    Abstract: This study aimed to assess the post-effort transcriptional changes of selected genes encoding receptors for chemokines and interleukins in young, physically active men to better understand the immunomodulatory effect of physical activity. The participants, aged 16–21 years, performed physical exercise tasks of either a maximal multistage 20 m shuttle-run test (beep test) or a repeated speed ability test. The expression of selected genes encoding receptors for chemokines and interleukins in nucleated peripheral blood cells was determined using RT-qPCR. Aerobic endurance activity was a positive stimulant that induced increased expression of CCR1 and CCR2 genes following lactate recovery, while the maximum expression of CCR5 was found immediately post-effort. The increase in the expression of inflammation-related genes encoding chemokine receptors triggered by aerobic effort strengthens the theory that physical effort induces sterile inflammation. Different profiles of studied chemokine receptor gene expression induced by short-term anaerobic effort suggest that not all types of physical effort activate the same immunological pathways. A significant increase in IL17RA gene expression after the beep test confirmed the hypothesis that cells expressing this receptor, including Th17 lymphocyte subsets, can be involved in the creation of an immune response after endurance efforts.

    Abstract: The participation of T cell subsets in the modulation of immunity in athletes triggered by maximal effort was investigated. In total, 80 physically active young men (range 16–20 years) were divided into 5 age groups: 16, 17, 18, 19, and 20 years old. They performed efficiency tests on mechanical treadmills until exhaustion. White blood cell (WBC) and lymphocyte (LYM) counts were determined, and the type 1 (Th1), type 2 (Th2) helper T cells, T helper 17 (Th17), and T regulatory (Treg) cell distribution and plasma levels of selected cytokines were analyzed. An increase in WBC and LYM counts after the test and in Th1 and Treg cells after the test and in recovery was observed. There were no changes in Th2 cells. An increase in interleukins (IL): IL-2 and IL-8 was observed. The IL-6 level was altered in all studied groups. IL-17A and interferon gamma (IFN-γ) levels were increased in all studied groups. The mechanism of differential T cell subset activation may be related to athletes’ age. The novel findings of this study are the involvement of Th17 cells in post-effort immune responses and the participation of IL-6 in post-effort and the long-term biological effect of endurance effort.

    Abstract: W e compared the effects of 16-week-training on rest metabolic rate, aerobic power, and body fat, and the post-exercise effects upon rest oxygen uptake and respiratory exchange ratio in overweight middle-aged females. Twenty nine overweight women (BMI 29.9 ± 1.2 kg*m -² ) participated in training (3 days a week). The subjects were divided onto groups of aerobic (AT) and strength (ST) training. The results showed that the total body mass decrease and VO 2 max increase did not differ in both groups. Decrease in waist circumference after 16 weeks was higher in the ST group. In the ST group fat-free mass increased during the first 8 weeks. Rest metabolic rate was increased significantly at 16th week compared to initial value in ST group only. Significant increase in post-exercise resting VO 2 and respiratory exchange ratio at 12 and 36 h was observed after the strength training session only. Increase in rest metabolic rate and post-exercise rest energy expenditure occurred after strength training but not after aerobic training despite the similar increase in aerobic power. The effect of 8-16 weeks of strength training on body mass decrease was higher in comparison to aerobic training.

    Abstract: Inflammation-induced processes commence with the activation of signalling pathways at the cellular level, which mobilize inflammatory cells and stimulate the secretion of chemokines, cytokines, and damage-associated molecular pattern molecules (DAMPs). Physical effort stimulates inflammation, contributing to muscle repair and regeneration. We have examined the impact of different protocols of progressive-effort tests on T-cell DAMP levels, extracellular cleavage products (fibronectin and hyaluronan), and Th-cell-related cytokine levels among soccer players. Thirty male soccer players with a median age of 17 (16–22) years performed different defined protocols for progressive exercise until exhaustion: (1) YO-YO intermittent recovery test level 1 (YYRL1, n = 10); (2) maximal multistage 20 m shuttle run (Beep, n = 10); and mechanical treadmill (MT, n = 10); and (3) shuttle-run test (n = 10). Blood samples were taken three times as follows: at baseline, post effort, and in recovery. Significantly higher post-effort concentrations of IL-4, IL-6, IL-10, and IFN-γ were observed in the Beep group, IL-4 in the YYRL1 group, and IL-6 and IFN-γ in the MT group as compared with the baseline values. Recovery values were significantly higher for concentrations of IL-4, IL-10, and IFN-γ in the YYRL1 group, only for IFN-γ in the Beep group, and for IL-6, IL-10, and INF-γ in the MT group as compared with the baseline values. Post-effort concentrations of DEFβ2, Hsp27, Fn, and UA in the Beep group and Hsp27 and HA in the YYRL1 group were significantly higher as compared with the baseline values. It seems the performed efficiency test protocols caused a short-term imbalance in Th1/Th2 cytokine levels without giving common molecular patterns. The rapidity of these changes was apparently related to specific physical movements and the type of running surface.

    Abstract: Molecular mechanisms of biological adaptation to training in professional soccer players are unclear. The aim of this study was to assess the impact of progressive physical effort on peripheral T-cells and their molecular response. Methods Thirteen soccer players form Pogo Szczecin S.A., a top league soccer club, (median age 21, range 18– 31, years old) performed progressive efficiency tests on a mechanical treadmill until exhaustion at the start (period 1) and the end (period 2) of a competition round. Venous blood T-lymphocyte subsets, selected hallmarks of cell death and plasma cytokine levels were determined by flow cytometry three times: pre-exercise, post-exercise, and in recovery. Results Although significant changes in T, Tc and Tc-naïve cell percentages were found in both periods, Th-naïve cell percentages were altered only in period 1. Post-exercise IL-10 plasma levels were higher than pre-exercise, while an increase in TNF-α levels was noticed in recovery from both periods. An increase in recovery IL-12p70 levels was observed in the second period. Increases in the percentage of T-cells with disrupted mitochondrial membrane potentials, elevated levels of phosphorylated H2AX histones and increases in early apoptotic T-cells were also observed. Conclusions The immune system in soccer players creates space for naïve CD3 + CD8 + cells by inducing mechanisms of cell death. It seems that the cumulative effect of physical activity during a competition round induced an adaptive mechanism, since the cell death process was induced faster during period 2.

    Abstract: Immunological alterations may led to the reduction in capacity and endurance levels in elite athletes by e.g. increased susceptibility to infections. There is a need to explain the impact of intensive physical effort on the CD4 + memory T cell subsets. Methods Fourteen participants median aged 19 years old (range 17–21 years) were recruited form Pogoń Szczecin S.A., soccer club. They performed progressive efficiency test on mechanical treadmill until exhaustion twice: during preparatory phases to spring and autumn competition rounds. We examined the influence of exhaustive effort on the selected CD45 + , especially CD4 + memory T cell subsets and inflammation markers determined before, just after the test and during recovery time. Results Significant changes in total CD45 + cells and decrease in T lymphocytes percentage after the run was observed. Significant fluctuations in T cells’ distribution were related not only to the changes in Th or Tc subsets but also to increase in naïve T cell percentage during recovery. Increase in TNF-α and IL-8 post-exercise, IL-6 and IL-10 plasma levels in recovery was also found. Conclusions The novel finding of our study is that the run performed on mechanical treadmill caused a significant release of CD4 + T naïve cells into circulation. Post-exercise increase in circulating NK cells is related with fast biological response to maximal effort. However, at the same time an alternative mechanism enhancing inflammation is involved.

J Strength Cond Res 35(8): 2279–2286, 2021—Mechanisms responsible for increasing athletes' physical capacity and induction of exercise-induced immunosuppression processes are not fully understood. The aim of the study was to monitor changes in percentages of lymphocyte subsets: T, Th, Tc, B, and NK cells in capillary blood of junior soccer players. Ten subjects median aged 18 years (range 17–19 years) were recruited form young soccer players. Capillary blood was collected 24 hours after each soccer match during the 8 weeks of the final phase of Central Junior League competition, and white blood cell (WBC) phenotyping was performed to determine the percentages of B lymphocytes, NK cells, and T-lymphocyte subsets. Cumulative match-time (a sum of time spend playing the game by each athlete during the observation period) was also calculated. Significant changes in the percentage of total lymphocytes ( p = 0.00005) and T cells ( p = 0.00006) were observed. The slight increases in lymphocytes' and Th cells' median percentages correlated with increasing cumulative match-time of studied subjects, although the correlation was not strong ( R = 0.24; p = 0.0205 and R = 0.30; p = 0.0035, for lymphocytes and Th cells, respectively). It seems that the exercise bouts are among considerable factors influencing the changes in WBC subsets, especially in CD3 + cells, among young soccer players. Regarding the number of games played and training loads, they are more susceptible to immunosuppression and subsequent infections and thus should be monitored regarding WBC phenotype assessment.

    Abstract: Purpose. Hypercholesterolaemia is a highly prevalent condition that has major health- and cost-related implications for the society. Aerobic-type exercise improves lipoprotein-lipid profiles, cardiorespiratory fitness and body composition in healthy young women. Thus, the aim of the study was to assess the impact of 9 weeks of low-high aerobic-type exercise on the lipid profile among young women. Methods. On the basis of the lipid profile, 64 women (median age, 21.8 years; range, 19.0-24.7 years) were divided into two groups: with low (LRH) and intermediate (IRH) risk of developing hypercholesterolaemia. The participants completed a 9-week-long low-high aerobic exercise programme. Before and after the training programme, we determined the lipid profile: triglycerides (TG), total cholesterol (TC), lipoprotein cholesterol: HDL-C and LDL-C, and glucose levels. Selected cardiorespiratory fitness variables and body composition were also determined. Results. It was found that aerobic-type fitness exercise in the IRH group caused statistically significant decreases in TC and TG levels in comparison with baseline values. Significant increase in maximum oxygen uptake and decrease in HDL-C in the LRH group were also observed. Conclusions. Aerobic fitness exercises, a combination of two alternating styles, could influence the blood lipid profile by decreasing plasma TC and TG levels. In non-athlete women, physical activity may be a good tool to prevent cardiovascular diseases.
