    Description: Cellular RNAs that do not function as messenger RNAs (mRNAs), transfer RNAs (tRNAs) or ribosomal RNAs (rRNAs) comprise a diverse class of molecules that are commonly referred to as non-protein-coding RNAs (ncRNAs). These molecules have been known for quite a while, but their importance was not fully appreciated until recent genome-wide searches discovered thousands of these molecules and their genes in a variety of model organisms. Some of these screens were based on biocomputational prediction of ncRNA candidates within entire genomes of model organisms. Alternatively, direct biochemical isolation of expressed ncRNAs from cells, tissues or entire organisms has been shown to be a powerful approach to identify ncRNAs both at the level of individual molecules and at a global scale. In this review, we will survey several such wet-lab strategies, i.e. direct sequencing of ncRNAs, shotgun cloning of small-sized ncRNAs (cDNA libraries), microarray analysis and genomic SELEX to identify novel ncRNAs, and discuss the advantages and limits of these approaches.

    Description: The bacterial envelope stress response (ESR) is triggered by the accumulation of misfolded outer membrane proteins (OMPs) upon envelope damage or excessive OMP synthesis, and is mediated by the alternative sigma factor, σ. Activation of the σ pathway causes a rapid downregulation of major mRNAs, which prevents further build‐up of unassembled OMPs and liberates the translocation and folding apparatus under conditions that require envelope remodelling. The factors that facilitate the rapid removal of the unusually stable mRNAs in the ESR were previously unknown. We report that in the ESR relies upon two highly conserved, σ‐controlled small non‐coding RNAs, RybB and MicA. By using a transcriptomic approach and kinetic analyses of target mRNA decay , RybB was identified as the factor that selectively accelerates the decay of multiple major mRNAs upon induction of the ESR, while MicA is proposed to facilitate rapid decay of the single mRNA. In unstressed bacterial cells, the two σ‐dependent small RNAs function within a surveillance loop to maintain envelope homeostasis and to achieve autoregulation of σ.

    Description: Recent systematic genome searches revealed that bacteria encode a tremendous number of small non-coding RNAs (sRNAs). Whereas most of these molecules remain of unknown function, it has become increasingly clear that many of them will act to modulate gene expression at the post-transcriptional level. Where studied in more detail, sRNAs have often been found to control the expression of outer membrane proteins (OMPs). Enterobacteria such as and are now known to encode at least eight OMP-regulating sRNAs (InvR, MicA, MicC, MicF, OmrAB, RseX and RybB). These sRNAs exert their functions under a variety of growth and stress conditions, including the σ -mediated envelope stress response. An sRNA–OMP network is emerging in which some sRNAs act specifically on a single mRNA, whereas others control multiple mRNA targets.

    Description: Motor dysfunction is an important clinical finding in patients with liver cirrhosis and mild forms of hepatic encephalopathy. The mechanisms and clinical appearance of motor impairment in patients with liver cirrhosis are not completely understood. We studied fine motor control in forty four patients with advanced liver cirrhosis (excluding those with hepatic encephalopathy grade II) and 48 healthy controls using a kinematic analysis of standardized handwriting tests. We analysed parameters of velocity, the ability to coordinate and the level of automatisation of handwriting movements. Furthermore, we studied the association between impairment of handwriting and clinical neuro-psychiatric symptoms. As compared with control subjects, patients showed a statistically significant reduction of movement peak velocity in all handwriting tasks as well as a substantial increase of number of velocity inversions per stroke. Using a z-score based assessment we found impairment of handwriting in fourteen out of forty four patients (31.8 %). The deterioration of handwriting was associated with clinical symptoms of motor dysfunction, such as bradykinesia, adiadochokinesia, dysmetria of upper extremities and gait ataxia. This is the first study that quantitatively investigates impairment of handwriting in patients with liver cirrhosis. Our findings suggest the application of kinematic analysis of handwriting for diagnostics of motor dysfunction in patients with mild forms of hepatic encephalopathy.

    Description: A federation approach for security in future distributed service delivery platforms for mobile users offers some key advantages over an integrated solution relying on a common choice of a standardized authentication technology. By agreeing on an exchange protocol for security assertions rather than on the detailed security mechanisms, Service Aggregators and Access Network Operators will be able to federate their customer offerings flexibly and jointly offer services. The consumer will find formerly separate offerings combined, and Service Operators will enjoy open interfaces towards the network's service delivery platform. Through the use of Security Assertion Markup Language (SAML), standardized assertion statements can be made not only for the user's identity, but also on attributes and authorizations associated with it. This will allow a seamless personalized service experience offering single sign-on across separate operational domains. An example from automobile telematics is used to illustrate the concepts.

    Description: A classical transport experiment was performed in a field plot of 2.5 m2 using the dye tracer brilliant blue. The measured tracer distribution demonstrates the dominant role of the heterogeneous soil structure for solute transport. As with many other published experiments, this evidences the need of considering the macroscopic structure of soil to predict flow and transport. We combine three different approaches to represent the relevant structure of the specific situation of our experiment: i) direct measurement, ii) statistical description of heterogeneities and iii) a conceptual model of structure formation. The structure of soil layers was directly obtained from serial sections in the field. The sub-scale heterogeneity within the soil horizons was modelled through correlated random fields with estimated correlation lengths and anisotropy. Earthworm burrows played a dominant role at the transition between the upper soil horizon and the subsoil. A model based on percolation theory is introduced that mimics the geometry of earthworm burrow systems. The hydraulic material properties of the different structural units were obtained by direct measurements where available and by a best estimate otherwise. From the hydraulic structure, the 3-dimensional velocity field of water was calculated by solving Richards' Equation and solute transport was simulated. The simulated tracer distribution compares reasonably well with the experimental data. We conclude that a rough representation of the structure and a rough representation of the hydraulic properties might be sufficient to predict flow and transport, but both elements are definitely required.
