    Abstract: This investigation was carried out on semen of three Gir bulls at Cattle Breeding Farm, Junagadh (India) for a period of 6 months. The aim was to assess the effect of different concentrations of Curcumin, viz., 0 µM (Control), 5 µM, 10 µM, and 15 µM in Andromed extender on sperm quality along with oxidative stress parameters of cryopreserved semen. Semen ejaculates (n=30, 10/bull) with 〉 70% initial motility were included in the study. Soon after initial evaluation, the semen was extended in Andromed extender @ 80 million sperm per ml and was divided into four equal aliquots. The Curcumin stock solution (10 mM) was added in these aliquots to get a final concentration of 0, 5, 10, and 15 µM and sperm motility was checked. It was soon filled and sealed in medium French straws (0.5 ml), equilibrated at 4°C for 4 h and frozen in LN2 vapour using conventional wide mouth freezer. Thawing of straws was done next day in water bath at 37°C for 30 seconds, and various sperm quality parameters including oxidative markers were assessed. The results showed that supplementation of Curcumin to freezing extender improved semen quality and antioxidant capacity. The post-thaw sperm motility, sperm viability, HOST reactive sperm and acrosomal integrity in semen extended with 15 µM Curcumin concentration (57.33±0.67, 73.20±1.05, 64.77±1.56 and 72.43±1.38 %, respectively) were significantly (p 〈 0.05) higher and sperm abnormality (18.00±0.41%) was significantly (p 〈 0.05) lower as compared to those of control extender and the 5 µM Curcumin group. Mean malondialdehyde (MDA) level in post-thaw seminal plasma did not differ significantly (p 〉 0.05) among the groups, while total antioxidants activity (TAC) with 15 µM Curcumin (476.36±16.52 μmol/L) was significantly (p 〈 0.05) higher as compared to control and 5 µM Curcumin group. It is concluded that Curcumin @ 15 µM in semen extender improves post-thaw sperm quality and antioxidant capacity of cryopreserved Gir bull semen.

    Abstract: The present investigation was undertaken on four mature Gir bulls (4-6 years) maintained at University farm,  Junagadh for 4 months during hot summer season. Semen (n=6 ejaculate/bull) was collected using artificial  vagina once a week from each bull. The fresh ejaculates were evaluated for physical attributes and oxidative  stress markers in the seminal plasma. The overall mean values recorded based on 24 ejaculates for ejaculate  volume, sperm concentration, mass motility (0-5 scale), individual motility, sperm viability, sperm abnormality,  plasma membrane integrity (HOST reactive sperm), and acrosome integrity were 6.20 ± 1.27 mL, 1316.83 ±  155.07 million/ mL, 3.75 ± 0.20, 84.83 ± 0.94 %, 87.58 ± 1.07 %, 9.63 ± 0.83 %, 79.58 ± 1.01 % and 91.21 ± 0.52  %, respectively. There was a significant P 〈 0.001) difference in ejaculate volume, sperm concentration and sperm  abnormality between bulls. The levels of oxidative stress parameters, viz. lipid peroxidation and glutathione S-transferase (GST) activity in the seminal plasma were 5.51 ± 0.26 μmol/mL and 16.30 ± 2.14 nmol/ mL/min, respectively. Individual motility, viability, HOST reactive spermatozoa, and acrosomal integrity were positively correlated, suggesting that any one or two attributeslike motility or HOS test can predict the semen quality. The  semen quality generally was in normal physiological range including oxidative markers even during heat stress.

    Abstract: Caffeine is widely known for its phosphodiesterase enzymes inhibiting, and its powerful reactive oxygen species scavenging property. This study aimed to evaluate the effect of caffeine supplementation @ 0 mM, 1 mM, 3 mM and 5 mM in AndroMed® extender on cryopreservation of Jaffarabadi buffalo semen. Twenty-four semen ejaculates (6/bull) with 〉 70% initial sperm motility were obtained from four mature Jaffarabadi bulls using AV. The ejaculates were split-diluted in an egg-yolk-free AndroMed® extender supplemented with different concentrations of caffeine and were cryopreserved using a standard protocol. The semen samples were evaluated for sperm quality parameters as well as oxidative stress parameters, viz., lipid peroxidation, and Glutathione-S-transferase (GST) enzyme activity in seminal plasma at pre-freeze (after equilibration) and post-thaw stage. The levels of caffeine had a significant effect on all these parameters, except sperm abnormalities, at both pre-freeze and post-thaw stages. Supplementation of caffeine in semen extender at 1 mM and 3 mM concentration showed a significant (p 〈 0.05) increase in post-thawed sperm motility (62.04 ± 0.84, 61.25 ± 1.01%), viability (64.21 ± 0.88, 64.25 ± 0.84%), acrosome integrity (58.08 ± 1.08, 57.30 ± 0.93%) and plasma membrane integrity (52.75 ± 0.89, 52.71 ± 0.74%) and a significant (p 〈 0.05) decrease in oxidative damage as evident by lower lipid peroxidation (MDA 7.62 ± 0.41, 7.80 ± 0.70 μM) and GST enzyme activity (31.15 ± 1.36, 29.54 ± 0.54 nmol CDNB/mL/min) as compared to control and 5 mM caffeine. The study concludes that the post-thaw quality of frozen semen of Jaffarabadi buffalo bulls improves significantly with decreased oxidative stress if the AndroMed® extender is supplemented with 1 and 3 mM concentration of caffeine over control.

    Abstract: Cleaning wastewater has become one of the most serious issues for a number of scientists and researchers in recent years, as water is the most basic need for the daily life of humans. There has been a focus on the removal of noxious pollutants from wastewater effluents by using nanocatalysts owing to their unique physicochemical actions and stability. Herein we manufactured TiO2 nanoparticles supported by activated carbon (AC-TiO2) using a cost-effective sonochemical method. The band structures of the AC-TiO2 and TiO2 were modified from 3.2 to 3.1 eV, thus increasing the catalytic activity. The structural, optical and anatase crystal phase properties, with morphological confirmation, were studied by applying UV-DRS, PL, FESEM, XRD, along with HRTEM, respectively. The specific surface area, calculated by BET analysis, was found to be ~241 m2/gm and ~46 m2/gm for AC-TiO2 and TiO2. The degradation efficiency of the as-prepared nanocatalysts against the very toxic but rarely studied organic textile dye pollutant RO 84 was investigated and 97% efficiency were found for the AC-TiO2 as compared to pure TiO2, which is a highly appreciated finding in the catalytic dye degradation application domain. Such surface-modified nanocatalysts could be further implemented for the treatment of wastewaters/waste effluents released from chemical industries, laboratories and other sources.

    Abstract: At present, chemical Si/Al sources are mainly used as precursor materials for the manufacturing of zeolites. Such precursor materials are quite expensive for commercial synthesis. Here, we have reported the synthesis of Ca-based zeolite from incense stick ash waste by the alkali-treatment method for the first time. Incense stick ash (ISA) was used as a precursor material for the synthesis of low Si zeolites by the alkali-treatment method. The as-synthesized zeolites were characterized by various instruments like particle size analyzer (PSA), Fourier transform infrared (FTIR), X-ray diffraction (XRD), field emission scanning electron microscope (FESEM), electron diffraction spectroscopy (EDS), transmission electron microscopy (TEM), and X-ray fluorescence (XRF). FTIR and XRD helped in the identification of the microstructure and crystalline nature of the zeolites and also confirmed the synthesis of Ca-based zeolite with two thetas at 25.7°. The microscopic analysis by FESEM and TEM exhibited that the size of synthesized Ca-rich zeolites varies from 200 to 700 nm and they are aggregated and cuboidal in shape. Additionally, structural, electronic, and density of states’ characteristics of gismondine (Ca2Al4Si4O16·9H2O) structures were evaluated by computational simulations (first principle, density functional theorem). The structural optimization of structures was carried out in the first stage under the lowest condition of total energy and forces acting on atoms for the lattice constant, as well as the available experimental and theoretical findings. The present research approach predicted the transformation of ISA waste into a value-added mineral, i.e., zeolite, which was further used for the removal of both heavy metals and alkali metals from fly ash-based wastewater using inductively coupled plasma-optical emission spectroscopy (ICP-OES).
