Abstract:
Mithilfe eines Pikosekunden‐IR‐Lasers (PIRL) können Proteine in weniger als einer Millisekunde aus intakten Geweben in die Gasphase katapultiert werden. Die ultraschnelle Desorption erfolgt durch eine impulsive Anregung intramolekularer Vibrationszustände (DIVE) der Wassermoleküle, die die Proteine innerhalb des Gewebes umgeben. PIRL‐DIVE extrahiert mit hohen Ausbeuten eine große Vielzahl unterschiedlicher Proteine, von wenigen kDa bis zu mehreren MDa. Die chemische Zusammensetzung der Proteine wird durch die Energie des PIRL nicht verändert. Der Nachweis von Enzymaktivitäten in den PIRL‐Extrakten weist darauf hin, dass die Konformation der Enzyme erhalten bleibt.
