In 2008, Huang and co-workers [15] modified the 6th position of A ring of baicalein and investigated the activity of nuclear factor-kappa B (NF-κB), activator protein-1 (AP-1), or cyclic-AMP response element binding protein (CREB)-mediated transcription. The results showed that analogues with O-alkyl group of the different carbon chain length or O-benzyl activated NF-κB transcription without TNFα stimulation.

2022, PhytomedicineCitation Excerpt :Cai et al. (2017) reported kidney injury and fibrosis induced by oral administration of high doses of baicalin (400, 800 and 1600 mg/kg) for 8 days in rats. Hence, future R&D of baicalein-based antifibrotics may include the following key directions: first, determining the safe dosing range of baicalein for long-term use, as needed for prevention and treatment of many fibrotic disorders; second, examining its clinical efficacy and safety in fibrotic conditions, such as CKD; finally, unravelling the key structural basis underpinning activities and toxicity, and designing better baicalein derivatives with more robust antifibrotic activity and better safety profiles—this is plausible, given that baicalein derivatives have already achieved altered anti-P-glycoprotein activity, which is related to multiple drug resistance (Lee et al., 2004), altered regulation of NF-kB-, AP-1-, or CREB-mediated transcription (Huang et al., 2008) and more potent CDK1 inhibition and cancer repression (Ha et al., 2016). IN1130 is a positive control antifibrotic in this study.

Zhao et al., 2019). This plant is also reported to comprise of other major flavonoids such as baicalin, wogonin, and oroxylin-A, that exerts wide range of pharmacological activities (S. T. Huang, Lee, Gullen, & Cheng, 2008). Due to its various biological activities, this plant is used in the Traditional Chinese Medicine (TCM), and are mentioned in various books of TCM such as Shennong Bencaojing and Bencao Gangmu (Compendium of Materia Medica), for the treatment of cold, hypertension, inflammation, insomnia, and lung and liver diseases (Weber, Wheat, & Currie, 2010; Gong et al., 2018; Zhu & Woerdenbag, 1995; Zhu & Woerdenbag, 1995).
