Abstract:
Cytotoxisch und somit für die Krebstherapie interessant sind die Derivate des Cephalotaxins, der Stammverbindung der aus südostasiatischen Eiben isolierten Harringtonine. Durch intramolekulare Pd‐katalysierte allylische Aminierung zum Spirocyclus (85% Ausbeute) und anschließende Heck‐Reaktion zum Siebenring (80% Ausbeute) gelang es nun, die Cephalotaxin‐Vorstufe 2 in zwei Stufen hocheffizient und selektiv aus dem leicht zugänglichen Amin 1 herzustellen. magnified image
