Abstract:
Ni,Fe‐haltige Kohlenmonoxid‐Dehydrogenasen (CODHs) katalysieren die reversible Reduktion von CO 2 zu CO. Mehrere anaerobe Mikroorganismen enthalten mehrere CODHs in ihrem Genom, von denen einigen, obwohl sie als CODHs annotiert sind, ein Cystein des kanonischen Bindungsmotivs für den Ni,Fe‐Cluster im aktiven Zentrum fehlt. Hier berichten wir über die Struktur und Reaktivität eines solchen abweichenden Enzyms, das als CooS‐V Ch bezeichnet wird. Seine Struktur weist das typische CODH‐Gerüst auf, enthält aber einen Eisen‐Schwefel‐Oxo‐Hybrid‐Cluster. Obwohl es eng mit den echten CODHs verwandt ist, katalysiert CooS‐V Ch weder die CO‐Oxidation noch die CO 2 ‐Reduktion. Das aktive Zentrum der CooS‐V Ch durchläuft eine redoxabhängige Umstrukturierung zwischen einem reduzierten [4Fe‐3S]‐Cluster und einem oxidierten [4Fe‐2S‐S*‐2O‐2(H 2 O)]‐Cluster. Hydroxylamin, ein langsam reagierendes Substrat von CooS‐V Ch , oxidiert den Hybrid‐Cluster in zwei strukturell unterscheidbaren Schritten. Insgesamt reichen geringfügige Änderungen in CODHs aus, um einen Fe/S/O‐Cluster anstelle des Ni,Fe‐Heterokuban‐Clusters der CODHs unterzubringen.
