Abstract:
L’avènement des nucléases programmables, notamment de CRISPR-Cas9, constitue une rupture technologique dans le domaine de l’ingénierie génétique. Le principe de ces méthodes est simple : il consiste à générer des cassures au niveau de séquences d’ADN cibles dans des cellules d’intérêt. Ces cassures sont ensuite réparées par un mécanisme de ligation non-homologue des extrémités pouvant conduire à l’inactivation d’un gène de la région modifiée, ou par recombinaison homologue permettant l’insertion d’un fragment de séquence apporté aux cellules. Les domaines d’application sont très nombreux : recherche fondamentale, thérapie génique, ingénierie écologique, biotechnologies, agriculture... Des exemples d’applications visant à améliorer la santé des animaux, à améliorer le bien-être animal ou l’éthique de la production, ou à modifier les produits pour une meilleure valeur santé ou nutritionnelle, sont présentés. L’utilisation de ces méthodes soulève de multiples questions (techniques, réglementaires, économiques, éthiques...) qui sont discutées.
