Abstract: The Pinelemabailongensis Wang & amp; Li, 2012 species group of the spider family Telemidae Fage, 1913 from Guangxi and Guizhou, China is revised. Previously, this species group contained two species: P.bailongensis and P.xiushuiensis Wang & amp; Li, 2016. In this study, four species are transferred from Telema Simon, 1882 to Pinelema Wang & amp; Li, 2012, and treated as members of the P.bailongensis species group. They are P.cordata (Wang & amp; Li, 2010) comb. n. , P.liangxi (Zhu & amp; Chen, 2002) comb. n. , P.strentarsi (Lin & amp; Li, 2010) comb. n. and P.zhewang (Chen & amp; Zhu, 2009) comb. n. Additionally, six new species belonging to the species group are described: P.cheni Zhao & amp; Li, sp. n. (♂♀), P.huoyan Zhao & amp; Li, sp. n. (♂♀), P.lizhuang Zhao & amp; Li, sp. n. (♂♀), P.wangshang Zhao & amp; Li, sp. n. (♂♀), P.wenyang Zhao & amp; Li, sp. n. (♂♀) and P.yunchuni Zhao & amp; Li, sp. n. (♂♀). A key to males is provided.

Abstract: In this study, we sequenced two complete mitogenomes from Deroceras laeve and Ambigolimax valentianus . The mitogenome of Ambigolimax valentianus represented the first such data from the family Limacidae. The lengths of the mitogenomes of Deroceras laeve and Ambigolimax valentianus were 14,773 bp and 15,195 bp, respectively. The entire set of 37 mitochondrial genes were identified for both mitogenomes. Compared with the mitogenome of Achatina fulica , the trnP _ trnA tRNA cluster was rearranged in both Deroceras laeve and Ambigolimax valentianus . The secondary structures of tRNA and rRNA genes for the two species were predicted. Phylogenetic analyses based on amino acid sequences supported (1) monophyly of Stylommatophora, (2) division of Stylommatophora into the ‘achatinoid’ clade (i.e., the suborder Achatinina) and the ‘non-achatinoid’ clade (i.e., the suborder Helicina), (3) placement of the Orthurethra in the ‘non-achatinoid’ clade, and (4) monophyly of each of the superfamilies Helicoidea, Urocoptoidea, Succineoidea, Arionoidea, Pupilloidea and Limacoidea. The exemplars of Helicidae, Philomycidae and Achatinellidae displayed many more mitochondrial gene rearrangements than other species of Stylommatophora.

Abstract: The complete mitochondrial genome (mitogenome) of Yponomeuta montanatus is sequenced and compared with other published yponomeutoid mitogenomes. The mitogenome is circular, 15,349 bp long, and includes the typical metazoan mitochondrial genes (13 protein-coding genes, two ribosomal RNA genes, and 22 transfer RNA genes) and an A + T-rich region. All 13 protein-coding genes use a typical start codon ATN, the one exception being cox1 , which uses CGA across yponomeutoid mitogenomes. Comparative analyses further show that the secondary structures of tRNAs are conserved, including loss of the Dihydorouidine (DHU) arm in trnS1 (AGN), but remarkable nucleotide variation has occurred mainly in the DHU arms and pseudouridine (TψC) loops. A + T-rich regions exhibit substantial length variation among yponomeutoid mitogenomes, and conserved sequence blocks are recognized but some of them are not present in all species. Multiple phylogenetic analyses confirm the position of Y. montanatus in Yponomeutoidea. However, the superfamily-level relationships in the Macroheterocera clade in Lepidoptera recovered herein show considerable difference with that recovered in previous mitogenomic studies, raising the necessity of extensive phylogenetic investigation when more mitogenomes become available for this clade.

Abstract: The flying squirrel genus Biswamoyopterus (Rodentia: Sciuridae: Pteromyini) was once considered to contain three species, Biswamoyopterus biswasi from northeastern India, B. laoensis from central Laos and B. gaoligongensis from southwest China, all identified from morphological characteristics of one or two specimens. However, based on similar morphological characteristics of two samples of the genus Biswamoyopterus collected recently from northern Laos and northern Myanmar, and the small genetic distances on mitochondrial DNA and nuclear DNA between them, the results strongly support these two samples as representatives of the same species. The phylogenetic analyses strongly support Biswamoyopterus as an independent genus of Pteromyini, as a sister group to Aeromys . Biswamoyopterus biswasi is distributed in the northern Indo-China peninsula, where it is exposed to a series of threats, such as intense hunting activity, illegal trade, and rapid habitat loss; this should warrant its classification as critically endangered according to the International Union for Conservation of Nature (IUCN) Red List criteria. Here, the molecular data for genus Biswamoyopterus and two new specimen records from northern Laos and northern Myanmar are presented.

Abstract: Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli . Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals 〈 1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.
