Abstract: The genus Theopropus Saussure, 1898 is distributed with five species in SE Asia, three of which were recorded in South China: T. elegans (Westwood), T. cattulus (Westwood) and T. sinecus Yang. After examining numerous specimens of Theopropus , we attempt to resolve some taxonomic confusion about Theopropus occurring in China. Those Chinese specimens that were inaccurately identified as T. cattulus represent a new subspecies of T. sinecus Yang: T. s. qiongae Wu & amp; Liu, ssp. nov. . A new species, T. xishiae Wu & amp; Liu, sp. nov. , is described from the rainforests of the southern slopes of the Himalayas. The records of T. elegans in China are also clarified. Biological characteristics of the species and subspecies, necessary illustrations, and ecological images are provided. The distribution of the known Chinese Theopropus species is discussed and mapped.

Abstract: During an investigation of Xylariaceae from 2019 to 2020, isolates representing eight Nemania (Xylariacese) species were collected from Yunnan, Guizhou and Hainan Provinces in China. Morphological and multi-gene phylogenetic analyses, based on combined ITS, α-actin, rpb2 and β-tubulin sequences, confirmed that six of them are new to science, viz. Nemania camelliae , N. changningensis , N. cyclobalanopsina , N. feicuiensis , N. lishuicola and N. rubi ; one is a new record ( N. caries ) for China and one is a known species ( N. diffusa ). Morphological descriptions and illustrations of all species are detailed. In addition, the characteristics of Nemania are summarised and prevailing contradictions in generic concepts are discussed.

Abstract: The genus Parachironomus has a cosmopolitan distribution including 85 valid described species worldwide. Species records and studies of the genus in the Tibetan Plateau are scarce. In this study, the genus Parachironomus from China is revised and two new species, Parachironomus wangi Liu & Lin, sp. nov. and Parachironomus nankaiensis Liu & Lin, sp. nov. , are described based on adult morphology and molecular data. Paracladopelma demissum Yan, Wang & Bu is placed in the genus Parachironomus as a new combination. A neighbor-joining tree was reconstructed based on all known Parachironomus COI DNA barcodes. A key to adult males of the genus Parachironomus from China is also provided.

Abstract: Shiraiaceae is an important family in Pleosporales (Dothideomycetes), which includes medical fungi and plant pathogens. Two hypocrellin-producing taxa, Shiraia bambusicola and a novel genus Rubroshiraia gen. nov. , typified by Rubroshiraia bambusae are treated in this article. Maximum likelihood analysis, generated via RAxML (GTR+G model), using a combined SSU, LSU, TEF1 and RPB2 sequence dataset, shows that Rubroshiraia is close to Shiraia and belongs to the family Shiraiaceae. Descriptions, illustrations and a taxonomic key are provided for the genera in Shiraiaceae. Rubroshiraia morphologically differs from Shiraia in having small and dark ascostromata and filiform ascospores. Production of the ascostromatal metabolites, hypocrellin A and B, were examined by HPLC and spectrophotometer. The content of hypocrellin A and B of specimen HKAS 102255 ( R. bambusae ) is twice that produced by HKAS 102253 ( S. bambusicola ). To clarify the relationship between R. bambusae and Hypocrella bambusae , type material of the latter was examined and provided the illustration.

Abstract: Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli . Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals 〈 1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.
