    Abstract: Klebsiella pneumoniae carbapenemase (KPC)-producing bacteria is a group of highly dangerous antibiotic resistant Gram-negative Enterobacteriaceae. They cause infections associated with significant morbidity and mortality. Therefore, the rapid detection of KPC-producing bacteria plays a key role in clinical microbiology. Matrix assisted laser desorption/ionization time-of- flight (MALDI-TOF) is a rapidly evolving technology that finds application in various clinical, scientific, and industrial disciplines. In the present study, we demonstrated three different procedures of carbapenemase-producing K. pneumoniae (KPC) detection. The most basic model of MALDI-TOF instrument MS Microflex LT was used, operating in the linear ion-positive mode, commonly used in modern clinical laboratories. The first procedure was based on indirect monitoring of carbapenemase production with direct detection of hydrolyzed carbapenem antibiotic degradation products in the mass spectrum. The second procedure was based on direct detection of blaKPC accompanying peak with an 11,109 Da in the mass spectrum of carbapenemase-producing K. pneumoniae (KPC), which represents the cleaved protein (pKpQIL_p019) expressed by pKpQIL plasmid. In addition, several unique peaks were detected in the carbapenemase-producing K. pneumoniae (KPC) mass spectrum. The third procedure was the identification of carbapenemase-producing K. pneumoniae (KPC) based on the protein fingerprint using local database created from the whole mass spectra. By comparing detection procedures, we determined that the third procedure was very fast and relatively easy. However, it requires previous verification of carbapenemase-producing K. pneumoniae (KPC) using other methods as genetic blaKPC identification, detection of carbapenem degradation products, and accompanying peak with 11,109 Da, which represents cleaved pKpQIL_p019 protein expressed by pKpQIL plasmid. Detection of carbapenemase-producing K. pneumoniae using MALDI-TOF provides fast and accurate results that may help to reduce morbidity and mortality in hospital setting when applied in diagnostic situations.

    Abstract: The main objective of this study is to evaluate the effect of selected essential oils thyme chemotype linalool (Thymus zygis L.), thyme chemotype tymol (Thymus vulgaris L.), eucalyptus (Eucalyptus globulus Labill.), lavender (Lavandula angustifolia Mill.), mint (Mentha piperita L.), almond (Prunbus dulcis Mill.), cinnamon bark (Cinnamomum zeylanicum Nees), litsea (Litsea cubeba Lour. Pers), lemongrass (Cympogon citrati L. Stapf), and ginger (Zingiber officinalis Rosc.) in the vapor phase on growth, sporulation, and mycotoxins production of two Aspergillus strains (Aspergillus parasiticus CGC34 and Aspergillus ochraceus CGC87), important postharvest pathogens of green and roasted coffee beans. Moreover, the effect of the essential oils (EOs) on the sensory profile of the coffee samples treated with EOs was evaluated. The major components of tested EOs were determined by gas chromatography and mass spectrometry (GC–MS) and gas chromatography with flame ionization detector (GC-FID). The results showed that almond, cinnamon bark, lemongrass, and litsea EOs are able to significantly inhibit the growth, sporulation, and mycotoxins production by toxigenic fungi. Sensory evaluation of coffee beans treated with EOs before and after roasting showed that some EOs (except lemongrass and litsea) do not adversely affect the taste and aroma of coffee beverages. Thus, application of the vapors of almond and cinnamon EOs appears to be an effective way that could serve to protect coffee during its transport and storage from toxigenic fungi.

    Abstract: The aim of this study was detection of gene leptin in muscles, liver and kidneys from pigs of breed Large White. Using Real time PRC method, we determined the Ct values of leptim gene in muscle, liver, kidney. The body weight of pigs ranged from 100 kg to 103 kg. The average body weight was 101.6 kg. The thickness of backfat ranged from 10 to 20 mm, average backfat thickness was 16 mm. The minimal Ct value of leptin gene in liver was 24.05 and the maximal value was 25.79. Average Ct value of leptin gene was 24.84. The minimal Ct value of leptin gene in muscle tissue was 25.83 and the maximal value was 27.05. Average Ct value of leptin gene was 26.41. The Ct value of leptin gene in liver ranged from 24.05 to 25.79. Average Ct value of leptin gene was 24.84. Leptin gene is expressed by porcine preadipocytes and leptin gene expression is highly dependent on dexamethasone induced preadipocyte differentiation. Hormonally driven preadipocyte recruitment and subsequent fat cell size may regulate leptin gene expression in the pig.

    Abstract: The present study evaluated the effect of probiotic in combination with bee pollen and propolis in diet of broilers on chemical and sensory characteristics of chicken breast meat. A total of 180 one-d-old chicks of mixed sex were randomly assigned to three dietary groups as follows: 1. control group (C); 2. basal diet supplemented with 400 mg bee pollen (ethanol extract) per 1 kg of feed and 3.3 g probiotic (Lactobacillus fermentum) per day in water (E1); 3. basal diet supplemented with 400 mg propolis (ethanol extract) per 1 kg of feed and 3.3 g probiotic (Lactobacillus fermentum) per day in water (E2). Chicken meat was analyzed for dry matter, crude protein, fat, ash, cholesterol, and energy value. Diet did not affect the chemical characteristics of chicken meat, except for supplementation with bee pollen and probiotic which resulted in increased fat content. As for sensory characteristics, dietary groups differ from each other in terms of aroma and overall acceptability. In addition, there was significant difference between the E1 group and groups C and E2 in terms of taste and tenderness. Significant differences were also detected between group E2 and groups C and E1 group in terms of juiciness. In conclusion, propolis + probiotic-supplemented group manifested the best results in terms of sensory characteristics. Propolis extract can be thus recommended as a potential supplement providing rich nutrients and biological active substances in chicken diet that is (together with probiotic) capable of improving the sensory quality of chicken breast meat.

    Abstract: The chemical composition and content of DBP (di-n-butyl phthalate) and DEHP (di-2-ethylhexyl phthalate) in pork shoulder before and after heat treatment in the package by the sous vide method was analyzed. The meat was analyzed raw and after heat treatment at 50 °C and 60 °C. The heat treatment time in the sous vide water bath was 4 hours, 4 hours + 1 hour after 24 hours, 8 hours and 8 hours and 1 hour after 24 hours. The fat content in meat treated at 50 °C was 6.04 in raw meat and after heat treatment after 4 hours. 7.51 and after 8 h. 6.81 g.100g-1, in the shoulder after heat treatment at 60 °C was after 4 hours 6.24 and after 8 h. 6.76 g.100g-1. The content of fatty acids during the sous vide treatment did not significantly change with exception of vaccenic acid, the content of which was statistically significantly reduced at temperatures of 50 °C also at 60 °C. The DBP content in raw shoulder at 50 °C during sous vide heat treatment it increased to 1.91 μg.g-1. The DEHP content during the heat treatment it increased to 23.95 μg.g–1 in the treatment of 4+1 hours.  The DBP content in raw shoulder after heat treatment at 60 °C increased to 1.84 μg.g–1 during treatment for 4+1 hours. The DEHP content decreased to 8.72 in the treatment of 4+1 hours and to 4.021 μg.g–1. Based on our results, we can conclude that at both monitored temperatures of sous vide method, the DBP content increased and the DEHP content decreased. The DBP content in raw shoulder at 50°C heat treatment increased to 1.91 μg.g-1 at 4+1 hour heat treatment and to 3.02 at 8+1 hour. The DEHP content increased to 23.95 μg.g-1 in the treatment of 4+1 hours. The content of DBP and DEHP in the packaging material before use was 29.08 μg.g-1, it gradually decreased with the length of the heat treatment, to 15.09 μg.g-1 in the treatment of 8+1 hours. The DEHP content in the unused package decreased to1.27 μg.g-1 at heat treatment of 8+1 hours. At the heat treatment at of 60°C in the packaging material gradually decreased to 3.18 μg.g-1. The DEHP content decreased to 2.54 μg.g-1.

    Abstract: This study aimed to evaluate the antifungal activity and the potential synergistic activity of ten plant extracts (EXs) and ten essential oils (EOs) obtained from the same plant species, namely Agrimonia eupatoria L., Arctium lappa L., Arnica montana L., Bidens cernua L., Corylus avellana L., Echinacea purpurea L. (Moench), Menta piperita L., Trifolium pratense L., Syzygium aromaticum L., and Angelica sinensis (Oliv.) Diels were used. Three species of the genus Candida, namely C. albicans, C. glabrata, and C. tropicalis, isolated from polluted estuarine water, were used in this study. The chemical composition of all EOs was evaluated by gas chromatography coupled with mass spectrometry (GC–MS) and gas chromatography with flame ionization detector (GC–FID) analysis. Firstly, the susceptibility of Candida spp. strains to EXs and EOs was screened by the agar disc diffusion method. Then the minimum inhibitory concentration (MIC) of the tested EXs and EOs was determined in a concentration range from 256 to 0.125 µL/mL. Following that, the effective EXs and EOs were divided into two groups (strong effect and weak effect) and tested in combination (strong + weak) at various concentrations depending on the tested strain to determine their potential synergistic effect using the chessboard pattern. The most sensitive strain to treatments with EXs and EOs tested alone and in mixtures was C. albicans. The most potent combinations with other EXs/EOs were observed with Syzygium aromaticum L., for both plant extract and essential oil, respectively. In this study the EOs and EXs tested in combination were much more effective than when acting on the growth of Candida spp. separately. So, the synergistic effect of the combinations of the EXs and EOs can lead to a reduction in the doses of commercial antifungals and can provide an effective way to reduce the infections caused by yeasts of the genus Candida.

    Abstract: Amygdalin is non-toxic, but when placed with certain amino acids in the small intestine, it leads to the production of toxic hydrocyanic acid. In our study, we observed the effect of amygdalin on the activity of Superoxide Dismutase (SOD) and Glutathione Peroxidase (GPx) in the kidney after a short administration. The experiment was performed on male California m eat rabbits of the P91 rabbit line (n = 20). Amygdalin was applied intramuscularly in the experimental group E1 and E2 daily for 14 days. Animals from experimental groups E3 and E4 were fed crushed apricot seeds. Apricot seeds were mixed into the feed mixture. The control group of animals was fed with a standard commercial feed mixture. Significant differences (p 〈 0.05) were observed in GPx activity in the control group and experimental considerations E1 and E2, where amygdalin was administered i.m. A significant difference was also observed among the control group and the E4 group, where amygdalin was administered to the animals per os. We also observed differences among SOD activity. Significant differences (p 〈 0.05) were found out in the control group and the experimental group E2 where amygdalin was administered i.m. and between the control group and the E4 group, where amygdalin was administered to the animals per os. We found significant differences between experimental groups E1:E2 and between experimental group E1 and experimental group E4. We did not find significant differences among the other groups (p 〉 0.05). Many aspects of amygdalin administration have not yet been explored, requiring other research.

    Abstract: Antibiotic resistance and identification of strains in Enterobacteriaceae genera isolated from milk, milk products and rectal swabs of chicken was examined in this experiment. After samples collection cultivation and identification of bacterial strain was done. MALDI TOF MS Biotyper for identification of Enterobacteriaceae strains was used. For susceptibility testing disc diffusion methodology was used according by EUCAST. Results showed high level of ampicillin resistance in isolates from milk and milk samples. The highest streptomycin resistance was detected in isolates from rectal swabs of chicken. After identification, we determined that S. enterica ser. Typhimurium, which was isolated from rectal swabs of chicken showed the most multi-resistance from all identificated strains of Enterobacteriaceae. The most isolates bacterial strain was E. coli, which showed resistance against four antibiotics from rectal swabs of chicken. Also our results showed that the higher resistance level is in rectal swabs of chicken like in milk samples.
