    Content: We evaluate and compare the effects of a monetary policy shock implemented by one certain tool of the people's bank of China ,say, open market operation, the required reserve ratio and interest rates, through constructing a mixed identification method that combines the pure sign restrictions method with some zero restrictions in a structural VAR model. We find, (i) a shock induced by open market sales or raising the required reserve ratio brings a stronger negative effect on real output, comparing with a shock induced by raising interest rate.(ii) a shock caused by raising interest rate has a bigger probability to bring price a persistent declining course. Our result implies the Chinese authority should give a priority to the instrument of interest rate when trying to tame inflation in the future

    Content: Engineered thick tissues require rapid blood perfusion upon implantation for survival. We have previously described a method to prevascularize (in vitro development of a vascular network) engineered tissues with endothelial cells and mural cells prior to implantation. That strategy has the potential to overcome the limitation of oxygen delivery by diffusion, and thus increase post-implantation survival rate. Pericytes are recruited to facilitate vessel maturation and stabilization during the formation of blood vessels. We hypothesized that the introduction of pericytes into the prevascularized tissue would enhance vessel formation and stimulate host anastomosis. Fibrin tissues were prevascularized by co-culturing human placental pericytes with endothelial colony forming cell-derived endothelial cells (ECFC-EC) from cord blood and normal human lung fibroblasts (NHLF) for 7 or 14 days in vitro. Tissues were then subcutaneously implanted to the dorsal side of SCID mice and retrieved 7 days later. Tissues with a low pericyte-fibroblast ratio developed a vessel network that was well-perfused with host circulation after 7 days in vivo culture. In contrast, pericytes alone or with a high pericyte-fibroblast ratio failed to develop significant in vitro vessel networks, and did not anastomose with the host circulation. Our results suggest that a low pericyte-fibroblast ratio can enhance the in vivo perfusion of engineered tissues. Additionally, we designed and constructed a model to control oxygen diffusion during both in vitro and in vivo culture using biocompatible Poly (methyl methacrylate) (PMMA) and low-density polyethylene (LDPE). Tissues were prevascularized in the devices without limiting oxygen diffusion in vitro for 7 days. Once tissues were implanted into the host, tissue access to oxygen was limited to a small opening facing the host skin side. Oxygen could diffuse through this opening, or be transported by convection following anastomosis with host vasculature. After 7 days, tissues were explanted. Blood perfusion is observed around the access point, but not the entire tissues, which is consistent with blood clotting following anastomosis.

    Content: Arterial stiffness is an independent risk factor for cardiovascular morbidity/mortality. It has been demonstrated that arterial stiffness is linked to arterial fibrosis manifested by increased synthesis of collagen and other extracellular matrix components. Integrins, Transmembrane receptors mediating cell-cell and cell-matrix signaling pathways, are involved in tissue fibrosis. Galectin-3, a novel marker for diagnosis and prognosis of heart failure patients, also plays an important role in fibrosis. However, the molecular mechanisms whereby galectin-3 induces vascular fibrosis are still unclear. We studied the role of integrin αv in Ang II-induced VSMCs arterial fibrosis and stiffness via a SMC specific knock-out of integrin αv mouse model (αv SMKO), induced in adult mice by injection of tamoxifen. We could not find any difference in vascular fibrosis in basal conditions between control and mutant mice. However, decreased arterial fibrosis was observed in αv SMKO mutant mice 28-day after Ang II perfusion. Analysis of RNA from aorta of control and mutant mice by Affymetrix microarrays indicated alteration of TGF-β pathway in Ang II-treated mutant mice. In order to examine the mechanism associated to the decreased fibrosis in VSMCs of αvSMKO mice, we used integrin αv-floxed VSMCs in culture and biochemical methods to analyze the phosphorylation of signaling components and fibrosis-related proteins synthesis following integrin αv inactivation and/or treatment of TGF-β1, Ang-II or galectin-3. Our results indicated that TGF-β1 or Ang-II increased the expression of collagen and fibronectin at the protein level as well as the phosphorylation of ERK and smad2/3 in the control cells, while inactivation of integrin αν partly inhibited the TGF-β1- and Ang-II-induced effects above. Integrin αv was required for Ang II-induced expression of galectin-3 in the VSMCs. Duolink method demonstrated that galectin-3 interacted directly with integrin αv. We also showed that galectin-3 activated AKT, ERK, and Wnt/β-catenin signaling components. The activation of AKT and Wnt/β-catenin signaling pathways, but not ERK signaling pathway, by galectin-3 was inhibited by the knock-down of integrin αv. At cellular level, galectin-3-induced an increase in cell proliferation, migration and synthesis of several fibrosis-related proteins were also significantly inhibited by knock-down of integrin αv. The specific inhibitor of AKT signaling pathway (LY294002) inhibited the activation of downstream Wnt/β-catenin signaling pathway and decreased the response of VSMCs to galectin-3 treatment. Our study indicates a role of integrin αv in the Ang II or TGF-β1 induced arterial fibrogenesis. Galectin-3, interacting with integrin αv, depends on integrin αv/AKT/Wnt/β-catenin signaling pathway to regulate the proliferation, migration and expression of fibrosis-related proteins in VSMCs. ; La rigidité artérielle est un facteur de risque indépendant de la morbidité/mortalité cardiovasculaire. Il a été démontré que la rigidité artérielle est liée à la fibrose artérielle qui se manifeste par une augmentation de la synthèse de collagène et d'autres composants de la matrice extracellulaire. Les intégrines, récepteurs transmembranaires intervenant dans les voies de signalisation cellule-cellule et matrice-cellule, sont impliquées dans la fibrose tissulaire. La galectine-3, un nouveau marqueur pour le diagnostic et le pronostic des patients souffrant d'insuffisance cardiaque, joue également un rôle important dans la fibrose. Cependant, les mécanismes moléculaires par lesquels la galectine-3 induit la fibrose vasculaire ne sont toujours pas élucidés. Nous avons étudié le rôle de l'intégrine αv des CMLV dans la fibrose et la rigidité artérielle induites par l'angiotensine II via une inhibition spécifique dans les CML de l'intégrine αv (αv SMKO), induite chez la souris adulte par injection de tamoxifène. Nous n'avons pas pu trouver de différence dans la fibrose vasculaire entre les conditions basales (souris témoins) et les souris mutantes. Cependant, une diminution de la fibrose artérielle est observée chez les souris mutantes αv SMKO 28 jours après la perfusion d'Ang II. L'analyse de l'expression de l'ARN de l'aorte de souris témoins et de souris mutantes par puces Affymetrix (microarrays) a indiqué une altération de la voie du TGF-ß chez les souris mutantes traitées à l'Ang II. Afin d'examiner le mécanisme associé à la fibrose dans les CMLV de souris αv SMKO, nous avons cultivés des CMLV génétiquement modifiés (intégrine αv floxé) pour analyser la phosphorylation des composants de la signalisation et la synthèse des protéines associées à la fibrose après l'inactivation de l'intégrine αv ou suite au traitement par TGF-β1, Ang-II ou galectine-3. Nos résultats ont indiqué que le traitement par le TGF-β1 ou l'Ang-II augmentaient l'expression du collagène et de la fibronectine au niveau protéique ainsi que la phosphorylation de ERK et smad2 / 3 dans les cellules témoins, tandis que l'inactivation de l'intégrine αv inhibait partiellement les effets du TGF-β1 et de l'Ang-II observés précédemment. L'intégrine αv est requise pour l'expression de la galectine-3 induite par l'Ang II dans les CMLV. La méthode Duolink a démontré que la galectine-3 interagit directement avec l'intégrine αv. Nous avons également montré que galectine-3 active les composants de signalisation AKT, ERK et Wnt/β-caténine. L'activation, par la galectine-3, des voies de signalisation AKT et Wnt/β-caténine, mais pas la voie de signalisation ERK, est inhibée par l'inactivation de l'intégrine αv. Au niveau cellulaire, l'augmentation de la prolifération cellulaire, de la migration et de la synthèse de plusieurs protéines liées à la fibrose induite par la galectine-3 sont également significativement inhibées par l'inactivation de l'intégrine αv. L'inhibiteur spécifique de la voie de signalisation AKT (LY294002) inhibe l'activation de la voie de signalisation Wnt / β-caténine en aval et diminue la réponse des CMLV au traitement par la galectine-3. Notre étude montre un rôle de l'intégrine αv dans la fibrogenèse artérielle induite par l'Ang II ou le TGF-β1. La galectine-3, interagissant avec l'intégrine αv, dépend de la voie de signalisation intégrine αv / AKT / Wnt / β-caténine pour réguler la prolifération, la migration et l'expression des protéines liées à la fibrose dans les CMLV.

    Content: In order to examine the new RMB exchange rate regime rigorously, we employ the STARTZ model to investigate the behavior of RMB NEER from mid-2006 to mid-2008. We find that a managed float with a target central parity and without an explicit band best describes the daily movement of the exchange rate between RMB and a basket of currencies. We also find some peculiar attributes of the RMB NEER such as small conditional variance and stronger effects of government interventions in foreign exchange market

    Content: Abstract: The importance and urgency of IPLE (intellectual property literacy education) are increasingly prominent. This paper aims to build an IPLE mode to provide support for the talent cultivation in university libraries facing opportunities and challenges in the service transformation period. The IPLE mode and content of the Regional Patent Information Centers (PIZ Regional Centers) in German universities and university libraries are studied through network research, literature research, and comparative analysis. Based on the practical experience, educational characteristics and advantages, this paper constructs an IPLE mode of university library that includes six modules. Building the IPLE mode of university libraries can provide reference for university libraries in the work of IPLE, and gradually form an IPLE system that meets the needs of users’ training, conforms to the university’s talent training objectives, and improves the development plan of the university libraries.
