Die vorliegende Dissertation untersucht am Beispiel des Enzyms Glucoseoxidase (GOD) die grundlegenden Aspekte der Affinitätstrennung von Glykokonjugaten,insbesondere von Glykoproteinen, also solchen mit Kohlenhydratanteilen. Als Adsorbentien dienen das zuckerbindende Lektin Concanavalin A (ConA) und das mit Polyethylenglykol-Derivaten modifizierte PEGylierte ConA. Deren Herstellung und Eigenschaften werden ausführlich dargestellt. Bei der Ermittlung der optimalen Adsorptionsbedingungen mittels Adsorptionsisothermen, Adsorptionskinetiken und Desorptionskinetiken der unspezifischen Adsorption zeigen beide Proteinliganden mit ihrer hohen Affinität, der niedrigen unspezifischen Adsorption zu GOD und der guten Stabilität eine hervorragende Eignung zu Affinitätstrennprozessen. Als Reaktionsschicht für die Proteinadsorption mittels der bei grobkörnigem Ausgangsmaterial nützlichen Expanded-Bed-Technik dient eine Matrix aus mit Agarose beschichteten Glasperlen.
