Series Statement: Methods in Molecular Biology, 630
Content: Once a tedious, highly skilled operation, reverse-transcription polymerase chain reaction (RT-PCR) has become a routine and invaluable technique used in most laboratories. In RT-PCR Protocols, Second Edition, expert researchers fully update the technologies presented in the popular previous edition, such as competitive RT-PCR, nested RT-PCR, RT-PCR from single cells, and RT-PCR for cloning. In addition, newer technologies are also explored, including multiplex RT-PCR, RT-LATE-PCR, and the greatly advanced field of real-time quantitative RT-PCR, while recent advances in creating the optimum RT-PCR reaction, e.g. RNA extraction, primer design, and reverse transcription, end the book with their indispensable input. Written in the highly successful Methods in Molecular Biology™ series format, chapters include brief introductions to their respective topics, lists of the necessary materials and reagents, step-by-step, readily reproducible protocols, and notes sections, highlighting tips on troubleshooting and avoiding known pitfalls. User friendly and up-to-date, RT-PCR Protocols, Second Edition acts as a handy companion to scientists from numerous diverse backgrounds who wish to explore further the marvels of gene expression.

Content: RT-PCR, the most widely used technique in biomedical research for analyzing gene expression, has proven a significant boon to the molecular investigation of disease pathogenesis. In RT-PCR Protocols, Joe O'Connell has assembled a panel of highly regarded molecular biologists and clinical researchers to describe in detail their most novel, useful, and interesting RT-PCR applications. Here the newcomer will find readily reproducible protocols for highly sensitive detection and quantification of gene expression, the in situ localization of gene expression in tissue, and the cloning of genes, as well as for analyzing T-cell clones and the differential expression of genes. For the expert seeking to extend the usefulness of RT-PCR, there are user-friendly applications that complement the latest technological advances, including laser-capture microdissection (LCM), real-time and quantitative PCR, microarray technology, cDNA cloning, and antibody engineering. Each protocol emphasizes the technical steps critical for experimental success and includes tips on avoiding potential pitfalls, notes explaining the method's scientific basis and its possible sources of error, and comments on how it might best be accommodated to various experimental circumstances. Diverse and wide-ranging, RT-PCR Protocols offers investigators powerful techniques to be used in the molecular investigation of disease pathogenesis and shows how RT-PCR can complement other technological advances in the design of today's new therapeutic strategies.

Series Statement: Tendencies: Identities, Texts, Cultures : TITC
Content: It is commonly accepted that identity or a sense of self is constructed by and through narrative - the stories we tell ourselves and each other about our lives. This book explores the complex relationships that exist between memory, nostalgia, writing and identity. The author examines a range of autobiographical and first-person fictional texts from holocaust literature, women's writing and popular fiction. Each text foregrounds issues of memory, history and trauma in the construction of identity. There are close readings of texts including Sylvia Fraser's My Father's House, Margaret Atwood's Cats Eye, Barbara Vine's A DarkAdapted Eye, Toni Morrison's Beloved, George Perec's W Or the Memory of Childhood, and Anne Michael's Fugitive Pieces. Reading these texts of memory shows that 'remembering the self' depends not on restoring an original identity, but on 're-membering', on putting past and present selves together, moment by moment, in a process of provisional reconstruction. This is a powerful contribution to the growing field of 'trauma' and holocaust studies. It will be of relevance to those working in the areas of literary and cultural studies, which are witnessing a steady growth of interest in autobiography, theories of narrative, and the relationship between trauma, history and memory.Key FeaturesClose readings of recent fiction and autobiography, including Toni Morrison's Beloved, Anne Michaels' Fugitive Pieces and the popularthrillers of Barbara VineAn interrogation of the models of memory which underpin the constructionof narratives and of identities, and of cultureA focus on meaning as constructed by the readers of the narrative textAn exploration of the myths of 'origin'

Content: The oceanic abyss (depths greater than 3000m), one of the largest environments on the planet, is characterized by absence of solar light, high pressures and remoteness from surface food supply necessitating special molecular, physiological, behavioural and ecological adaptations of organisms that live there. Sampling by trawl, baited hooks and cameras we show that the Chondrichthyes (sharks, rays and chimaeras) are absent from, or very rare in this region. Analysis of a global data set shows a trend of rapid disappearance of chondrichthyan species with depth when compared with bony fishes. Sharks, apparently well adapted to life at high pressures are conspicuous on slopes down to 2000m including scavenging at food falls such as dead whales. We propose that they are excluded from the abyss by high-energy demand, including an oil-rich liver for buoyancy, which cannot be sustained in extreme oligotrophic conditions. Sharks are apparently confined to ca 30% of the total ocean and distribution of many species is fragmented around sea mounts, ocean ridges and ocean margins. All populations are therefore within reach of human fisheries, and there is no hidden reserve of chondrichthyan biomass or biodiversity in the deep sea. Sharks may be more vulnerable to over-exploitation than previously thought.
