Ziel dieserArbeit war, Rheum-Wurzelextrakte auf ihre antifungale Wirksamkeit gegen pilzliche Phytopathogene (Fokus: Echter Mehltau) hinsichtlich der Eignung ihrer aktiven Inhaltsstoffe als Komponenten für eine alternative Pflanzenschutzstrategie zu testen. Basierend auf der analytischen Charakterisierung mittels LC-MS umfasste das mehrstufige Monitoring-system Mycelwachstums- und Blattsegmenttests, histochemische Studien, Antioxidantien-/Protein-Messungen und Transkriptabundanzanalysen sowie Feldversuche. Am effektivsten sowohl gegenüber nekrotrophen und hemibiotrophen Pathogenen als auch obligat biotrophen Pilzen selbst unter Freilandbedingungen enrvies sich ein Extrakt aus Rheum rhabarbarum L. 'The Sutton'. Die Mehltau-Wirkung beruht dabei sowohl auf polyphenolischen Verbindungen der Flavan-3-ol- als auch der Stilben-Gruppe, wobei neben einer direkten antifungalen Aktivität auch induzierte Resistenzmechanismen in den Wirtspflanzen nachgewiesen werden konnten. Formulierte Mischpräparate auf Basis von Rheum-Wurzelextrakten bergen daher ein hohes antifungales Potential.

The aim of this work was to investigate the impact of Rheum root extracts that include antifungal compounds with regard to their utilization as components for an alternative plant protection strategy. Based on chemical structure elucidation by LC-MS, the antifungal efficacy of the extracts against phytopathogens like powdery mildew fungi was tested by using a multistep monitoring system including mycelial growth and leaf segment tests, histochemical observations, enzyme kinetics, transcript abundance analyses and protein assays as well as field trials. Results showed that specific polyphenolic mixtures inhibit mycelial growth of several necrotrophic and hemibiotrophic pathogens. Heterogeneous polyphenols (flavan-3-ols and stilbene agents) were proven as bioactive substances that inhibit biotrophic barley powdery mildew pustule formation, even under field conditions. Certain extracts (Rheum rhabarbarum L. 'The Sutton') showed not only a direct inhibition of pathogen growth, but also some evidence for induced resistance in host plants with a high antifungal potential for developing mixed formulations from these botanicals in the future.
