[spa] Durante la realización de la presente Tesis Doctoral se ha establecido que el método más eficiente para la síntesis de la (4ªS,5R)-4a,5-dimetil-4,4a,5,6,7,8-hexahidronaftalen-2(3H)-ona (-)-3 desarrollado hasta el presente consiste en la doble alquilación de la (R)-3-metilciclohexanona para generar un precursor de la lactona de enol bicíclica de Piers. La apertura de la misma por reacción con el derivado lítico del metilfosfonato de dimetilo y subsiguiente reacción de Wadsworth-… [+]Emmons intramolecular rinde la enona (-)-3 de manera satisfactoria. Por contra, para el acceso al enantiómero (+)-3, el método más eficiente consiste en la preparación de la cetona de Wieland-Miescher y la transformación del grupo carbonilo aislado en un grupo metilo mediante un proceso de integración diastereoselectivo. Se ha establecido la configuración relativa del C13 y la configuración absoluta del diterpenoide (+)-nakamurol A, mediante síntesis total del (-)-nakamurol A. La identificación de los datos de RMN (1H y 13C) comparando los descritos para el producto natural y el sintetizado así como la medida del poder rotatorio del producto sintetizado permiten elucidar en base al proceso de síntesis la configuración absoluta del producto natural. Se ha determinado de forma inequívoca la configuración del C13 mediante un proceso de síntesis enantioespecífico que tiene como etapa clave una epoxidación asimétrica de Sharpless. La configuración queda establecida no sólo en base de la esperada selectividad en base al reactivo empleado, sino también por análisis pormenorizado de los datos de RMN. La primera síntesis total del nakamurol A, consta de 20 etapas. También se ha establecido un ruta más corta (16 etapas) pero que carece de estereoselectividad en la formación del centro estereogénico en C13. Se ha establecido la configuración absoluta del sesquiterpenoide xilarenal A mediante síntesis total y comprobación de la identidad en los datos de RMN y poder rotatorio entre los registrados a partir de la muestra sintetizada y los descritos para el producto natural. Se ha establecido la estereoselectividad en las reacciones de gamma-oxidación y alfa'-alquilación en el compuesto bicíclico intermedio 3. ; [eng] This PhD was initially directed to establishing an efficient method for the synthesis of enantiomerically pure (4aS,5R)-hexahydro-4a,5-dimethyl-2(3H)-naphthalenone (-)-3. The synthesis was started from (R)-3-methylcyclohexanone and involved the preparation of the Piers enol lactone in enantiopure form as the key intermediate. Treatment of this lactone with lithium salt of dimethyl methylphosphonate gave the bicyclic enone (-)-3. Alternatively, enantiomerically pure (+)-3 was prepared using the … [+]Wieland-Miescher ketone as the starting material. The transformation of the isolated ketone to the methyl group was carried out in a diasterioselective manner. The total synthesis of the enantiomer of the marine sponge diterpenoid Nakamurol A and determination of the absolute configuration of this natural product are reported. This first synthetic entry to thelepogane-type diterpenoids involves the use of the bicyclic enone (-)-3, which after a tandem difunctionalization process and elongation of the side chain leads to the formation of an advanced intermediate incorporating a methylketone. From the latter, two approaches to ent-Nakamurol A are reported: the straightforward but nonselective way, by reaction with vinylmagnesium bromide, and a longer but stereocontrolled route, through the primary allylic alcohol, which is submitted to a Sharpless epoxidation followed by a tellurium-promoted reductive-epoxide ring-opening cascade reaction. The total synthesis of the sesquiterpenoid Xylarenal A is reported. This first synthetic entry to this eremophylan terpenoid with an exocyclic vinyl aldehyde unit involves the use of the bicyclic enone (+)-3, with after an '-alpha alkylation and a gamma-oxidation leads to the formation of a new bicyclic ketone. From the latter, sequential acylation, ozonolysis of the allyl side chain and alpha-methylenation of the resulting aldehyde gives Xylarenal A.
