    Inhalt: De diagnose kanker kan een grote invloed hebben op het dagelijks leven van mensen, hun lichamelijke gezondheid en psychisch functioneren. In de laatste decennia zijn verschillende psychologische behandelingen ontwikkeld om mensen te helpen met hun omgang met kanker. In verscheidene gerandomiseerde gecontroleerde onderzoeken (RCTs) is onderzocht of deze interventies effectief zijn in het verbeteren van psychisch functioneren, met een groot aantal studies die aantonen dat psychologische behandelingen mensen met kanker kunnen helpen bij het verbeteren van hun stemming en functioneren. In de meeste van deze onderzoeken is bestudeerd hoe mensen gemiddeld als groep veranderen, ten aanzien van hun omgang met kanker. Daardoor weten we nog weinig over hoe personen van elkaar kunnen verschillen in hun respons op psychologische zorg. Om hier meer inzicht in te geven, was het doel van dit proefschrift om subgroepen van personen te identificeren die verschillende aanpassingspatronen laten zien tijdens het ontvangen van psychologische zorg. Het onderzoek richtte zich zowel op negatieve als positieve gevolgen als voorspellers van aanpassing aan kanker. De resultaten laten duidelijk zien dat mensen met kanker verschillen in hun respons op psychologische zorg. In het bijzonder toonde het onderzoek aan dat een klein maar substantieel aantal mensen ondanks psychologisch zorg klachten als somberheid, angst en vermoeidheid bleef ervaren. De bevindingen suggereren dat zowel naturalistisch onderzoek als interventie onderzoek zich in de toekomst meer zou moeten richten op het bestuderen van subgroepen van mensen met kanker, omdat mensen verschillen in hun omgang met het ziekteproces en verschillend reageren op psychologische zorg. ; A diagnosis of cancer can have a significant effect on people's daily life, their physical health and psychological functioning. During the past decades, different types of psychological interventions have been developed to help people to adapt to cancer, and numerous randomized controlled trials have examined the effectiveness of these interventions in improving people's functioning, suggesting their efficacy in improving patients' mood. The majority of these RCTs mainly examined how cancer patients, as a group, changed with respect to changes in adaptation. Therefore, little is known about how individuals differ from each other in their changes while receiving psychological care. In order to fill in this gap, the current thesis aimed to identify subgroups of people showing distinct patterns of adaptation to cancer over a period of receiving psychological care. We focused on both negative and positive outcomes as well as on predictors of adaptation to cancer. Results clearly confirmed that people with cancer vary in their responses to receiving psychological care. In particular, this research identified a small but substantial number of people who were having difficulties in coping with cancer, even while receiving psychological care. Our findings suggest that future research, both naturalistic and RCT studies, should focus more on the examination of subgroups of people with cancer, as individuals differ from each other in adaptation to the cancer experience and respond differently to receiving psychological care.

    Inhalt: Studies of cell walls have long contributed to understanding plant function. Relevance to the production of biofuels from biomass has recently added urgency to the study of plant cell walls. Arabinogalactan proteins (AGPs) are highly glycosylated glycoproteins at the plant plasma membrane and cell wall. Consisting of a core polypeptide surrounded by glycans, AGPs contain abundant galactosyl and arabinosyl residues and often some glucuronosyl, rhamnosyl, or other residues. Previous study revealed that AGPs in the moss Physcomitrella patens contain unusual 3-O-methyl-rhamnosyl residues (3-O-Me-Rha) in amounts as high as 15 mole percent. The goals of this dissertation were to identify and evaluate Physcomitrella genes that are candidates to encode the rhamnosyl 3-O-methyltransferase that forms 3-O-Me-Rha. A Mycobacteria gene encoding a rhamnosyl 3-O-methyltransferase and an Arabidopsis gene encoding a glucuronosyl 4-O-methyltransferase were used as queries in bioinformatics searches of the Physcomitrella genome. Eleven candidate genes were identified and, through homologous recombination in Physcomitrella , targeted knockouts of these genes were generated. Based on reductions in 3-O-Me-Rha/Rha content ratio in AGPs of the knockouts, two genes, KO1 and KO9 , were selected for further study. A third gene, KO11 , was selected for further study because it shares a domain with the Arabidopsis glucuronosyl 4-O-methyltransferase. Further study of KO1 led to revision of its gene model. A phylogenic tree suggested that the revised KO1 protein contains a LpxB domain, which is characteristic of bacterial lipid A synthesis. The Physcomitrella ko1 knockout exhibited phenotypic effects including abnormal growth in protonema and rhizoids, reduced 3-O-Me-Rha/Rha in AGPs, and reduced lignin-like content in cell walls. Heterologous expression of KO1 did not produce detectable 3-O-Me-Rha in tobacco AGPs. Further bioinformatics study of KO9 showed it to be an ortholog of caffeoyl-CoA O-methyltransferase of lignin biosynthesis. The Physcomitrella ko9 knockout did not exhibit altered lignin-like content in its cell walls. Heterologous expression of KO9 in tobacco did not produce detectable 3-O-Me-Rha in tobacco AGPs. The Physcomitrella ko11 knockout exhibited no effect on 3-O-Me-Rha/Rha in AGPs, and no 3-O-Me-Rha was detected in AGPs from transgenic KO11 tobacco. It remains uncertain whether any of the candidate genes encodes rhamnosyl 3-O-methyltransferase in Physcomitrella .
