegen das Tabugesetz plötzlich um, was schließlich zur Ermordung Cooks führte. Kein Seefahrer vor James Cook unternahm so ausgedehnte Reisen, verbrachte so lange Zeiträume ununterbrochen auf See und kehrte mit so umfangreichen neuen Kenntnissen weiter Teile der Erde zurück.
