New anti-inflammatory sterols from a gorgonian Pinnigorgia sp (2016)

Description: Chemical investigation on the EtOAc-soluble fraction from the MeOH/DCM extract of a gorgonian sp. afforded two new sterols, 11-acetoxy-24 -methyl-3 ,5 ,6 -trihydroxy-9,11-secocholest-7-en-9-one ( ) and 5 ,6 -epoxy-(22 ,24 )-ergosta-8,22-diene-3 ,7 -diol ( ). The structures of sterols and were elucidated on the basis of spectroscopic analysis and by comparison of their spectroscopic data with those of related analogues. Both and were shown to significantly inhibit the accumulation of the pro-inflammatory iNOS and COX-2 protein in LPS-stimulated RAW264.7 macrophage cells.

Description: Pinnigorgiols A–C ( – ), three novel 9,11-secosterols with a rare carbon skeleton arrangement, were isolated from a gorgonian coral identified as sp. Compounds – possess novel carbon skeletons, which include a tricyclo[5,2,1,1]decane ring. The structures of – were established on the basis of spectroscopic methods. The novel compounds – were tested for in vitro cytotoxicity in hepatic stellate cells (HSCs) and displayed inhibitory effects on the generation of superoxide anions and the release of elastase by human neutrophils. Proliferation of HSCs plays a key role in the pathogenesis of liver fibrosis.

Description: Two novel 16-membered tetraene marcolides, bafilomycins N ( ) and O ( ), along with two known analogues, JBIR-100 ( ) and bafilomycin K ( ) were produced from isolate GIC10-1, a sp. strain that was originally cloned from bacterial communities associated with marine sponge sp. The structures of new bafilomycins and were established by using spectroscopic methods and comparing the spectroscopic data with those of known related metabolites. Compounds and were proven to be the first 14-methylbafilomycins to be isolated. Cytotoxicity of these compounds toward various cancer cell lines also is described.

Description: Eight new briarane diterpenoids, designated briarenolides M–T ( – ), were isolated from an octocoral identified as sp. The structures of briaranes – were established by spectroscopic methods and by comparison of the spectroscopic data with those of known analogues. The relationship between H and C NMR chemical shifts of 2-hydroxybriaranes possessing of (A) a Δ -conjugated diene moiety or (B) a Δ -conjugated diene moiety are described. Briarenolides M ( ), P ( ), S ( ), and T ( ) were found to inhibit the accumulation of the pro-inflammatory inducible nitric oxide synthase (iNOS) protein and briarenolides N ( ), P ( ), and T ( ) were found to inhibit the accumulation of the pro-inflammatory cyclooxygenase-2 (COX-2) protein in LPS (lipopolysaccharide)-stimulated RAW264.7 macrophage cells.

Description: A new 16-membered diene macrolide, bafilomycin M ( ), was produced from a sp. GIC10-1, which was originally isolated from a marine sponge sp. The structure of was established by spectroscopic methods and this compound was found to exhibit cytotoxicity toward K-562, HL-60, SUPT-1, and LNCaP tumor cells with IC values 0.060, 0.011, 0.047, and 0.389 μg/mL, respectively.

Description: Two new eunicellin-type diterpenoids, cladieunicellins R ( ) and S ( ), were isolated from the octocoral and their structures were elucidated by spectroscopic methods. Eunicellin was proven to be the first 2,6-hemiketal eunicellin and was found to exhibit moderate cytotoxicity toward the MOLT-4, K562, and SUP-T1 cells.

Description: A novel 6,12-dichlorobriarane diterpenoid, briarenolide J ( ), was isolated from an octocoral identified as sp. The structure of briarane was elucidated by spectroscopic methods. Briarenolide J ( ) is the first metabolite of a briarane-related natural product found to possess a chlorine atom at C-12 and this compound was found to display inhibitory effects on the generation of superoxide anions and the release of elastase by human neutrophils with IC values of 14.98 and 9.96 μM, respectively.

Description: Three natural clovane-related sesquiterpenoids, 2beta-acetoxyclovan-9alpha-ol (1), 9alpha-acetoxyclovan-2beta-ol (2) and clovan-2beta,9beta-diol (3), were isolated from the gorgonian coral Rumphella antipathies. The structures of clovanes 1-3 were elucidated by spectroscopic methods and by comparison of the spectral data with those of known clovane analogues. This is the first time that clovanes 1-3 have been isolated from a natural source. Clovanes 1 and 2 displayed inhibitory effects on the generation of superoxide anions and the release of elastase by human neutrophils.
