    Description: The plant-pathogenic bacterium Xanthomonas campestris pv. vesicatoria employs a type III secretion (T3S) system to translocate bacterial effector proteins into eukaryotic host cells. The membrane-spanning secretion apparatus consists of 11 core components and several associated proteins with yet unknown functions. In this study, we analyzed the role of HrpB1, which was previously shown to be essential for T3S and the formation of the extracellular T3S pilus. We provide experimental evidence that HrpB1 localizes to the bacterial periplasm and binds to peptidoglycan, which is in agreement with its predicted structural similarity to the putative peptidoglycan-binding domain of the lytic transglycosylase Slt70 from Escherichia coli. Interaction studies revealed that HrpB1 forms protein complexes and binds to T3S system components, including the inner membrane protein HrcD, the secretin HrcC, the pilus protein HrpE, and the putative inner rod protein HrpB2. The analysis of deletion and point mutant derivatives of HrpB1 led to the identification of amino acid residues that contribute to the interaction of HrpB1 with itself and HrcD and/or to protein function. The finding that HrpB1 and HrpB2 colocalize to the periplasm and both interact with HrcD suggests that they are part of a periplasmic substructure of the T3S system.

    Description: Background and aims: The radial growth of roots largely affects and reorganizes the porous or crack networks of soils and substrates. We studied the consequences of a radial steric constriction on the root growth and the feedback force developed by the root on the solid phase. Methods: We developed an original method of photoelasticity to measure in situ root forces. By changing the gap width (0.5 to 2.3 mm) between two photoelastic disks we applied variable radial constrictions to root growth and simultaneously measured the corresponding radial forces. Changes in morphology and forces of primary roots of chick pea (Cicer arietinum L.) seedlings were recorded by time-lapse imaging every 24 min up to 5 days. Results: The probability of root entering the gap depended on the gap size but was also affected by circumnutation. Compared to non-constrained root controls, no significant morphological change (elongation, diameter) was measured outside the gap zone. Inside the gap zone, outer cortex cells were compressed, the central cylinder was unaffected. Radial forces were increasing with time but no force levelling was observed even after 5 days. Conclusions: Radial constrictions applied to roots did not significantly reduce their growth. The radial force was related to the root strain in the gap.

    Description: Activated factor X (FXa) is an important player in the coagulation cascade responsible for thrombin generation, which is activated during atrial fibrillation. Increasing evidence suggests that FXa influences cell signalling in various cell types by activating protease-activated receptors (PARs). It is so far not known if molecular effects of FXa affect atrial signal transduction. To study the effects of FXa, human atrial tissue slices were cultivated with FXa up to 24 h. Additionally, rapid pacing was applied at 4 Hz to resemble atrial fibrillation. The inhibitory impact of FXa antagonist (Rivaroxaban), protease-activated receptor 1 antagonist (SCH79797), and protease-activated receptor 2 antagonist (GB83) were analysed under experimental conditions. The exposure of atrial tissue to FXa resulted in the 1.7 fold upregulation of PAR2-mRNA, activation of MAP kinases (ERK1/2) and NF-κB signalling. Furthermore FXa increased the expression of adhesion molecule ICAM-1 (1.82±0.20), chemokine IL-8 (1.94±0.20), as well as prothrombotic molecule PAI-1 (1.52±0.17). The combination of rapid pacing and FXa caused significant upregulation of PAR1 (2.82±0.22), PAR2 (2.66±0.40), ICAM-1 (2.13±0.25), IL-8 (2.22±0.24), LOX-1 (2.59±0.35), and PAI-1 (2.65±0.52) at the mRNA level. Rivaroxaban and GB83 prevented upregulation of PARs, ICAM-1, LOX-1, IL-8, and activation of MAP kinases. The elevation in the expression of PAI-1 was hindered in the presence of SCH79797, or Rivaroxaban. The present study indicates that FXa mediates inflammatory signalling in atrial tissue. Importantly, FXa and tachyarrhythmia act synergistically to increase expression of protease-activated receptors and inflammatory mediators. Rivaroxaban prevented effectively FXa-induced molecular effects in human atrial tissue particularly during rapid pacing.

    Description: To evaluate the correlation between the 1993 and 2000/2007 World Health Organization (WHO) classification with the outcome in patients with high-grade meningiomas. Between 1985 and 2004, 73 patients diagnosed with atypical or anaplastic meningiomas were treated with radiotherapy. Sections from the paraffin-embedded tumor material from 66 patients (90%) from 13 different pathology departments were re-evaluated according to the first revised WHO classification from 1993 and the revised classifications from 2000/2007. In 4 cases, the initial diagnosis meningioma was not reproducible (5%). Therefore, 62 patients with meningiomas were analyzed. All 62 tumors were reclassified according to the 1993 and 2000/2007 WHO classification systems. Using the 1993 system, 7 patients were diagnosed with WHO grade I meningioma (11%), 23 with WHO grade II (37%), and 32 with WHO grade III meningioma (52%). After scoring using the 2000/2007 system, we found 17 WHO grade I meningiomas (27%), 32 WHO grade II meningiomas (52%), and 13 WHO grade III meningiomas (21%). According to the 1993 classification, the difference in overall survival was not statistically significant among the histologic subgroups ( = .96). Using the 2000/2007 WHO classifications, the difference in overall survival became significant ( = .02). Of the 62 reclassified patients 29 developed tumor progression (47%). No difference in progression-free survival was observed among the histologic subgroups ( = .44). After grading according to the 2000/2007 WHO classifications, significant differences in progression-free survival were observed among the three histologic groups ( = .005). The new 2000/2007 WHO classification for meningiomas showed an improved correlation between the histologic grade and outcome. This classification therefore provides a useful basis to determine the postoperative indication for radiotherapy. According to our results, a comparison of the published data for future treatment decision-making remains difficult when the histologic diagnosis has not been based on the new improved classification system.

    Description: Die im vorliegenden Beitrag besprochene Studie untersucht die Interaktionen zwischen exterozeptiven und interozeptiven Afferenzen. Als eines der Ergebnisse zeigte sich, dass die Inselregion im Gehirn aus beiden Reizkategorien nicht nur eine Körperrepräsentation, sondern offensichtlich auch eine Art Raumrepräsentation (“peripersonal space”) generiert. Als Nebenergebnis zeigte sich, dass die vegetative Kontrolle einzelner Körperbereiche davon abzuhängen schien, inwieweit die Körperrepräsentation in die dazugehörige Raumrepräsentation projiziert wurde. Ziel der Behandlung nicht strukturell bedingter vegetativer Dysfunktionen ist demnach die Verbesserung der zentralen Integration extero- und interozeptiver Reize, d.h. der Körperwahrnehmung als Ganzes. Da es sich um interne Integrationsprozesse handelt, die direkten Techniken nicht zugänglich sind, können Behandlungsansätze maximal die Rahmenbedingungen für die besagten Prozesse verbessern. Hier empfehlen sich v.a. sanfte und indirekte Techniken der salutogenetisch orientierten Osteopathie. The reviewed study presented in this article analyzes the interactions between exteroceptive and interoceptive afferences. The results show that the lobus insularis of the brain generates from both categories of stimuli not only a body representation but apparently also a kind of space representation (peripersonal space). As additional result the vegetative control of single body parts seems to depend on how the body representation is projected within the associated space representation. The aim of treating non structural vegetative dysfunctions is to improve the central integration of extero- an interoceptive stimuli, meaning the body awareness as a whole. Since these intern integration processes are not accessible through direct techniques the treatment approach can at most improve the basic conditions of theses processes. Mainly smooth and non direct techniques of the salutogenetic oriented osteopathy are recommended.
