Der Abschlussbericht zum obengenannten Projekt beschreibt und bewertet die Ergebnisse zur Untersuchung der S-Lost induzierten Pathomechanismen in humanen Keratinozyten, Lungenepithelzellen und Lungenendothelzellen im Vergleich zu den Schädigungen bei UV-Bestrahlung und Zytostatikabehandlung. Es wird aufgezeigt, dass eine S-Lost Intoxikation von 5 Minuten bis 6 Stunden zu einer zeitabhängigen und konzentrationsabhängigen, sowie zelltypabhängigen Anschaltung von Apoptoseprozessen, einer Bildung von Freien Radikalen, Aktivierung, katalytischer Entkopplung und Expression von Stickstoffmonoxidsynthasen (NOS) und der Aktivierung und Umverteilung des antiapoptotischen Enzyms Akt führt. Die S-Lost-Schädigungsmechansimen lassen sich dabei nicht direkt mit UV- und Zytostatika- induzierten Zellschädigungsmechanismen vergleichen. Die aus den Ergebnissen resultierenden Interventionsmöglichkeiten bei S-Lost Intoxikation werden kurz aufgezeigt. Als Interventionsansatz wurde die Behandlung mit Tetrahydrobiopterin (BH4) zur Aufhebung der katalytischen Entkopplung der eNOS gewählt. Das Potential aber auch die Probleme einer möglichen BH4 Behandlung werden aufgezeigt.(AUT)
