Der Jungschriftsteller Ben Mears (Rob Lowe) kehrt nach Jerusalem's Lot zurück, den Ort, in dem er aufwuchs und den die Einheimischen "Salem's Lot" nennen. Hier machte er die schrecklichste Erfahrung seiner Kindheit. Diese Erinnerung lastet noch immer schwer auf ihm und er möchte an den Ort des Schreckens von damals, das Marsten-Haus zurückkehren und sich seinen Ängsten stellen. Jedoch ist das Haus, das als verflucht gilt und von allen gemieden wird, merkwürdigerweise mittlerweile an einen seltsamen Antiquitäten-Händler (Donald Sutherland) vermietet worden. Und der hat einen mysteriösen Partner, den niemand bisher zu Gesicht bekommen hat. Kurze Zeit darauf beginnt eine seltsame Mordserie, die immer weiter ihre Kreise zieht und sich bald zur Epidemie ausbreitet. Mears und seine Freunde vermuten Vampirismus dahinter und beginnen, während mehr und mehr Bewohner des Ortes sterben oder auf mysteriöse Weise verschwinden, ihren Kampf gegen das Böse. Dreh- und Angelpunkt ist das Marsten-Haus...
