Goal of the dissertation was the analysis of the antibiotic stress response of Staphylococcus aureus against the important antibiotics vancomycin and linezolid and in addition the establishment of new protocols for gel-free proteomics. The analysis of the vancomycin stress response (article 1) was one of the most comprehensive gel-free proteomic studies so far. We could show that several enzymes producing precursors for the cell wall were upregulated after vancomycin stress. During the dissertation a new protocol for the enrichment of dilute proteins based on the solid phase extraction system StrataClean has been established (article 2). The protocol is superior for the quantitative analysis of extremely dilute proteins. With this new protocol the linezolid stress response of S. aureus has been elucidated (article 3). We could show that linezolid stress induces a temporary growth arrest and specific regulations. In addition, also changes in the cell morphology could be detected. Combining this results a new hypothesis, that linezolid dissolves transertion complexes, which are important for cell division could be established.
