    Abstract: Traditional Chinese opera, such as Pingju , requires actors to master sophisticated performance skills and cultural knowledge, potentially influencing brain function. This study aimed to explore the effects of long-term opera training on the dynamic amplitude of low-frequency fluctuation (dALFF) and dynamic functional connectivity (dFC). Methods Twenty professional well-trained Pingju actors and twenty demographically matched untrained subjects were recruited. Resting-state functional magnetic resonance imaging (fMRI) data were collected to assess dALFF differences in spontaneous regional brain activity between the actors and untrained participants. Brain regions with altered dALFF were selected as the seeds for the subsequent dFC analysis. Statistical comparisons examined differences between groups, while correlation analyses explored the relationships between dALFF and dFC, as well as the associations between these neural measures and the duration of Pingju training. Results Compared with untrained subjects, professional Pingju actors exhibited significantly lower dALFF in the right lingual gyrus. Additionally, actors showed increased dFC between the right lingual gyrus and the bilateral cerebellum, as well as between the right lingual gyrus and the bilateral midbrain/red nucleus/thalamus, compared with untrained subjects. Furthermore, a negative correlation was found between the dALFF in the right lingual gyrus and its dFC, and a significant association was found between dFC in the bilateral midbrain/red nucleus/thalamus and the duration of Pingju training. Conclusion Long-term engagement in Pingju training induces neuroplastic changes, reflected in altered dALFF and dFC. These findings provide evidence for the interaction between artistic training and brain function, highlighting the need for further research into the impact of professional training on cognitive functions.

    Abstract: Steady-State Visual Evoked Potentials (SSVEPs) are widely used in spatial selective attention. In this process the two kinds of visual simulators, Light Emitting Diode (LED) and Liquid Crystal Display (LCD), are commonly used to evoke SSVEP. In this paper, the differences of SSVEP caused by these two stimulators in the study of spatial selective attention were investigated. Results indicated that LED could stimulate strong SSVEP component on occipital lobe, and the frequency of evoked SSVEP had high precision and wide range as compared to LCD. Moreover a significant difference between noticed and unnoticed frequencies in spectrum was observed whereas in LCD mode this difference was limited and selectable frequencies were also limited. Our experimental finding suggested that average classification accuracies among all the test subjects in our experiments were 0.938 and 0.853 in LED and LCD mode, respectively. These results indicate that LED simulator is appropriate for evoking the SSVEP for the study of spatial selective attention.

    Abstract: Circulating tumor cells (CTCs) play a key role in cancer progression, especially metastasis, due to the rarity and heterogeneity of CTCs, fewer researches have been conducted on them at the molecular level. However, through the Gene Expression Omnibus (GEO) database, this kind of minority researches can be well integrated, the gene expression differences between CTCs and primary tumors can be identified, and molecular targets for CTCs can be found. Methods We analyzed 7 sets of gene chips (GSE82198, GSE99394, GSE31023, GSE65505, GSE67982, GSE76250, GSE50746) obtained by GEO. Analysis of differentially expressed genes (DEGs) between CTCs and corresponding primary tumors by NetworkAnalyst. Metascape tool for Gene Ontology (GO) / Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis of differential genes and visual display. Cytoscape performs protein-protein interaction (PPI) analysis and obtains the hub genes. Renal cancer patients’ clinical specimens to verify the correctness of enrichment results. Prognostic analysis of hub genes in kidney cancer patients using the Kaplan–Meier plotter survival analysis tool. Results We obtained a total of 589 DEGs. The GO / KEGG enrichment results indicate that the DEGs are mainly concentrated in cell adhesion, epithelial-mesenchymal transition (EMT), and apoptosis. Renal cancer clinical specimens suggest that CTCs have epithelial and mesenchymal types. At the same time, PSMC2 can be used as a poor prognostic indicator for renal cancer patients. Conclusions In summary, our study suggests that compared with primary tumors, CTCs mainly change cell adhesion, EMT, and apoptosis. PSMC2 can be used as a poor prognostic factor.

    Abstract: Rhenium disulfide (ReS 2 ) with distinct physicochemical properties has shown promising potential in disease theranostics, such as drug delivery, computed tomography (CT), radiotherapy, and photothermal therapy (PTT). However, the synthesis and post-modification of ReS 2 agents for different application scenarios are time- and energy-consuming, which seriously hinders the clinical translation of ReS 2 . Herein, we proposed three facile excipient strategies for different theranostic applications of ReS 2 just through the flexible use of commercial ReS 2 powder. Three excipients, including sodium alginate (ALG), xanthan gum (XG), and ultraviolet-cured resin (UCR), were used to prepare different dosage forms of commercial ReS 2 powder, like hydrogel, suspension, and capsule, respectively. These dosage forms of ReS 2 with distinct characteristics showed great potential for second near-infrared window PTT against tumours, gastric spectral CT imaging, and functional evaluation of the digestive tract in vivo . In addition, these ReS 2 formulations exhibited good biocompatibility both in vitro and in vivo , showing a promising prospect for clinical transformation. More importantly, the facile excipient strategies for commercial agents pave a bridge to the development and wide bioapplication of many other theranostic biomaterials.

    Abstract: Major civil and military infrastructures are usually located in mountainous areas with complex geological structures. When subjected to impact loads caused by operational blasting or seismic activities, the inherent flaws or joints in the hosting rock mass may grow irreversibly and eventually and cause the entire structure to collapse. It is, therefore, desirable to understand the mechanical and cracking behaviors of flawed rock structures under dynamic loads. In this work, the cement mortar is used to make the plate specimens, and two different types of flaws, i.e., the open and the closed (resin-filled) flaws, with different inclination angles from 0° to 90° with respect to the loading direction are prefabricated. The nominal strength, crack initiation, and failure characteristics of the specimens under different loading rates are investigated using the split Hopkinson bar (SHPB) system in conjunction with the digital image correlation (DIC) technique. The results indicate that the dynamic nominal strength of both two types of specimens shows similar loading rate and flaw angle dependency, specifically, monotonically increasing with the increase of the loading rate while increasing first and then decreasing with increasing the flaw inclination angle. The strength of specimens with an open flaw is obviously lower than that of the specimens with a closed flaw at a similar loading rate. Besides, the failure mode of specimens with an open flaw is mainly an X-shaped tensile or tensile-shear mixed form regardless of the flaw inclination, whereas the resin-filled flawed specimens always fail in the form of an atypical X-shaped shear pattern with obvious randomness. Moreover, the stress field around the crack tip is carefully extracted based on the obtained displacement field from DIC analysis; the stress intensity factors at crack initiation onset also show flaw inclination- and loading rate-dependent behavior. The strength, cracking behavior, and stress field around the tip during the dynamic loading are closely related to the friction effect between the flaw surfaces. The findings in this work provide some basic insights into the cracking mechanism of rock with open and closed flaws under dynamic loading conditions.

    Abstract: Background: SLC1A5, a ferroptosis regulator gene, plays a dual role in cancer regulation. However, the roles of SLC1A5 in pancreatic adenocarcinoma (PAAD) remain elusive. Methods: SLC1A5’s expression and somatic mutation information were determined by TCGA, GEO, Oncomine, and cBioPortal databases. Its prognostic value was assessed in TCGA cohort and was validated in three independent cohorts. The effects of SLC1A5 on the tumor immune microenvironment were analyzed by the CIBERSORT algorithm, ssGSEA method, and TISIDB and TIMER databases. The “oncoPredict” R package, TIDE algorithm, ImmuCellAI online tool, and GSE35141 and GSE59357 datasets were used to ascertain its therapeutic correlations. GSEA and Western blot were applied to reveal the effects of SLC1A5 on the mTORC1 signaling pathway and ferroptosis process. The biofunctions of SLC1A5 were assessed by MTT, wound-healing, Transwell, and xenograft assays. Results: SLC1A5 was significantly upregulated in the PAAD samples but was not commonly accompanied with somatic mutation (2.3%). Overexpression of SLC1A5 led to a poor prognosis and was identified as an independent prognostic factor. Moreover, high SLC1A5 expression suppressed the antitumor immune process by changing the infiltrating levels of immune cells. As for therapeutic correlations, SLC1A5 was related to the efficacy of dasatinib, sunitinib, sorafenib, and imatinib but may not predict that of radiotherapy, chemotherapeutic drugs, and immune checkpoints inhibitors (ICIs). Notably, the overexpression of SLC1A5 could activate the mTORC1 signaling pathway and may increase the cellular sensitivity to ferroptosis. Finally, the overexpression of SLC1A5 markedly promoted proliferation, migration, and invasion of pancreatic cancer cells. At the in vivo level, SLC1A5 deletion inhibited tumor growth in a mice xenograft model. Conclusions: SLC1A5 prefers to play as an accomplice rather than an opponent in PAAD. Our findings provide novel insights into PAAD treatment.

    Abstract: Nonlinear modulation of the dopamine signaling on brain functions can be estimated by the interaction effects of dopamine-related genetic variations. We aimed to explore the interaction effects of COMT rs4680 and DRD2 rs1076560 on intra-network connectivity using independent component analysis. In 250 young healthy adults, we identified 11 meaningful resting-state networks (RSNs), including the salience, visual, auditory, default-mode, sensorimotor, attention and frontoparietal networks. A two-way analysis of covariance was used to investigate COMT×DRD2 interactions on intra-network connectivity in each network, controlling for age, gender and education. Significant COMT×DRD2 interaction was found in intra-network connectivity in the left medial prefrontal cortex of the anterior default-mode network, in the right dorsolateral frontal cortex of the right dorsal attention network and in the left dorsal anterior cingulate cortex of the salience network. Post hoc tests revealed that these interactions were driven by the differential effects of DRD2 genotypes on intra-network connectivity in different COMT genotypic subgroups. Moreover, even in the same COMT subgroup, the modulation effects of DRD2 on intra-network connectivity were different across RSNs. These findings suggest a network-dependent modulation of the DA-related genetic variations on intra-network connectivity.
