Abstract: The article presents the results of the application of the Antimicrobial Array II Plus test system based on the Randox® Biochip immunomicrochip technology in fish semi-finished products (frozen fish cutlets from pike) produced in the territory of the Russian Federation. The samples were taken from a retail chain in Moscow. The sensitivity of this method is 0,9 micrograms/l. As a result of our experiments, we found that this method has sensitivity and specificity in an amount not higher than that established in the Technical Regulations of the Eurasian Economic Union «On the Safety of fish and Fish Products» (EAEU TR 040/2016).

Abstract: Proteorhodopsins (PRs), bacterial light-driven outward proton pumps comprise the first discovered and largest family of rhodopsins, they play a significant role in life on the Earth. A big remaining mystery was that up-to-date there was no described bacterial rhodopsins pumping protons at acidic pH despite the fact that bacteria live in different pH environment. Here we describe conceptually new bacterial rhodopsins which are operating as outward proton pumps at acidic pH. A comprehensive function-structure study of a representative of a new clade of proton pumping rhodopsins which we name “mirror proteorhodopsins”, from Sphingomonas paucimobilis ( Spa R) shows cavity/gate architecture of the proton translocation pathway rather resembling channelrhodopsins than the known rhodopsin proton pumps. Another unique property of mirror proteorhodopsins is that proton pumping is inhibited by a millimolar concentration of zinc. We also show that mirror proteorhodopsins are extensively represented in opportunistic multidrug resistant human pathogens, plant growth-promoting and zinc solubilizing bacteria. They may be of optogenetic interest.

Abstract: NMDA ( N -methyl- d -aspartate) receptor antagonists are promising tools for the treatment of a wide variety of central nervous system impairments including major depressive disorder. We present here the activity optimization process of a biphenyl-based NMDA negative allosteric modulator (NAM) guided by free energy calculations, which led to a 100 times activity improvement (IC 50 = 50 nM) compared to a hit compound identified in virtual screening. Preliminary calculation results suggest a low affinity for the human ether-a-go-go-related gene ion channel (hERG), a high affinity for which was earlier one of the main obstacles for the development of first-generation NMDA-receptor negative allosteric modulators. The docking study and the molecular dynamics calculations suggest a completely different binding mode (ifenprodil-like) compared to another biaryl-based NMDA NAM EVT-101.

Abstract: Serial femtosecond crystallography (SFX) takes advantage of extremely bright and ultrashort pulses produced by x-ray free-electron lasers (XFELs), allowing for the collection of high-resolution diffraction intensities from micrometer-sized crystals at room temperature with minimal radiation damage, using the principle of “diffraction-before-destruction.” However, de novo structure factor phase determination using XFELs has been difficult so far. We demonstrate the ability to solve the crystallographic phase problem for SFX data collected with an XFEL using the anomalous signal from native sulfur atoms, leading to a bias-free room temperature structure of the human A 2A adenosine receptor at 1.9 Å resolution. The advancement was made possible by recent improvements in SFX data analysis and the design of injectors and delivery media for streaming hydrated microcrystals. This general method should accelerate structural studies of novel difficult-to-crystallize macromolecules and their complexes.

Abstract: The article presents the results of studying the corrosiveness of seven complex disinfectants containing, along with peracetic acid (PAA), hydrogen peroxide and acetic acid. All samples were encrypted. As test objects, details of the chicken processing line at the meat processing plant were used: bushings, fingers of a picking machine, round rubber discs, steel chain links, part of the conveyor belt and other parts of various shapes. It was found that all tested disinfectants based on PAA have a corrosive effect on the test objects of the chicken processing line. The most aggressive in effect were the means with the codes 02/22 and 03/22. The use of such disinfectants is economically unprofitable for meat processing enterprises.

Abstract: The article presents the results of studying the content of active substances in a number of acid disinfectants based on peracetic acid: peracetic acid, acetic acid, hydrogen peroxide, used directly on the chicken processing line of a large meat processing plant. Since some disinfectants had too strong a destructive effect on the parts of the chicken processing line, a comparison of the experimental results with the data of the manufacturer was carried out. Most of the products showed compliance with the content of active substances declared by the manufacturers.
