    Content: Chromothripsis is a form of genomic instability that begins with a shattering event leading to clustered chromosomal aberrations. Chromothriptic cells acquire multiple genomic alterations in confined genomic regions restricted to one or a few chromosomes. Due to the very high prevalence of chromothripsis in different types of cancers especially Sonic Hedgehog medulloblastomas, osteosarcomas and neuroblastomas and due to the link between chromothripsis and poor prognosis for cancer patients, finding therapeutic options to specifically target tumor cells with chromothripsis is highly relevant. Chromothripsis is linked to homologous recombination (HR) repair deficiency in a number of tumor types. Therefore, synthetic lethality may potentially be used, whereby HR repair deficiencies of cancer cells are exploited by applying a PARP inhibitor, which spares repair-proficient normal cells. In the primary screen, I searched for potential additive or synergistic partners of a PARP inhibitor (BGB290) and also searched for drugs that are potent as single agents. The analysis of the primary screen done in 15 cell lines identified romidepsin and nanaomycin A as promising candidates to eliminate tumor cells with chromothripsis. To confirm the potential additivity or synergism of these drugs with the PARP inhibitor, a secondary screen was done where drugs were titrated against each other in 8×8 matrix settings. The secondary screen revealed a strong synergistic interaction between BGB290 and romidepsin in chromothriptic Sonic Hedgehog (SHH) medulloblastoma patient derived xenograft spheroid models but antagonism in non-chromothriptic control cells. Functional assays revealed that the synergistic effect between BGB290 and romidepsin was not TP53 dependent. Romidepsin and BGB290 combination treatment caused G2 cell cycle arrest and apoptosis in chromothriptic DAOY cells. The mitotic rate was decreased by romidepsin and BGB290 combination treatment in DAOY cells as shown by acetyl-αtubulin and phospho-histone H3 immunostaining. Gene expression analysis revealed that the romidepsin and BGB290 combination treatment was associated to the regulation of MYC target genes. To further analyze the link between MYC expression and the effects of the combination treatment, I used an inducible MYC ON / OFF system. When MYC was overexpressed, romidepsin and BGB290 showed additivity, while in the MYC OFF state the additive effect was replaced by antagonism. This indicates that MYC overexpression plays a role in the interaction between BGB290 and romidepsin. Lastly, the in vivo efficacy of BGB290 and romidepsin combination treatment was confirmed in patient-derived xenograft mouse models.

    Content: This study examines the relationship between career commitment and employee career success (objective and subjective success) in middle-level employees working in the service sector. Further, the study investigates the moderating effect of work-life balance on the relationship between career commitment and career success. By analyzing data from 360 middle level working employees, our empirical results show that career commitment has a positive and significant effect on the objective and subjective career success of employees. Work-life balance positively moderates the relationship between career commitment and subjective career success, whereas it fails to moderate the relationship between career commitment and objective career success. The study contributes by providing a better understanding of the employee’s perception of career commitment and career success and their management in emerging markets. Avenues for future research are identified.

    Content: Purpose – The purpose of this paper is to provide an exploratory analysis Islamic finance products for housing finance in the UK market and make a comparative study of all available products in the market in order for drawing out the best product presented. To this end, the study examines the key Sharī'ah compliant products offered by different Islamic financial service providers for home financing in the UK and finds as to which product offers the best value for money among its competitors. Design/methodology/approach – The study reviews the various methods available and attempts to justify their use for both parts of the study, part 1 of which deals with the methods used for the review of the key principles of Islamic banking and the surrounding subject area, while the other part discusses the variables and approaches used for the cross-product analysis. The research method adopted is qualitative in nature using first and secondary data. The research of first part of the study is qualitative and is conducted through a literature review. The second part of the study involved collecting primary data with regards to the Islamic housing finance market in the UK. Findings – The research finds that if a prospective applicant for an Islamic housing finance does not have sufficient savings for a high deposit and accepts the Sharī'ah compliance products of Islamic home financing providers in the UK, then the Ahli United Manzil Home purchase programme is the most suitable and competitive; although highest application fees incur. Otherwise the applicant will have to wait until the Sharī'ah compliance products have been thoroughly established. Practical implications – The study recommends that an independent Sharī'ah advisory Board be set up in order to confirm and analyse the Sharī'ah issues pertaining to Islamic home financing, in an objective manner. Originality/value – The paper provides (so far as the authors are aware) the first cross-product analysis of the products offered by Islamic housing finance providers in the UK with the aim of showing the best home financing mode in terms of cost and also the product that complies most with the Sharī'ah
