Objetivo: Desenvolver e sintetizar novos inibidores de metaloproteinases (iMMPs) associados à estratégia adesiva etch-and-rinse. Avaliar a sua viabilidade celular e resistência adesiva à dentina, aos seis meses. Materiais e Métodos: Depois da síntese iMMPs (K8, K12, K16) e da sua análise da inibição enzimática, o grau de conversão do monómero foi calculado analisando os espectros de Fourier Transform Near-Infrared Spectroscopy (FTIR), a sua solubilidade foi calculada utilizando o software ACD/Percepta (ACD/Labs algorithms model). A citotoxicidade foi medida por ensaio de MTT em duas linhas celulares NIH 3T3 e MG-63. Os ensaios de resistência adesiva foram feitos por microtração (μTBS) e baseados nas normas ISO/TS 11405:2015. Trinta molares hígidos foram aleatoriamente dividos em seis grupos (n=5) e testados às vinte e quatro horas e aos seis meses: OPT FL (Optibond FL- Kerr Corporation, Orange, CA, USA); K8 (Optibond FL+ K8); K12 (Optibond FL + K12); K16 (Optibond FL + K16); CHX0,2 (Optibond FL + CHX 0,2%) e CHX 2 (Optibond FL + CHX 2%). Os dados foram analisados com ANOVA de medidas repetidas (a=0,05), e fez-se caracterização da analíse ultramorofologica de todos os dentes. Resultados: O inibidor K12 mostrou ter a maior inibição enzimática estatisticamente significativa. O grau de conversão do monómero apresentou tendência para uma diminuição relativamente ao OPT FLcom excepção do K8. Os inibidores não são tóxicos na concentração de 20 μl. Os testes de μTBS não apresentam diferenças significativas (p〉0,05) quando comparados com o grupo de controlo OPT FL. A associação do inibidor K12 e CHX a 0,2% ao sistema adesivo parece manter a camada híbrida mais estável morfologicamente passados seis meses.
