    Description: An efficient method for the synthesis of flavones and γ‐benzopyranones has been developed utilizing a mild Sonogashira coupling and 18‐crown‐6 ether mediated 6‐ cyclization of ‐alkynoylphenyl acetates. By using this strategy, flavones and γ‐benzopyranones bearing electron‐donating groups, halogens, and simple alkyl substituents were synthesized in satisfactory yields. A facile, mild, and selective method for the synthesis of flavones and γ‐benzopyranones is reported. ‐Alkynoylphenyl acetates were obtained from either acyl chloride derivatives or substituted salicylates. Upon removing the acetate group, flavones and γ‐benzopyranones were synthesized within 15 min with the aid of 18‐crown‐6 ether. A library of flavones and γ‐benzopyranones was established.

    Description: : NMR spectroscopy and isothermal titration calorimetry show that calcium ions are chelated by the hydroxylated THF ring of acetogenins, which results in formation of complexes that aid the Ca cations in penetrating cell membranes and in elevating the intracellular calcium level (see picture). This disruption of intracellular calcium homeostasis induces mitochondrial depolarization and mediates cell toxicity.

    Description: Copper catalysed oxidative coupling reaction of formamides with β‐keto esters and 2‐carbonyl‐substituted phenols successfully proceeded through direct C–H bond activation of formamides. The corresponding carbamates were formed with high stereoselectivity under mild reaction conditions. This protocol was successfully applied to the synthesis of three novel halogenated carbamates and a carbaryl insecticide derivative. Our results suggest the use of 6.0 equiv. TBHP is crucial for this type of reaction. Synthesis of carbamates was accomplished through the reaction of β‐keto esters or 2‐carbonyl‐substituted phenols with formamides using copper chloride and an oxidant. Sixteen aliphatic as well as fourteen aromatic carbamates were prepared in good to excellent yields. The methodology was applied to the synthesis of halogenated carbamates and a carbaryl insecticide derivative.

    Description: Hepatic ischemia‐reperfusion (IR) injury is a common clinical problem and ROS may be a contributing factor on IR injury. The current study evaluates the potential protective effect of saffron ethanol extract (SEE) in a rat model upon hepatic IR injury. Caspases 3 and terminal deoxynucleotidyl transferase‐mediated dUTP biotin nick end labeling (TUNEL) results showed increased cell death in the IR samples; reversely, minor apoptosis was detected in the SEE/IR group. Pretreatment with SEE significantly restored the content of antioxidant enzymes (SOD1 and catalase) and remarkably inhibited the intracellular ROS concentration in terms of reducing p47phox translocation. Proteome tools revealed that 20 proteins were significantly modulated in protein intensity between IR and SEE/IR groups. Particularly, SEE administration could attenuate the carbonylation level of several chaperone proteins. Network analysis suggested that saffron extract could alleviate IR‐induced ER stress and protein ubiquitination, which finally lead to cell apoptosis. Taken together, SEE could reduce hepatic IR injury through modulating protein oxidation and our results might help to develop novel therapeutic strategies against ROS‐caused diseases.

    Description: The selective synthesis of various dideuterochalcones as potentially bioactive deuterium‐labeled products is presented, by means of the highly controlled partial deuteration of antidiabetic chalcone derivatives. The benefits of continuous‐flow processing in combination with on‐demand electrolytic D gas generation has been exploited to avoid over‐reaction to undesired side products and to achieve selective deuterium addition to the carbon–carbon double bond of the starting enones without the need for unconventional catalysts or expensive special reagents. The roles of pressure, temperature, and residence time proved crucial for the fine‐tuning of the sensitive balance between the product selectivity and the reaction rate. The presented flow‐chemistry‐based deuteration technique lacks most of the drawbacks of the classical batch methods, and is convenient, time‐ and cost‐efficient, and safe. : Various dideuterochalcones have been synthesized selectively by means of the highly controlled partial deuteration of antidiabetic chalcone derivatives. The benefits of continuous‐flow processing were exploited in combination with on‐demand electrolytic generation of D gas (see figure) to avoid over‐reaction to undesired side products and to achieve selective deuterium incorporation without the need for special reagents.
