    Abstract: Razvoj molekularne biologije i molekularne genetike posebno tehnologije rekombinantne DNK uz istovremeno usavrsavanje tehnologija koje omogucavaju manipulacije u bezcelijskom sistemu kao sto su kultura celija i tkiva rezultirao je u formiranju oblasti biotehnologije sa specificnim osobinama u odnosu na klasicne definicije. Integracijom najpropulzivnijih naucnih disciplina kao sto su biologija, biohemija, molekularna biologija, genetika hemijski inzenjering i informatika otvorene su neogranicene mogucnosti i izazovi u sprecavanju bolesti humane populacije, biljaka i zivotinja kontroli insekata bez pesticida, povecanju produktivnosti stoke, povecanju proizvodnje i poboljsanju kvaliteta hrane i redukciji zivotne sredine. Osnovne komponente moderne biotehnologije, prema Presley-u i Lantin (2000) su: (i) genomika - molekularna karakterizacija svih gena i produkata gena organizma; (ii) bioinformatika - organizovanje podataka dobijenih iz analize genoma u prihvatljivu formu; (iii) transformacija - unosenje gena koji kontrolise osobinu od interesa u genom zeljenog organizma (mikroorganizmi biljke, animalni sistemi) i (iv) dijagnostika - brza i pouzdanija identifikacija prisustva ili odsustva gena u genomu organizma od interesa (identifikacija patogena, prenatalna dijagnostika, molekularno oplemenjivanje biljaka koriscenjem molekularnih markera i sl). Svojstva organizma su odredjena njegovim genetickim materijalom, odnosno molekulom dezoksiribonukleinske kiseline (DNK). Watson i Crick (1953) su prvi objasnili strukturu molekula DNK kao dvolancane zavojnice. Visi organizmi sadrze set linearnih DNK molekula - hromozoma i kompletan set hromozoma organizma je genom. Svaki genom je podeljen u serije funkcionalnih jedinica odnosno gena. Svojstva organizma zavise od gena ali njihovo ispoljavanje zavisi i od mnogih drugih faktora, ukljucujuci da li se gen odgovoran za tu osobinu eksprimira, specificnih celija u kojima se eksprimira a posebno kako gen i njegov produkt intereaguju sa spoljasnjom sredinom. Poseban aspekt u okviru koriscenja biotehnologije se javlja kao interakcija stranog gena sa genomom organizma u koji je integrisan. Primena tehnologije rekombinatne DNK ima nekoliko prednosti u odnosu na tradicionalno oplemenjivanje. Geneticke modifikacije biljaka kao i klasicno oplemenjivanje imaju isti cilj, dobijanje biljaka sa poboljsanim svojstvima. Klasicnim metodama oplemenjivanja prenosi se kompletan set gena iz roditeljskih linija u novo potomstvo. Time se ne prenosi samo jedan ili dva zeljena gena vec stotine drugih gena, neki pozeljni, a neki ne. Takodje, tradicionalne metode oplemenjivanja se zasnivaju na prenosu genetickog materijala izmedju jedinki iste ili veoma bliske vrste. Primenom biotehnologije moguce je preneti jedan ili vise pozeljnih gena iz bilo koje evolucione kategorije u istu ili drugu kategoriju organizma i na taj nacin stvoriti geneticki modifikovane organizme (GMO) sa zeljenim osobinama. U radu ce biti dat pregled stanja u oblasti primene biotehnologije u svetu i kod nas.

    Abstract: Modern agriculture, breeding procedures, as well as competition among breeding institutions contribute to further reduction of already narrowed diversity of soybean commercial varieties. The objective of the study was to characterize eighteen soybean cultivars from three different breeding programs for agro-morphological traits and to reveal genetic diversity using molecular markers. Morphological description was performed with 13 qualitative and 9 quantitative traits. The genetic relationships were estimated using 21 RAPD markers. PIC was calculated for RAPD data, while the diversity of qualitative traits was described by Shannon genetic diversity index. Cluster analysis based on qualitative morphological characters showed clear separation of genotypes on the basis of their plant growth type. PC analysis performed for quantitative traits divided genotypes according to their maturity group. Grouping pattern based on molecular marker data was in agreement with pedigree of cultivars. A great similarity was found, primarily between the varieties under the same institution, and then among all examined varieties. Comparison of three methods in the assessment of diversity indicated that morphological markers might provide useful information in breeding process and allow classification by pedigree to some extent, but RAPD markers were found to be superior in assessing differences among genetically very similar genotypes.

    Abstract: The paper presents the results of the analysis of genetic variability of eight populations of silver fir (Abies alba Mill.) in Serbia obtained using SSR markers. The genomic DNA was isolated from tissue of needles of all eight populations. Due to the costly and lengthy process a small number of the SSR markers for Abies alba have been developed, so in this study were used the microsatellite markers of related species. The obtained results indicate a low level of the genetic variability between natural populations of silver fir. The total number of alleles detected with nine SSR markers in eight studied populations of silver fir is 28. The range of alleles varies from two for NFF15 to six for SF78 with an average of 3.1 alleles per locus. The mean value of genetic similarity between populations is 0.92. The smallest genetic similarity between pairs of populations is 0.82 (Dubocica Bare and Stara Planina; Dubocica Bare and Tara) and the greatest genetic similarity is 1 (Zlatar and Stara Planina, Zlatar and Tara, Stara Planina and Tara). A basic insight into the level of genetic diversity of natural populations of silver fir in Serbia, which are located in a relatively small area, has been given using a set of SSR markers. The obtained results can be used in the future strategy for the management and regeneration of silver fir forests.

    Abstract: Panels of diverse materials have proven to be very useful in evaluation of the organization of genetic diversity available for breeding at phenotypic and genotypic levels. In this study, a panel consisting of several groups of maize inbreds was tested using microstelite markers. The aim of this study was to test genetic diversity and define population structure of twenty-four genotypes differing in kernel type (dent, flint, popcorn, sweet maize) and kernel color (white, yellow, orange) with 21 SSRs. Genetic diversity parameters such as number of alleles, gene diversity, observed heterozygosity, PIC (Polymorphism Information Content) and number of unique alleles were determined. In addition, genetic distances according to Rogers distance were calculated. The values for all parameters were high, reflecting high genetic divergence of analyzed germplasm. Distance matrix based on UPGMA cluster analysis showed moderate congruence to available pedigree/origin or kernel type/color data. Genetic structure of tested genotypes was defined using Bayesian model-based clustering, without a considerable difference in comparison to cluster data analysis. The determined diversity parameters along with the results concerning genetic structure analysis provided a valuable information for improved selection efficiency.

    Abstract: During the 2014-2017 period, wheat samples were collected from discoloured spikes affected by Fusarium head blight (FHB) from 20 locations in Serbia. After isolation, fungi were cultivated on potato dextrose agar (PDA) at 25oC for 7 days. Based on the in situ identification on PDA, 36 isolates of the section Liseola were selected for further analyses. The pathogenicity of all isolates was confirmed on wheat leaves. The virulence of isolates was determined by measuring the lengths of spots formed at the inoculation leaf site. In order to prove the presence of the species Fusarium verticillioides, a pair of primers FV-F2/FV-R was used. This pair of primers amplifies the sequence of the gaoB gene, and it proved to be specific for the stated species. Moreover, for the same purpose, a pair of primers VER1-VER2 based on the calmodulin partial gene was used. The reference isolate RBG 1603 Q27 was used as a positive control. The pair of primers VER1-VER2 produced a band of the expected size - 578 bp in 18 isolates, while using FV-F2/FV-R, a 370bp long band confirmed the presence of F. verticillioides in 16 samples. Sixteen out of 18 isolates verified with VER1-VER2 were also identified as F. verticillioides with FV-FS/FV-R. No amplification was observed in a negative control.

    Abstract: Nine maize hybrids were grown with the aim to determine variations in chemical composition of the grain. Analyzed hybrids were grown in split-plot experimental design with tree replications in Sremska Mitrovica and Zemun Polje, during the summer of 2010 and 2011. Grain yield, protein, starch, oil, as well as, antioxidants like phytate, soluble phenolics and free sulfhydryl groups (PSH) were analyzed. Unfavorable meteorological conditions affected not only grain yield, but its chemical composition too, by decreasing starch, phenolics and PSH and increasing protein, oil and phytate concentrations. Hybrids from the FAO 600 maturity group achieved higher grain yield during unfavourable conditions, with higher starch and phytate contents and decreased levels of PSH and phenolics. Hybrids from the FAO 500 maturity group were much more stable in respect of protein and oil contents, irrespective to year and location. The grain yield did not have the same source of variations as the factors which determine nutritional quality, an exception being oil content, which decreased with the increase in the grain yield.

    Abstract: Taste and nutritional value make sweet corn a valued plant and an important component of the human diet worldwide. Kernel nutritive composition of sweet corn has been reported in various papers, but a description of carotenoid and tocopherols profile, especially after cooking is scarce. Therefore, the present study was carried out to compare the carotenoid and tocopherol content in sweet corn before and after cooking. Contents of b-carotene, lutein+zeaxanthin and tocopherols (d-T, b+g-T, a-T) in the kernels of twelve sweet corn hybrids were determined by High-Performance Liquid Chromatography (HPLC) and were expressed as the mean value of three independent measurements. Both genotype and cooking affected the content of the carotenoids and tocopherols in the kernel. The highest content of total carotenoids before and after cooking was found in hybrid ZP486/1su (27.77/45.28 µg/g) whereas the lowest content was in hybrid ZP 355su (10.27 µg/g) before cooking i.e. in hybrid ZP 347su (24.55 µg/g) after cooking. The cooking resulted in a significant increase in the content of total carotenoids and tocopherols, lutein+zeaxanthin, and b-carotene in all hybrids, except the ZP504su in which the b-carotene content decreased. An increase in a-tocopherol after cooking was observed in hybrids ZP485/1su and ZP484/1su, while a decrease was in hybrids ZP481/1su, ZP486/1su and ZP477/2su. The results showed that increasing micronutrient content is genotype-dependent. This study confirmed that cooking increases the nutritional value of sweet corn and gives it additional value in terms of functional food.
