    Kurzfassung: Multiple sclerosis (MS) is an inflammatory and demyelinating disease of the central nervous system (CNS). Mesenchymal stem cells (MSC) have been shown to ameliorate symptoms in experimental autoimmune encephalomyelitis (EAE), a model of MS. Using cloned MSC labeled with clinically approved small particles of iron oxide (SPIO) for treatment of EAE we analyzed the tissue localization of transferred cells. Treatment with unlabeled MSC led to disease amelioration compared to controls. In contrast, treatment with SPIO-labeled MSC lead to increase in disease severity. Treatment with SPIO alone did not alter disease course. After transplantation labeled and nonlabeled MSC were detected in the CNS and the liver with significantly more SPIO-labeled cells present in the CNS. Iron deposition was present in the group treated with SPIO-labeled MSC, indicating that in vivo the initially cell surface-bound iron detached from the MSC. These results could be of great importance for imaging of patients in the clinical setting, indicating that in vivo application of SPIO-labeled MSC needs to be performed with caution because the cell-derived exposure of iron can lead to disease aggravation.

    Kurzfassung: Air embolism (AE), especially when affecting the brain, is an underrated and potentially life-threatening complication in various endovascular interventions. This study aims to investigate experimental AEs using a new model to generate micro air bubbles (MAB), to assess the impact of a catheter on these MAB, and to demonstrate the applicability of this model in vivo. Materials and Methods: Micro air bubbles were created using a system based on microfluidic channels. The MAB were detected and analyzed automatically. Micro air bubbles, with a target size of 85 µm, were generated and injected through a microcatheter. The MAB diameters proximal and distal to the catheter were assessed and compared. In a subsequent in vivo application, 2000 MAB were injected into the aorta (at the aortic valve) and into the common carotid artery (CCA) of a rat, respectively, using a microcatheter, resembling AE occurring during cardiovascular interventions. Results: Micro air bubbles with a highly calibrated size could be successfully generated (median: 85.5 µm, SD 1.9 µm). After passage of the microcatheter, the MAB were similar in diameter (median: 86.6 µm) but at a lower number (60.1% of the injected MAB) and a substantially higher scattering of diameters (SD 29.6 µm). In vivo injection of MAB into the aorta resulted in cerebral microinfarctions in both hemispheres, whereas injection into the CCA caused exclusively ipsilateral microinfarctions. Conclusion: Using this new AE model, MAB can be generated precisely and reproducibly, resulting in cerebral microinfarctions. This model is feasible for further studies on the pathophysiology and prevention of AE in cardiovascular procedures.

    Kurzfassung: The German healthcare system is characterized by digital change. In recent years, a trend towards the entry of start-ups and medium-sized enterprises into this sector has developed. These types of companies in particular are facing enormous challenges due to the industry and the ongoing digital change. This article presents the fundamental developments and challenges of these companies. Further solutions developed by the Tech4Age research group are presented briefly. Finally, this article describes how these solutions can be integrated into a formal concept for the promotion and further development of SMEs and start-ups in the German health industry in order to enable them quickly and comprehensively for the development of excellent digital health products.

    Kurzfassung: In everyday life, individuals often need to make choices about the difficulty level of tasks they wish to perform. Here, we investigate age- and gender-related differences in the monitoring of discrepancies between the difficulty of a given task and one’s own performance level, and in the likelihood to select task difficulties that match one’s performance level. Male and female children, teenagers, younger adults, and older adults (total N = 160) were asked to play a modified version of the BINGO game. Task difficulty was operationalized as the number of cards played simultaneously. We expected that (a) discrepancies between individuals’ self-selected difficulty levels and their objectively assessed maximum manageable task difficulty (MMTD) would be lowest in early adulthood; (b) children and teenagers, on average, would select relatively difficult task difficulties; and (c) males would overestimate their performance levels, on average, to a greater extent than females. As predicted, younger adults selected task difficulties closest to their MMTD. All other age groups, including older adults, chose task difficulties above their MMTD. The expected gender differences were restricted to children, with boys showing more pronounced performance overestimations than girls. Children and teenagers fluctuated more in their difficulty choices than adults, and many of them, especially boys, occasionally chose difficulty levels far beyond their performance capabilities. We conclude that task-difficulty choices are an interesting topic for lifespan studies. Future research should systematically vary the physical risk involved in a task, and also include the presence of peers.

    Kurzfassung: Cat breeders aim to suppress the sexual cycle for a defined period. This is a challenge as most preparations available for this intention are orally administered progestins; however, these drugs may have side effects, especially when used over a long period of time. Long-acting gonadotropin-releasing hormone agonist implants have been used successfully to suppress the oestrous cycle in cats for a longer period. After removal of the implant, the duration of action was shortened; however, the re-occurrence of normal cyclicity cannot be foreseen. The search for alternatives is therefore ongoing. Clinical challenges Another possibility for oestrus control is orally administered melatonin preparations; however, previous studies have shown that the effect was not satisfactory. After subcutaneous application of a melatonin implant developed for induction of oestrus in sheep (Melovine; Ceva Santé Animale), duration of oestrus suppression was shown to differ highly in individuals and in some cases no effect was observed. Nevertheless, it is used for short-term oestrus suppression in female cats. Aims This article provides an overview of recent studies and clinical experiences of the use of melatonin as a contraceptive in cats. It explains the function of melatonin in cats, discusses previous studies, provides clinical experience with Melovine (27 cases) and elucidates advantages and disadvantages. Evidence base Information provided in this article is drawn from the published literature and the author’s own clinical experience.

    Kurzfassung: The aim of the study was to evaluate the efficacy of a 4.7 mg deslorelin implant in tom cats. Methods Nine mature male cats were included in the deslorelin group and five cats in the control group. Before the study started, all cats were confirmed to have distinct sexually dimorphic behaviour. Blood samples were taken on the implantation day, at day 7 and at day 15, then monthly, in order to measure serum dihydrotestosterone (DHT) and 17beta(β)-oestradiol concentrations. The deslorelin group (n = 9) was divided into two subgroups: five cats (cats 1–5) were neutered in the postimplantation period during suppression of sexually dimorphic behaviour, and four cats (cats 6–9) were neutered after re-expression of sexually dimorphic behaviour. The control group cats (n = 5) were castrated without administration of the implant. Results Sexually dimorphic behaviours ceased within a mean ± SD of 13–58 days (23.30 ± 14.17) after implantation. DHT concentration decreased within 30 days. The mean duration of suppression was 26.5 ± 7.42 months and reactivation coincided with increased DHT values reaching preimplantation concentrations within 1 month. 17β-oestradiol concentrations significantly correlated with DHT concentrations ( P 〈 0.01). For cats castrated during suppression of sexual behaviour, the length of the long axes of the nuclei of Leydig cells, the diameter of seminiferous tubules and the height of the epithelium of the seminiferous tubules did not change until 3–6 months after implantation, whereas at 12 and 32 months the measured values were even lower than in the control group. For cats castrasted after reactivation, the length of long axes of the nuclei of Leydig cells and the diameter of seminiferous tubules approached the values of the control group between 4 and 6 months after reactivation. Conclusions and relevance A deslorelin implant (4.7 mg) suppresses sexually dimorphic behaviour in tom cats without any side effects and with full reversibility; however, duration of suppression is highly individual.
