    Kurzfassung: Wire and arc additively manufactured (WAAM) parts and structures often present internal defects, such as gas pores, and cause irregularities in the manufacturing process. In order to describe and assess the effect of internal defects in fatigue design, this research study investigates the fatigue strength of wire arc additive manufactured structures covering the influence of imperfections, particularly gas pores. Single pass WAAM structures are manufactured using titanium alloy Ti-6Al-4V and round fatigue, tensile specimen are extracted. Tensile tests and uniaxial fatigue tests with a load stress ratio of R = 0.1 were carried out, whereby fatigue test results are used for further assessments. An extensive fractographic and metallographic fracture surface analysis is utilized to characterize and measure crack-initiating defects. As surface pores as well as bulk pores are detected, a stress intensity equivalent ∆Keqv transformation approach is presented in this study. Thereby, the defect size of the surface pore is transformed to an increased defect size, which is equivalent to a bulk pore. Subsequently, the fatigue strength assessment method by Tiryakioğlu, commonly used for casting processes, is applied. For this method, a cumulative Gumbel extreme value distribution is utilized to statistically describe the defect size. The fitted distribution with modified data reveals a better agreement with the experimental data than unmodified. Additionally, the validation of the model shows that the usage of the ∆K modified data demonstrates better results, with a slight underestimation of up to about −7%, compared to unmodified data, with an overestimation of up to about 14%, comparing the number of load cycles until failure. Hence, the presented approach applying a stress intensity equivalent transformation of surface to bulk pores facilitates a sound fatigue strength assessment of WAAM Ti-6Al-4V structures.

    Kurzfassung: Frequency response (FR) analysis allows the characterization of gas diffusion occurring within a porous solid system. The shape of the pressure response curves obtained after a volume modulation in the reactor gives essential information about the gas adsorption and desorption properties of the porous material, e.g., zeolites, which is in contact with a certain gas environment, as well as information about the transport phenomena such as diffusion. In this work, a simulation model developed in COMSOL Multiphysics® is introduced to reproduce the experimental behavior of the tested solid/gas systems. This approach covers, for the first time, a coupling of computational fluid dynamics (CFD), porous media flow, and a customized mass adsorption/desorption function to simulate the behavior of real frequency response systems. The simulation results are compared to experimental data obtained from the interaction of propane in MFI zeolites as well as additional data from the literature to evaluate the model validity. Furthermore, a small variation study of the effect of simulation parameters such as the mass of the sample, bed porosity, or geometry is performed and analyzed. The essential advantage of this model with respect to other analytical approaches is to observe the spatial pressure and adsorption distribution (along with other local effects) of the gas within the porous material. Thus, local environments can be visualized, and non-idealities can, therefore, be detected in contrast to the general integral simulation approach.

    Kurzfassung: Transient uptake curves for propane gas in a bed of HZSM-5 using a volumetric frequency response setup (batch system) were obtained. Thereby, a perturbation, such as a change in volume, was applied to the solid/gas system, and the resulting change in pressure was detected. Two cases of mass transfer limitations (bed diffusion control and micropore diffusion control) were compared, and it was concluded that, in the presented case, micropore diffusion is the rate-determining process. The obtained micropore diffusion coefficient for propane in HZSM-5 was, on average, about 1.2 × 10−10 m2 g−1, which is in good agreement with other frequency response studies shown by literature data. The homemade setup and the modeling presented in this work serve as the basis for ongoing numerical simulations.

    Kurzfassung: We developed a new mobile ultrasound device for long-term and automated bladder monitoring without user interaction consisting of 32 transmit and receive electronics as well as a 32-element phased array 3 MHz transducer. The device architecture is based on data digitization and rapid transfer to a consumer electronics device (e.g., a tablet) for signal reconstruction (e.g., by means of plane wave compounding algorithms) and further image processing. All reconstruction algorithms are implemented in the GPU, allowing real-time reconstruction and imaging. The system and the beamforming algorithms were evaluated with respect to the imaging performance on standard sonographical phantoms (CIRS multipurpose ultrasound phantom) by analyzing the resolution, the SNR and the CNR. Furthermore, ML-based segmentation algorithms were developed and assessed with respect to their ability to reliably segment human bladders with different filling levels. A corresponding CNN was trained with 253 B-mode data sets and 20 B-mode images were evaluated. The quantitative and qualitative results of the bladder segmentation are presented and compared to the ground truth obtained by manual segmentation.

    Kurzfassung: Definitive radiochemotherapy of locally advanced head and neck squamous cell cancer (HNSCC) achieves high locoregional tumor control rates; but is frequently associated with long-term toxicity. A future direction could be a de-escalation strategy focusing on treated volume rather than radiotherapy dose. This analysis evaluates radiotherapy dose and volume parameters of patients treated with a standard contouring approach in a clinical trial context compared with a revised volume-reduced contouring approach. In this case, 30 consecutive patients from the CheckRad-CD8 trial treated at a single study center were included in this analysis. Treatment toxicity and quality of life were assessed at the end of radiotherapy. Standard treatment plans (ST) following state of the art contouring guidelines that were used for patient treatment and volume reduced treatment plans (VRT) according to a revised simulated approach were calculated for each patient. Planning target volumes (PTV) and mean doses to 38 organs-at-risk structures were compared. At the end of radiotherapy patients reported high rates of mucositis; dysphagia and xerostomia. In addition; patient reported quality of life as assessed by the EORTC QLQ-HN35 questionnaire deteriorated. Comparing the two contouring approaches; the elective PTV_56 Gy and the high risk PTV_63 Gy (shrinking field) were significantly smaller in the VRT group. Significant reduction of mean dose to structures of the oral cavity; the larynx as well as part of the swallowing muscles and the submandibular glands was achieved in the simulated VRT-plan. Treatment de-intensification by reduction of the irradiated volume could potentially reduce treatment volume and mean doses to organs at risk. The proposed contouring approach should be studied further in the context of a clinical trial.

    Kurzfassung: Background and Aims: Pulmonary Hypertension (PH) represents an aetiologically and clinically heterogeneous disorder accompanied by a severely impaired prognosis. Key steps of PH pathogenesis are vascular and right ventricular myocardial remodelling entailing the re-occurrence of fetal variants of the cell adhesion modulating protein fibronectin (Fn) being virtually absent in healthy adult tissues. These variants are liberated into circulation and are therefore qualified as excellent novel serum biomarkers. Moreover, these molecules might serve as promising therapeutic targets. The current study was aimed at quantifying the serum levels of two functionally important fetal Fn variants (ED-A+ and ED-B+ Fn) in patients suffering from PH due to different aetiologies compared to healthy controls. Methods: Serum levels of ED-A+ and ED-B+ Fn were quantified using novel ELISA protocols established and validated in our group in 80 PH patients and 40 controls. Results were analysed with respect to clinical, laboratory, echocardiographic and functional parameters. Results: Serum levels of ED-A+ Fn (p = 0.001) but not ED-B+ Fn (p = 0.722) were significantly increased in PH patients compared to healthy controls. Thus, the following analyses were performed only for ED-A+ Fn. When dividing PH patients into different aetiological groups according to current ESC guidelines, the increase in ED-A+ Fn in PH patients compared to controls remained significant for group 1 (p = 0.032), 2 (p = 0.007) and 3 (p = 0.001) but not for group 4 (p = 0.156). Correlation analysis revealed a significant relation between ED-A+ Fn and brain natriuretic peptide (BNP) (r = 0.310; p = 0.002), six minutes’ walk test (r = −0.275; p = 0.02) and systolic pulmonary artery pressure (PAPsys) (r = 0.364; p 〈 0.001). By logistic regression analysis (backward elimination WALD) including a variety of potentially relevant patients’ characteristics, only chronic kidney disease (CKD) (OR: 8.866; CI: 1.779–44.187; p = 0.008), C reactive protein (CRP) (OR: 1.194; CI: 1.011–1.410; p = 0.037) and ED-A+ Fn (OR: 1.045; CI: 1.011–1.080; p = 0.009) could be identified as independent predictors of the presence of PH. Conclusions: Against the background of our results, ED-A+ Fn could serve as a promising novel biomarker of PH with potential value for initial diagnosis and aetiological differentiation. Moreover, it might contribute to more precise risk stratification of PH patients. Beyond that, the future role of ED-A+ Fn as a therapeutic target has to be evaluated in further studies.

    Kurzfassung: The measurement of acoustic emission data in experiments reveals informative details about the tribological contact. The required recording rate for conclusive datasets ranges up to several megahertz. Typically, this results in very large datasets for long-term measurements. This in return has the consequence, that acoustic emissions are mostly acquired at predefined cyclic time intervals, which leads to many blind spots. The following work shows methods for effective postprocessing and a feature based data acquisition method. Additionally, a two stage wear mechanism for bearings was found by the described method and could be substantiated by a numerical simulation.

    Kurzfassung: Physical simulations of tribo contacts in internal combustion engines can act as a supporting tool to match upcoming guidelines and emission restrictions. In particular, the scuffing resistance of the contact between the piston ring and cylinder liner suffers under decreasing oil viscosity and limitation of antiwear additives. This paper aims to provide an experimental method to simulate the scuffing of the piston ring/cylinder liner-contact and to validate this method with real engine parts and the literature from engine tests. The experimental methodology uses a linear tribometer TE77 to test specimens from original piston rings and liners under reciprocating motion. Additionally, the ring specimen is given the opportunity to perform secondary movements (ring twisting, ring turning) and to run under deficient lubrication conditions similar to the engine. A specially designed test strategy enables the reproducible creation of seizure of the tribosystem. The seizure resistance of two engine oils, tested for validation, correlates with the known engine performance. Therefore, the model test system can be seen as a reproducible tool for simulating seizure of a ring/liner-system, showing similar trends and wear mechanisms as in an engine. Surface analysis depicts similarities between the scuffed surfaces of an engine and the model and discusses the origin of seizure based on the model specimens together with the relevant literature.

    Kurzfassung: In this second part of our systematic review on the research area of 3D cell culture in micro-bioreactors we give a detailed description of the published work with regard to the existing micro-bioreactor types and their applications, and highlight important results gathered with the respective systems. As an interesting detail, we found that micro-bioreactors have already been used in SARS-CoV research prior to the SARS-CoV2 pandemic. As our literature research revealed a variety of 3D cell culture configurations in the examined bioreactor systems, we defined in review part one “complexity levels” by means of the corresponding 3D cell culture techniques applied in the systems. The definition of the complexity is thereby based on the knowledge that the spatial distribution of cell-extracellular matrix interactions and the spatial distribution of homologous and heterologous cell–cell contacts play an important role in modulating cell functions. Because at least one of these parameters can be assigned to the 3D cell culture techniques discussed in the present review, we structured the studies according to the complexity levels applied in the MBR systems.
