    Abstract: Urbānajā vidē dzīvie organismi atrodas nepārtrauktā saskarē ar dažāda veida piesārņojumu. Gan zemfrekvences elektromagnētiskais lauks (ZF EML), gan SiO 2 nanodaļiņas ir vispārzināmi urbānās vides piesārņojuma veidi. Kombinēta zemferekvences elektromagnētiskā lauka un SiO 2 ietekme uz šūnām ir maz pētīta. Šī pētījuma mērķis bija noteikt ZF EML un bioloģiski aktīvu (diametrā mazākas par 20 nm) SiO 2 nanodaļiņu ietekmi uz augu gametiskajām šūnām. Kā modeļa objekts pētījumā tika izmantotas Cyclamen persicum gametiskās šūnas vienkodola attīstības stadijā. Gametiskās šūnas tika kultivētas MS barotnē. Pirms ievietošanas ZF EML šūnas 1 h tika inkubētas barotnē, kam pievienoja SiO 2 nanodaļiņu suspensiju (1 mg/10 ml). Šūnas tika apstrādātas ZF EML 50 Hz ar blīvumu 50, 100, 200 un 400 μT. Šūnu relatīvās fluorescences izmaiņas tika noteiktas ar plūsmas citometru (BD FACSJazz ® ). Parādīts, ka gan SiO 2 klātūtne kultivācijas barotnē gan šūnu ievietošana ZF EML statistiski ticami palielina gametisko šūnu fluorescenci. Šūnu apstrāde ar ZF EML, pēc to inkubācijas SiO 2 nanodaļiņu suspensijā, statitiski ticami samazināja šūnu relatīvo fluorescenci. Rakstā tiek apspriesti iespējamie šāda efekta iemesli

    Abstract: The mitochondrial DNA (D-loop of mtDNA) sequencing method was used for investigation of various populations of Garganey Anas querquedula and Common Pochard Aythya ferina in the Western Palearctic. Reconstructed phylogenetic relationships based on comparison of D-loop sequences enabled to identify genetic similarity on the level of individual birds and to designate populations with different genetic structure. Identified haplotypic variability indicated the existence of two populations of Garganey with a different genetic structure in Europe. Some genetic variability was also observed among populations of Common Pochard of different origin. Haplotypic variability at different levels was found in both investigated species, despite a considerable overlap in wintering sites used by Garganeys and Common Pochards breeding in various regions.

    Abstract: Frequency of callusogenesis, type and capacity of regeneration, cell ploidy, DNA methylation level, histological features, autofluorescence and photobleaching effects were studied in flax calli grown on medium supplemented with different concentrations of carbon (C) nanoparticles. The presence of carbon nanoparticles on medium decreased the percentage calli formation. Detailed histological study led to better understanding of changes in calli caused by nanoparticles. Ploidy variation in calli cells significantly depended on carbon nanoparticle concentration in the medium. Different types of calli regeneration associated with different C nanoparticles concentration in medium were observed: somatic embryogenesis (direct or indirect) or organogenesis. C nanoparticle dose-dependent DNA hypermethylation in flax calli cells were found. Increased repair ability during laser irradiation was found in calli grown on medium with carbon nanoparticles

    Abstract: Accessions with no germinating seeds are a common problem in plant gene banks and research institutions. Our goal was to elaborate and apply an in vitro method of germination and multiplication of old aged seeds of red and alsike clover and alfalfa. Eighteen clover and five alfalfa accessions were used for germination in vitro. Most of the accessions had produced seeds more than 20 years ago and the seeds did not germinate in soil. Seed pre-treatment with different concentrations of potassium permanganate, as well as addition of phytohormones, AgNO3 and activated carbon to germinating media were tested. Plantlets for all germinated accessions were obtained, even in the case when seeds were 44-year-old (alfalfa). Germination rate in vitro not always correlated with seed age and ranged from 2 to 72%. Pre-treatment with potassium permanganate was effective both for seeds sterilisation and germination stimulation. Most germinated seeds formed phenotypically normal seedlings with all organs. In vitro multiplication of obtained clover plants was performed. Best results were achieved by micropropagation of stem segments approximately 2 mm in length. Number of finally obtained plants depended not only on seed germination ability, but also on micropropagation ability in relation to genotype, and on acclimatization success in soil after in vitro cultivation

    Abstract: Baltais āboliņš (Trifolium repens L.) ir tauriņziežu dzimtas augs, plaši izplatīts gan pilsētvidē, gan ārpus tās. Tas tiek izmantots kā lauksaimniecības augs biomasas iegūšanai, un tā ir arī populāra daudzgadīgos zālienus veidojošā suga - pilsētvidē balto āboliņu sēj zālājos. Baltais āboliņš plaši pārstāvēts dažādos biotopos ar mainīgu ārējo faktoru ietekmi, tai skaitā pilsētvidē. Tādēļ tas ir piemērots modeļobjekts antropogēnās ietekmes pētīšanai. Darba mērķis bija noteikt baltā āboliņa Latvijas populācijas ģenētisko daudzveidību, izmantojot iPBS (inter primer binding sites) - uz retrotranspazoniem balstītu metodi. Iegūtie rezultāti salīdzināti starp trim augu paraugkopām: pilsētvidē augoši augi (Rīgas pilsētas teritorija), dabiskā vide (Latvijas novadu teritorijas) un selekcionētā šķirne 'Daile'. Lai gan polimorfo lokusu skaits visās pētītajās grupās bija augsts (89%), tomēr Šanona (Shannon) indekss, kas raksturo grupu iekšējo ģenētisko daudzveidību, pētāmajām grupām svārstījās tikai ap 0.3. Šāds rezultāts norāda uz to, ka iekšējo grupu polimorfismu nosaka neliels ģenētiski atšķirīgu augu skaits. Galveno koordināšu analīzē, kas balstīta uz Šanona indeksu, visi pētītie paraugi izveidoja trīs grupas. Vienā no tiem grupējās šķirnes 'Daile' augi, savukārt baltais āboliņš no pilsētvides un Latvijas reģioniem izveidoja divas atšķirīgas grupas.

    Abstract: Mute swan Cygnus olor is the most abundant breeding swan species in Rīga and Latvia. Since manmade habitats are the preferred breeding habitats of this species - Cygnus olor is of particular interest in studies of urban biodiversity. The latest records show that 30-40 pairs breed annually in Rīga, 400-700 individuals migrate through the area, and 120-150 individuals stay over winter. The goal of our study was to determine the genetic variation of the mute swan population in Rīga. Blood samples were collected from 47 individual birds found in different sites in Rīga or elsewhere in Latvia. The universal retrotransposon based iPBS markers were used to estimate diversity. Three primers (2076, 2080, and 2415) amplified 64 DNA fragments; of these 18 (28%) were polymorphic. The average diversity index based on polymorphic markers for the studied individuals was 0.425. All individuals, with the exception of two pairs of individuals, could be separated by means of these markers. The grouping of individuals was not be associated with their sampling location, gender and age. The genetic diversity indexes were compared between these groups. The breeding individuals (including their cygnets) had bands not showing variation among breeding birds as compared to the not breeding ones.

    Abstract: According to the Global Strategy for Plant Conservation, 75% of endangered species should be preserved in ex situ collections till 2020. The genus Liparis has a lot of recognised taxons, but only one species, Liparis loeselii (L.) Rich., grows in Europe. L. loeselii is a rare and endangered orchid species occurring in Europe. In Latvia L. loeselii is classified as the third category of endangered and protected species. To develop the best conservation strategy, the knowledge concerning the genetic differences of protected plants in a particular area is crucial. For this purpose, the genetic diversity of L. loeselii populations from different Latvian habitats was tested. The inter-retrotransposon amplified polymorphism method (iPBS) was used for population genetic diversity evolution. In total, 54 accessions from nine habitats were collected and analysed. L. loeselii leaves have a high content of phenols that reduce the quality of extracted DNA. It was found that the percentage of polymorph loci varied among the populations of L. loeselii growing in different habitats; some of the populations were genetically homogeneous. The genetic diversity levels of L. loeselii populations are related with the population age and the growing conditions.

    Abstract: Nanoparticles (nano-scale particles (NSPs)) are defined as particles with dimensions less than 100 nm. SiO 〈 sub 〉 2 〈 /sub 〉 nanoparticles are one of the most widely common nanoparticles in the environment, particularly in urban areas. The sources of SiO 〈 sub 〉 2 〈 /sub 〉 nanoparticles are very different, including natural nanoparticles, anthropogenic and engineered nanoparticles. The SiO 〈 sub 〉 2 〈 /sub 〉 nanoparticles could be considered a source of different pollution effects on leaving organisms. Nevertheless, knowledge of the mechanisms, through which the SiO 〈 sub 〉 2 〈 /sub 〉 nanoparticles affect cells, is incomplete. The aim of the research was to elaborate a method to determine changes in relative fluorescence of both somatic and immature gametic plant cells in presence of SiO 〈 sub 〉 2 〈 /sub 〉 nanoparticles. Relative cell fluorescence was measured with BD FACSJazz® cell sorter using 488 nm exciting laser light. Mean cell fluorescence was determined for samples of purified cell suspension. Gates of different size and shape were preliminary tested to find those with the lowest CV. Cell plots were created by BS FACS Software 1.0.0.650. The densest part of the plot was gated using oval-shaped gate. The gate included 95-99% of all cells. A logarithmic scale in arbitrary fluorescence units was applied to determine cell relative fluorescence. More than 10 000 cells were gated and analysed from each sample. Somatic cell culture from callus culture initiated from leaves of flax ( 〈 em 〉 Linum usitatissimum) 〈 /em 〉 was obtained. The relative fluorescence of the somatic cells had large distribution, since the cells differ by many parameters (size, shape, metabolism etc.). Immature pollen cells (one-nucleus stage) as best for SiO 〈 sub 〉 2 〈 /sub 〉 nanoparticles influence investigation were found. The influence of SiO 〈 sub 〉 2 〈 /sub 〉 nanoparticles on several plant species ( 〈 em 〉 Cyclamen persicum, Tilia cordata, Hordeum vulgare 〈 /em 〉 and 〈 em 〉 Triticum 〈 /em 〉 〈 em 〉 aestivum 〈 /em 〉 ) immature pollen cells were investigated. A significant increase in relative cell fluorescence was observed for all mentioned plant species cells after incubation in SiO 〈 sub 〉 2 〈 /sub 〉 nanoparticles suspension. It was found that cell relative fluorescence was dependent on cultivation duration in SiO 〈 sub 〉 2 〈 /sub 〉 nanoparticles suspension.
