    Description: Reduced apoptosis is associated to cancer development. Agents able to restore the programmed cell death responsiveness of cancer cells are foreseen as potential effective cancer therapies. In this study, we report that a glutathione-S-derivative, S-acetyl-glutathione (Sag), induces significant apoptosis in three human lymphoma cell lines, including Daudi, Raji and Jurkat cells while it had no or little effect on either Hut-78 lymphoma cells or the normal BT lymphocytes. We used Annexin-V FACS analysis and DNA laddering to demonstrate that Sag activated apoptosis in the three sensitive cell lines in a dose- and time-dependent-fashion. Using mercury orange staining and FACS analysis, we showed that Sag generated an intracellular GSH depletion in Daudi, Raji and Jurkat cells but not in Hut-78 or the normal BT cells. These data provide direct evidence that Sag specifically activates programmed cell death in lymphoma cells through, at least in part, a depletion of intracellular GSH rather than an increase, as previously suggested. Because of its selective effect on cancer cells, Sag appears as a promising new lymphoma cell apoptosis inducer with potential clinical value for lymphoma patients.

    Description: Negative correlation between serum IgE levels and production of IFN-gamma by lymphocytes and positive correlation between serum IgE levels and production of IL-4 by lymphocytes was detected in 12 children with allergic asthma and recurrent respiratory diseases. Deficiency of reduced glutathione in whole blood and some disorders in phagocytic and oxidative burst activity of monocytes were observed in these children. Use of reduced glutathione, L-cysteine and anthocyane (Recancostat, Clear Vision, Switzerland) resulted in elevation of IFN-gamma production, lymphocyte response to mitogens, NK cell activity, increase in percentage of naive CD4(+) T lymphocytes (refreshment effect) and improvement of clinical status. Positive clinical results were lasted during 6 months.

    Description: The possibility of fabricating SiC ceramics using special wood-based materials has been shown in previous research. To form accurate SiC bodies from wood-based green templates, special shaping techniques are necessary to fulfill the requirements on the finished ceramic products. Due to the fact that wood-based green templates undergo shrinkage during carbonization, no suitable forming processes for the wood-based green templates have been presented so far. In this publication different raw materials and 3D shaping production processes and their influence on green body properties are described. It was possible to produce green bodies with a MOR higher than 15 N/mm² and a density in the range above 0.82 g/cm³. It is shown that reduction of resin content decreases the strength significantly and a reduction to 30 mass % resin content reduces the MOR of flat pressed panels to the defined limit. The extruded samples showed a significantly lower strength (mean 18.1 N/mm², standard deviation 8.6 N/mm²). Injection molding was not applied successfully. The findings show that shape pressing and extrusion were applied successfully, but the green body properties differed significantly from the results achieved by flat pressing. Vorhergehende wissenschaftliche Untersuchungen haben bereits die Möglichkeit der Herstellung von SiC-Keramiken aus speziellen Holzwerkstoffen dargestellt und dabei durch spezielle holzbasierte Grünkörper vergleichbare Eigenschaftsprofile von herkömmlichen Hochleistungskeramiken erreicht (z. B. Sinterkeramiken). Um holzwerkstoffbasierte, maßstabsgetreue SiC-Körper herstellen zu können, die die Anforderungen von keramischen Produkten erfüllen, sind jedoch spezielle Formgebungsverfahren erforderlich. Auf Grund der Tatsache, dass holzbasierte Grünkörper während des Carbonisierungsprozesses einer Schrumpfung unterliegen, konnten aber bisher noch keine geeigneten Verfahren zur Umformung von komplex geformten holzwerkstoffbasierten Grünkörpern dargestellt werden. In dieser Publikation werden verschiedene Ausgangsrohstoffe und 3D-Gestaltungs-/Produktionsprozesse und deren Einfluss auf die Grünkörpereigenschaften beschrieben. Es konnten Grünkörper mit einem MOR höher als 15 N/mm² und einer Dichte von über 0,82 g/cm³ hergestellt werden. Des Weiteren konnte ein deutlicher Einfluss des Harzgehaltes auf die Festigkeit nachgewiesen werden, so dass sich bei einer Reduktion des Harzgehaltes auf 30 Masse- % der MOR von im Flachpressverfahren hergestellten Platten bis zum Grenzwert verringerte. Extrudierte Probekörper zeigten signifikant geringere Festigkeitswerte (Mittelwert 18.1 N/mm², Standardabweichung 8.6 N/mm²). Das Spritzgussverfahren konnte nicht erfolgreich angewendet werden. Die Ergebnisse zeigen jedoch, dass sich das Formpressen und die Extrusion erfolgreich anwenden lassen, dabei aber deutliche Abweichungen der Grünkörpereigenschaften zum Flachpressverfahren vorliegen.

    Description: Intracellular glutathione (GSH) plays an important regulatory role in the host response to viral infections. Replenishment of intracellular GSH is a desirable yet challenging goal, since systemic GSH supplementation is rather inefficient due to a short half-life of GSH in blood plasma. Further, GSH is not taken up by cells directly, but needs to be broken down into amino acids and resynthesized to GSH intracellularly, this process often being impaired during viral infections. These obstacles may be overcome by a novel glutathione derivative S-acetylglutathione (S-GSH), which is more stable in plasma and taken up directly by cells with subsequent conversion to GSH. In the present study, in vitro effects of supplementation with S-GSH or GSH on intracellular GSH levels, cell survival and replication of human herpes simplex virus type 1 (HSV-1) were studied in human foreskin fibroblasts. In addition, in vivo effects of supplementation with S-GSH or GSH on HSV-1-induced mortality were studied in hr/hr mice. In cell culture, viral infection resulted in a significant decrease of intracellular GSH levels. S-GSH efficiently and dose-dependently (5 and 10 mM tested) restored intracellular GSH, and this replenishment was more efficient than with GSH supplementation. In mice, S-GSH, but not GSH, significantly decreased HSV-1-induced mortality ( P 〈0.05). The data suggest that S-GSH is a suitable antiviral agent against HSV-1 both in vitro and in vivo, indicating that this drug may be of benefit in the adjunctive therapy of HSV-1 infections.

    Description: The effect of reduced glutathione (GSH) and S-acetylglutathione (S-acglu) treatment on several tumor cell lines and normal cells in vitro was investigated. GSH and S-acglu applied at concentrations of 1 mM and 2 mM induced apoptosis in malignant cells as shown by DNA-fragmentation and staining of apoptotic cells with 7-amino-actinomycin D while viability and growth of normal cells were not significantly influenced by this treatment. The results demonstrated that GSH and S-acglu may be selective inducers of apoptosis in malignant cells.
